Ag(I) complexes with natural products and triphenylphosphine: activity against tumor cell lines and mycobacterium tuberculosis.

Cancer and tuberculosis remain among the leading causes of morbidity and mortality worldwide, and the emergence of drug resistance in both diseases highlights the urgent need for new therapeutic strategies. In this context, metal-based compounds combined with bioactive natural products have emerged as promising alternatives to conventional drugs. In the present study, a series of silver (I) complexes containing natural products (curcumin, lawsone, lapachol or alizarin) and triphenylphosphine (PPh3) (complexes 1–4) were synthesized and structurally characterized by IR spectroscopy, elemental an

Optimization of Doxorubicin Loading into Citrus Lemon-Derived Nanoparticles and Evaluation of Their Antitumor Effects on Tumor Cell Lines.

Recently, lipid bilayer nanoparticles, similar to extracellular vesicles, have been identified in various plant juices and extracts. These plant-derived nanoparticles have attracted attention in the medical field for their intrinsic anti-inflammatory and antioxidant properties and for the advantages of inexpensive and abundant raw materials. Although citrus lemon-derived extracellular vesicle-like nanoparticles (cNPs) exert selective antitumor effects on several cancer cell lines, information on these effects remains limited. Therefore, in this study, we focused on cNPs as nanocarriers for eff

Cytotoxic Effects of New Palladium(II) Complexes with Thiazine or Thiazoline Derivative Ligands in Tumor Cell Lines.

The synthesis of analogs of cisplatin, which is a widely used chemotherapeutic agent, using other metal centers could be an alternative for cancer treatment. Pd(II) could be a substitute for Pt(II) due to its coordination chemistry similarity. For that reason, six squared-planar Pd(II) complexes with thiazine and thiazoline ligands and formula [PdCl2(L)] were synthesized and characterized in this work. The potential anticarcinogenic ability of the compounds was studied via cytotoxicity assay in three different human tumor cell lines, i.e., epithelial cervix carcinoma (HeLa), promyelocytic leuk

Molecular Characterization of the Response to Conventional Chemotherapeutics in Pro-B-ALL Cell Lines in Terms of Tumor Relapse.

Little is known about optimally applying chemotherapeutic agents in a specific temporal sequence to rapidly reduce the tumor load and to improve therapeutic efficacy. The clinical optimization of drug efficacy while reducing side effects is still restricted due to an incomplete understanding of the mode of action and related tumor relapse mechanisms on the molecular level. The molecular characterization of transcriptomic drug signatures can help to identify the affected pathways, downstream regulated genes and regulatory interactions related to tumor relapse in response to drug application. We

Dubious effects of methadone as an "anticancer" drug on ovarian cancer cell-lines and patient-derived tumor-spheroids.

BACKGROUND: The opioid agonist D,L-methadone exerts analgesic effects via the mu opioid receptor, encoded by OPRM1 and therefore plays a role in chronic pain management. In preclinical tumor-models D,L-methadone shows apoptotic and chemo-sensitizing effects and was therefore hyped as an off-label "anticancer" drug without substantiation from clinical trials. Its effects in ovarian cancer (OC) are completely unexplored. METHODS: We analyzed OPRM1-mRNA expression in six cisplatin-sensitive, two cisplatin-resistant OC cell-lines, 170 OC tissue samples and 12 non-neoplastic control tissues. Pro-a

Bothrops Jararaca Snake Venom Modulates Key Cancer-Related Proteins in Breast Tumor Cell Lines.

Cancer is characterized by the development of abnormal cells that divide in an uncontrolled way and may spread into other tissues where they may infiltrate and destroy normal body tissue. Several previous reports have described biochemical anti-tumorigenic properties of crude snake venom or its components, including their capability of inhibiting cell proliferation and promoting cell death. However, to the best of our knowledge, there is no work describing cancer cell proteomic changes following treatment with snake venoms. In this work we describe the quantitative changes in proteomics of MCF

Functional and genomic characterization of three novel cell lines derived from a metastatic gallbladder cancer tumor.

