Photo-Induced Stereoselective Synthesis of C-Glycosides via Conformation-Locked Glycosyl Radical Coupling.

C-glycosides are crucial scaffolds in the development of carbohydrate-based drugs, owing to their favorable pharmacological properties and metabolic stability. However, the stereoselective synthesis of specific anomers under mild conditions remains a significant challenge, particularly for the β-anomer. In this study, we developed a visible-light-mediated, photocatalyst- and metal-free strategy for the stereoselective synthesis of β-alkyl C-glycosides via radical coupling. The pivot of this methodology is the utilization of bulky silyl-protected 2,3,5,6-tetrafluoropyridin-4-ylthio (SPyf) glyco

Conformational determinants of the activity of antiproliferative factor glycopeptide.

The antiproliferative factor (APF) involved in interstitial cystitis is a glycosylated nonapeptide (TVPAAVVVA) containing a sialylated core 1 α-O-disaccharide linked to the N-terminal threonine. The chemical structure of APF was deduced using spectroscopic techniques and confirmed using total synthesis. The synthetic APF provided a platform to study amino acid modifications and their effect on APF activity, based on which a structure-activity relationship (SAR) for APF activity was previously proposed. However, this SAR model could not explain the change in activity associated with minor alter

Synthesis and Glycosidase Inhibition Studies of Novel Exoglycals Targeting GH3 Family Enzymes: Insights from Comparative Analysis with Macrolide Antibiotics.

Glycosidases are key enzymes involved in carbohydrate metabolism. Members of the GH3 family have emerged as therapeutic targets due to their roles in natural product biosynthesis and disease. Transition-state analogues represent a powerful strategy for glycosidase inhibition, and exoglycals (C-glycosylidenes) have gained interest as conformational mimics of glycosidic bond hydrolysis. Herein, we report the synthesis of novel exoglycals bearing diverse substituents, including terminal alkynes and thymidyl residues, via a modified Julia-olefination strategy from sugar-derived lactones and substi

[Antiganglioside antibodies--their pathophysiological effects on Guillain-Barré syndrome and variants].

Gangliosides, N-acetylneuraminic acid (sialic acid)-bearing glycosphingolipids, are believed to reside in clusters within membrane microdomains, called lipid rafts or glycosynapse. Recent studies demonstrated that antiganglioside antibodies play an important role in the pathogenesis of Guillain-Barré syndrome (GBS) and Fisher syndrome (FS). The anti-GM1 antibodies are likely to damage peripheral nerves through complement activation with dysfunction of voltage-gated sodium channels. Some antiganglioside antibodies may cause dysfunction of voltage-gated calcium channels without complement activa

Ion mobility-mass spectrometry of complex carbohydrates: collision cross sections of sodiated N-linked glycans.

Currently, the vast majority of complex carbohydrates are characterized using mass spectrometry (MS)-based techniques. Measuring the molecular mass of a sugar, however, immediately poses a fundamental problem: entire classes of the constituting monosaccharide building blocks exhibit an identical atomic composition and, consequently, also an identical mass. Therefore, carbohydrate MS data can be highly ambiguous and often it is simply not possible to clearly assign a particular molecular structure. A promising approach to overcome the above-mentioned limitation is to implement an additional gas

Study of biodegradation behavior of chitosan-xanthan microspheres in simulated physiological media.

Microspheres of a polyelectrolyte complex hydrogel were prepared from chitosan and xanthan after interaction between the two polyionic polymers. Their biodegradation was studied vs. chitosan. Simulated gastric fluid (SGF, pH 1.2) and intestinal fluid (SIF, pH 7.5) both as biodegradation media and phosphate buffered saline (PBS, pH 7.4) as a negative control were used. The degradation studies were performed at 37 degrees C at 240 rpm permanent stirring to mimic the physiologic conditions. High performance liquid chromatography (HPLC) was carried out to quantify the chitosan degradation products

Sequence-specific 1H-NMR assignments and folding topology of human CD59.

CD59 is a recently discovered cell-surface glycoprotein that restricts lysis by homologous complement and has limited sequence similarity to snake venom neurotoxins. This paper describes the first results of a two-dimensional NMR study of CD59 prepared from human urine. Nearly complete 1H-NMR assignments were obtained for the 77 amino acid residues and partial assignments for the N-glycan and the glycosylphosphatidylinositol (GPI) anchor. These results together confirm that the C-terminal residue of the mature protein is Asn 77 and that the urine-derived form retains the nonlipid part of the G

Structural and immunochemical analysis of three alpha-limit dextrin oligosaccharides.

Complete structures are described for three urinary oligodextrins from one patient with type II and one patient with type III glycogen storage disease. GLC-MS, direct probe MS, and 1H NMR demonstrate two heptasaccharides and one hexasaccharide containing only alpha 1-4 and alpha 1-6 linkages. The observation that all three oligosaccharides were present in urine of both patients and the occurrence of alpha 1-4 and alpha 1-6 linkages in characteristic sequences indicates that the oligodextrins are limit dextrins derived from alpha-amylolytic degradation of glycogen. The binding affinities of the

Conformationally Restricted Glycoconjugates Derived from Arylsulfonamides and Coumarins: New Families of Tumour-Associated Carbonic Anhydrase Inhibitors.

The involvement of carbonic anhydrases (CAs) in a myriad of biological events makes the development of new inhibitors of these metalloenzymes a hot topic in current Medicinal Chemistry. In particular, CA IX and XII are membrane-bound enzymes, responsible for tumour survival and chemoresistance. Herein, a bicyclic carbohydrate-based hydrophilic tail (imidazolidine-2-thione) has been appended to a CA-targeting pharmacophore (arylsulfonamide, coumarin) with the aim of studying the influence of the conformational restriction of the tail on the CA inhibition. For this purpose, the coupling of sulfo

Iron (III) isomaltoside 1000.

Intravenous (iv.) iron is now the recommended treatment for iron deficiency anemia if oral preparations have failed or in those undergoing hemodialysis. Iron isomaltoside is a new iv. iron preparation, licensed since 2009 in the UK and Europe. The iron is tightly bound within a nonionic isomaltoside carbohydrate matrix, as opposed to most other iv. iron preparations that use branched polymers to form a carbohydrate shell. This conformation produces a low immunogenic potential, which allows high single-dose infusions to adequately replenish stores. Two Phase III, open-label, noncomparative, mul

Inhibition of HIV entry by carbohydrate-binding proteins.

Carbohydrate-binding proteins (CBP) can be isolated from a variety of species, including procaryotes (i.e. cyanobacteria), sea corals, algae, plants, invertebrates and vertebrates. A number of them, in particular those CBP that show specific recognition for mannose (Man) and N-acetylglucosamine (GlcNAc) are endowed with a remarkable anti-HIV activity in cell culture. The smallest CBP occur as monomeric peptides with a molecular weight of approximately 8.5 kDa. Many others are functionally dimers, trimers or tetramers, and their molecular weight can sometimes largely exceed 50 kDa. CBP can cont

MRS reveals additional hexose N-acetyl resonances in the brain of a mouse model for Sandhoff disease.

Sandhoff disease, one of several related lysosomal storage disorders, results from the build up of N-acetyl-containing glycosphingolipids in the brain and is caused by mutations in the genes encoding the hexosaminidase beta-subunit. Affected individuals undergo progressive neurodegeneration in response to the glycosphingolipid storage. (1)H magnetic resonance spectra of perchloric acid extracts of Sandhoff mouse brain exhibited several resonances ca 2.07 ppm that were not present in the corresponding spectra from extracts of wild-type mouse brain. Characterization of polysaccharide conformational epitopes by surface plasmon resonance.

SPR techniques can provide a wealth of insight into the nature of protein-carbohydrate interactions. Information not obtained readily by other methodologies can be gathered relatively quickly in a label-free manner with low sample consumption. This chapter focused on two applications in which SPR has been used to map conformational epitopes on bacterial polysaccharides recognized by protective antibodies. In one example, methods for demonstrating the conformational nature of the epitope recognized by an anti-GBS antibody were described. Dramatic epitopic stabilization at 2 repeating units with

Neutral core oligosaccharides of bovine submaxillary mucin--use of lead tetraacetate in the cold for establishing branch positions.