BACKGROUND: Gallbladder cancer (GBC) is the most common tumor of the biliary tract. The incidence of GBC shows a large geographic variability, being particularly frequent in Native American populations. In Chile, GBC represents the second cause of cancer-related death among women. We describe here the establishment of three novel cell lines derived from the ascitic fluid of a Chilean GBC patient, who presented 46% European, 36% Mapuche, 12% Aymara and 6% African ancestry. RESULTS: After immunocytochemical staining of the primary cell culture, we isolated and comprehensively characterized thre

Expression of the miR-150 tumor suppressor is restored by and synergizes with rapamycin in a human leukemia T-cell line.

miR-150 functions as a tumor suppressor in malignancies of the lymphocyte lineage and its expression is significantly reduced in these cells. However, the mechanism of miR-150 repression is unknown and so are pharmacological interventions that can reverse it. Here, we report that reduced expression of miR-150 in human Jurkat T-cell acute lymphoblastic leukemia (T-ALL) cells is mediated by constitutive mTOR signaling, a common characteristic of T-ALL cell lines and clinical isolates. Activating mTOR signaling in non-malignant T cells also resulted in a significant miR-150 down-regulation. Conve

Synergistic cytotoxicity against human tumor cell lines by oncolytic adenovirus dl1520 (ONYX-015) and melphalan.

AIMS AND BACKGROUND: In light of the need for more selective anticancer therapy, much work has been directed at developing compounds or biological agents that target functions specific to cancer cells. To this end, numerous viruses have been engineered to exploit the dependence of cancer cells on particular anomalies that contribute to their rogue proliferative activity, such as dysfunctional p53, overactive mitogenic signaling, or a defective interferon response. The oncolytic human adenovirus dl1520 (ONYX-015) was engineered to propagate specifically in p53-deficient tumors, which comprise o

The distribution and expression profiles of human Aspartyl/Asparaginyl beta-hydroxylase in tumor cell lines and human tissues.

Human aspartyl beta-hydroxylase (HAAH) is a highly conserved enzyme that hydroxylates epidermal growth factor-like domains in transformation-associated proteins. Previous studies showed that the HAAH gene was overexpressed in many human malignancies. The present study investigates the distribution and expression profiles of HAAH in a variety of tumor cell lines and human tissues. One hundred and four cases of carcinomas were retrieved from the pathology archives of Department of Pathology, First Affiliated Hospital of Medical College of Xi'an Jiaotong University. Expression of Jun and Fos proteins in ovarian tumors of different malignant potential and in ovarian cancer cell lines.

Jun and Fos proteins form the transcription factor activating protein 1 (AP-1). They play a role in cell proliferation, malignant transformation and invasion in various tumors. The aim of the current study was to characterize the role of AP-1 in ovarian cancer. Fifty-six ovarian tumors of different invasive potential including 13 metastases as well as 5 ovarian cancer cell lines were analyzed by Western blot analysis regarding their expression of pc-Jun, Jun B, Jun D, c-Fos, Fos B, Fra-1 and Fra-2. In addition, invasion, cell proliferation and migration assays were performed. The expression of

Establishment and characterization of 4 new human pancreatic cancer cell lines: evidences of different tumor phenotypes.

OBJECTIVES: Pancreatic cancer still remains a challenge for its biological complexity and lack of effective therapeutic strategies. Establishing new pancreatic cancer cell lines is therefore of paramount importance to clarify its biology. METHODS: We established and characterized 4 new pancreatic cancer cell lines (PP78, PP109, PP117, and PP161) according to their genetic (K-Ras, TP53, CDKN2A, and MADH4; DNA fingerprinting; karyotype), cytostructural (cytokeratins 7, 8, 18, and 19 vimentin, and ezrin), and functional profiles (doubling time; migration assay). RESULTS: K-Ras, TP53, and CDKN2A

Specific amino acid deficiency alters the expression of genes in human melanoma and other tumor cell lines.

This study determined the effect of tyrosine (Tyr) and phenylalanine (Phe) deprivation on protein expression and phosphorylation of mitogen-activated protein kinase kinase 4 (MKK4)/stress-activated protein/Erk kinase (SEK1), a metastasis suppressor gene. Differential display and suppressive subtractive hybridization techniques identified genes modulated by Tyr and Phe deprivation. Expression of MKK4/SEK1 protein varied widely among human A375, A375SM and SB2 melanoma, PC-3 and DU145 prostate cancer, and MDA-MB-231 breast cancer cell lines and within the different lines. Diminished capacity for p53 in mediating a radiation-induced G1 arrest in established human tumor cell lines.

It has been reported that the p53 gene mediates an ionizing radiation-induced G1 arrest in mammalian cells. To further characterize this important phenomenon, a panel of seven human diploid fibroblast cell strains and 14 human tumor cell lines from a variety of sources with both wild-type and mutant p53 status were assayed for their susceptibility to G1 arrest after gamma-ray irradiation by a continuous labeling [3H]thymidine incorporation technique. An irreversible G1-block involving 20-70% of the cell population was observed in diploid fibroblasts irradiated with 4 Gy. The block was abolishe

Two cell lines established from mixed müllerian tumors of the uterus. Morphologic, immunocytochemical, and cytogenetic analyses.

To clarify the cellular origin and characteristics of malignant mixed müllerian tumor (MMMT), the authors investigated two cell lines (designated as FU-MMT-1 and FU-MMT-2) established from two patients with heterologous MMMT of the uterus. Both cell lines propagated continuously for 83 and 55 serial passages over 1.5 years, respectively. Morphologically, FU-MMT-2 was a mixture of carcinoma cells and sarcoma cells with predominance of carcinoma cells; FU-MMT-1 only had a sarcomatous element with distinct rhabdomyoblastic differentiation. Immunocytochemically, the sarcoma cells of each cell line

Differences among human tumor cell lines in the expression of glutathione transferases and other glutathione-linked enzymes.

A large number of human tumor cell lines of various origins have been investigated with respect to expression of glutathione-linked enzymes in the cytosol fraction. The amounts of the different enzymes were estimated by use of activity measurements and by silver staining or immunoblot analysis after electrophoresis of cytosol fractions purified by affinity chromatography on S-hexylglutathione Sepharose. Class Pi glutathione transferase was the most abundant enzyme in most tumor cells; the cell lines HepG2 and Raji were exceptions in not expressing significant amounts of this enzyme. HepG2 cell

Initiation and characterization of cultured tumor lines from spontaneous reticulum cell sarcoma of SJL/J mice.

Three reticulum cell sarcoma lines (LA1, LA6, and LA8) have been established from SJL/J spontaneous "Hodgkin's-like" reticulum cell sarcoma. All cultures are lymphoid with blast-like morphology by light and electron microscopy and produce the type B neoplasm (Dunn classification) when injected into young SJL/J mice. Identification of these tumors as lymphocytic and monocytic was investigated by surface markers, histochemical staining, and phagocytic function. LA6 and LA8 bear receptors for the Fc portion of immunoglobulin complement receptors; Thy 1.2 antigens and surface immunoglobulin were n

Spleen tyrosine kinase (SYK) inhibition suppresses growth of gastrointestinal neuroendocrine tumor cells: a pilot study in two cell lines.

Gastrointestinal neuroendocrine tumors (GI-NETs) lack effective targeted options beyond somatostatin analogs and mTOR inhibitors. Spleen tyrosine kinase (SYK) is a non-receptor kinase with emerging roles in solid tumors and available small-molecule inhibitors. We explored whether SYK is a plausible therapeutic target in GI-NET using two human cell lines. SYK expression in GI-NET cells was confirmed by immunofluorescence. Cells were exposed to a selective SYK inhibitor (BI-1002494), and proliferation was quantified using both 2D and 3D models. Both GI-NET models expressed SYK and exhibited redu

Anti-CD19 inhibits the growth of human B-cell tumor lines in vitro and of Daudi cells in SCID mice by inducing cell cycle arrest.

In this report, we extend our previous findings that IgG or F(ab')2 fragments of HD37 anti-CD19 antibody (Ab) in combination with the immunotoxin (IT), RFB4-anti-CD22-deglycosylated ricin A chain (dgA) (but neither reagent alone), prolonged the survival of SCID mice with disseminated human Daudi lymphoma (SCID/Daudi mice) to 1 year at which time they still remained tumor-free. We explored the mechanisms by which the HD37 Ab exerts antitumor activity in vivo by studying its activity in vitro. We found that it has antiproliferative activity (IC50 = 5.2 - 9.8 x 10(-7) mol/L) on three CD19+ Burkit