Oligosaccharide alditols were released from bovine submaxillary mucin by alkaline borohydride treatment. The fractions containing smaller neutral oligosaccharides were separated by HPLC, to give, in the mono-, di- and trisaccharide-alditol sizes, 19 different structures, in addition to three fucosylated tetrasaccharide alditols. Molecules were identified by NMR spectroscopy, electrospray-mass-spectrometry-mass-spectrometry, permethylation analyses, and highly selective Pb(OAc)4 oxidation at -30 degrees C, followed by borohydride reduction. Pb(OAc)4 oxidation was found to be generally applicabl

Elongation of the N-glycans of fowl plague virus hemagglutinin expressed in Spodoptera frugiperda (Sf9) cells by coexpression of human beta 1,2-N-acetylglucosaminyltransferase I.

Spodoptera frugiperda (Sf9)-cells differ markedly in their protein glycosylation capacities from vertebrate cells in that they are not able to generate complex type oligosaccharide side chains. In order to improve the oligosaccharide processing properties of these cells we have used baculovirus vectors for expression of human (beta 1,2-N-acetylglucosaminyltransferase I (hGNT-I), the enzyme catalysing the crucial step in the pathway leading to complex type N-glycans in vertebrate cells. One vector (Bac/GNT) was designed to express unmodified GNT-I protein, the second vector (Bac/tagGNT) to expr

Exploring the structural insights on human laforin mutation K87A in Lafora disease--a molecular dynamics study.

Lafora disease (LD) is an autosomal recessive, progressive form of myoclonus epilepsy which affects worldwide. LD occurs mainly in countries like southern Europe, northern Africa, South India, and in the Middle East. LD occurs with its onset mainly in teenagers and leads to decline and death within 2 to 10 years. The genes EPM2A and EPM2B are commonly involved in 90 % of LD cases. EPM2A codes for protein laforin which contains an amino terminal carbohydrate binding module (CBM) belonging to the CBM20 family and a carboxy terminal dual specificity phosphatase domain. Mutations in laforin are fo

Influence of Modification of Cysteine Residues in the Carbohydrate-Binding Domain on Cellulase Activity: Achieving Enhanced Performance and pH-Sensitive Recovery.

The conversion of cellulosic biomass into fermentable sugars represents a pivotal stage in the environmentally friendly and sustainable production of clean fuels and chemicals. In this paper, a new strategy to enhance enzyme performance was proposed. This strategy can achieve pH-sensitive recovery of cellulase while improving the catalytic activity and stability of the enzyme. This strategy was achieved by means of a chemical modification of the cysteine residue (-SH) of the carbohydrate-binding domain (CBD) of cellulase with a PEG-based modifier (Mal-PEG-FA). It was shown that chemical modifi

Genetic testing of two Pakistani patients affected with rare autosomal recessive Fanconi-Bickel syndrome and identification of a novel SLC2A2 splice site variant.

Fanconi-Bickel syndrome (FBS) is a rare autosomal recessive carbohydrate metabolism disorder caused by mutations in SLC2A2 encoding the glucose transporter 2 (GLUT2) protein. The clinical manifestations include hepatomegaly, conditional hypo/hyperglycemia, rickets, short stature and proximal renal tubular dysfunction. GLUT2 regulates monosaccharide homeostasis through sugar sensing and transmembrane transportation during high/low glucose levels. In the current study, we present two siblings suffering from FBS. The patients presented with doll-like facies, failure to gain weight and height, abd

D2O-Labeled Single-Cell Raman Spectroscopy Reveals the Evolutionary Trajectory of Helicobacter pylori During the Phenotypic Silent Window.

Helicobacter pylori is a carcinogenic pathogen with a high gastric cancer risk. The rapid emergence of antibiotic resistance severely compromises treatment efficacy. However current conventional approaches fail to capture early molecular events preceding detectable resistance. Here, we used a gradient intermittent antibiotic exposure model to simulate stepwise resistance evolution and longitudinally acquired single-cell Raman spectra across adaptive cycles. By integrating two-dimensional correlation Raman spectroscopy (2D-COS), we resolved the temporal sequence of molecular alterations during
