Actions of noradrenaline on myenteric neurons in the guinea pig gastric antrum.

We used intracellular electrophysiological recording to study the actions of noradrenaline on myenteric neurons in the guinea pig gastric antrum. Noradrenaline caused a dose-dependent inhibition of the stimulus-evoked cholinergic fast excitatory postsynaptic potentials (EPSPs). Noradrenaline had no effect on the postsynaptic response to acetylcholine, suggesting a presynaptic site of action. The slow EPSP was also presynaptically inhibited by noradrenaline. In only 5% of the neurons, noradrenaline caused a postsynaptic depolarization, accompanied by increased input resistance and enhanced exci

Effect of bethanechol on gastric acid secretion and serum gastrin concentration after proximal gastric vagotomy.

It has been suggested that maximal acid output decreases after vagotomy as a result of reduced cholinergic activity and that maximal acid secretion can be restored to prevagotomy levels with cholinergic agonists. To test this, a maximally tolerated dose of bethanechol (50 micrograms/kg/hr) or saline was infused intravenously in nine duodenal ulcer patients who had been treated by proximal gastric vagotomy. Bethanechol infusion significantly (p less than 0.05) increased basal acid secretion without affecting basal serum gastrin concentrations. Despite stimulation of basal acid secretion, bethan…

The lateral parabrachial nucleus and central angiotensinergic mechanisms in the control of sodium intake induced by different stimuli.

Angiotensin II (ANG II) is a typical facilitatory stimulus for sodium appetite. Surprisingly, hyperosmolarity and central cholinergic stimulation, two classical antinatriorexigenic stimuli, also facilitate NaCl intake when they are combined with injections of the α2-adrenoceptor/imidazoline agonist moxonidine into the lateral parabrachial nucleus (LPBN). In the present study, we tested the relative importance of central angiotensinergic and cholinergic mechanisms for the control of water and NaCl intake by combining different dipsogenic or natriorexigenic stimuli with moxonidine injection into

Lithocholyltaurine interacts with cholinergic receptors on dispersed chief cells from guinea pig stomach.

Although bile acids damage gastric mucosa, the mechanisms underlying tissue injury induced by these agents are not well understood. To determine whether bile acids alter gastric secretory function, we investigated the actions of sodium cholate, deoxycholate, lithocholate, and their taurine and glycine conjugates on a highly homogeneous population of gastric chief cells. Lithocholyltaurine (LCT), a particularly injurious bile acid, caused a threefold increase in pepsinogen secretion (detectable with 100 nM and maximal with 10 microM LCT). When combined with other secretagogues, increasing conce

Comparison of the effects of histamine and N(alpha)-methylhistamine on neuronal function in the guinea-pig oesophagus and ileum.

The effects of histamine and N(alpha)-methylhistamine, two components of gastric juice, on vagal and transmural stimulation of the guinea-pig isolated oesophagus were compared with their effects on cholinergic and on non-adrenergic-non-cholinergic (NANC) neuronal responses in the isolated ileum, both tissues having been pretreated with mepyramine (1 microM). Histamine (< or = 10 microM) and N(alpha)-methylhistamine (< or = 1 microM) had no significant effect on either vagal or transmural stimulation in the oesophagus. GPR139-mediated cholinergic circuit from the medial septum to the ventral hippocampal CA3 modulates cognitive impairment in Alzheimer's disease.

G-protein coupled receptor 139 (GPR139) has been detected in the medial septum (MS) cholinergic neuron, but its roles in cholinergic circuits associated with Alzheimer's disease (AD) remain unclear. Using a variety of experimental approaches, including the establishment of AD mouse models, viral injection, immunohistochemistry, ELISA, TUNEL apoptosis assay, Golgi staining, electrophysiological recording, fiber photometry, and behavioral tests, we found that knockdown of GPR139 in MS cholinergic neurons exacerbated early-stage cognitive decline, apoptosis, and synaptotoxicity in the basal foreb

The cholinergic anti-inflammatory pathway: towards innovative treatment of rheumatoid arthritis.

The efferent vagus nerve can regulate inflammation via its principal neurotransmitter acetylcholine (ACh), a concept referred to as the 'cholinergic anti-inflammatory pathway'. ACh interacts with members of the nicotinic acetylcholine receptor (nAChR) family, in particular with the alpha7 subunit (alpha7nAChR), which is expressed not only by neurons but also macrophages and other cells involved in the inflammatory response. In these inflammatory cells, the stimulation of alpha7nAChR by ACh and other alpha7nAChR-specific agonists suppresses the release of proinflammatory cytokines. Recent work

Muscarinic cholinergic binding in rat brain.

Binding sites with high affinity and specificity for [(3)H]quinuclidinyl benzilate (QNB) are present in homogenates of rat brain. The characteristics of the binding sites resemble those of muscarinic cholinergic receptors. Specific binding is saturable with respect to [(3)H]QNB and tissue concentration and is time-, temperature-, and pH-dependent. The bimolecular rate of association (2.0 x 10(8) M(-1) min(-1)) and dissociation (1.2 x 10(-2) min(-1)) at 35 degrees indicate a dissociation constant of 60 pM and a density of 65 pmol/g of brain. New Multitarget Molecules Derived from Caffeine as Potentiators of the Cholinergic System.

Cholinergic deficit is a characteristic factor of several pathologies, such as myasthenia gravis, some types of congenital myasthenic syndromes, and Alzheimer's Disease. Two molecular targets for its treatment are acetylcholinesterase (AChE) and nicotinic acetylcholine receptor (nAChR). In previous studies, we found that caffeine behaves as a partial nAChR agonist and confirmed that it inhibits AChE. Here, we present new bifunctional caffeine derivatives consisting of a theophylline ring connected to amino groups by different linkers. All of them were more potent AChE inhibitors than caffeine.

Muscarinic receptor subtypes in airways.

Muscarinic receptor subtypes in the airways appear to subserve different physiological functions. M1-receptors facilitate neurotransmission through parasympathetic ganglia and enhance cholinergic reflexes, but are also localized to alveolar walls. M2-receptors act as autoreceptors on post-ganglionic cholinergic nerves and inhibit acetylcholine release. There is some evidence that they may be defective in asthma (as a consequence of airway inflammation?) and this may enhance cholinergic reflexes and account for beta-blocker-induced asthma. M2-receptors in airway smooth muscle may also counterac

Short-term exposure to a high-fat diet leads to neuroinflammation and impairs memory and cholinergic signaling in the hippocampal CA3 region of male mice.

Consumption of a high-fat diet (HFD) can lead to cognitive dysfunction and neuroinflammation in the hippocampus, particularly the CA3 region, which is vital for associative memory. Cholinergic input from the basal forebrain to the hippocampus is critical for regulating excitability, plasticity, and overall cognitive function in this area. Neuroinflammation may disrupt the expression of the α7 nicotinic acetylcholine receptor (α7nAChR), essential for the anti-inflammatory cholinergic pathway. We sought to assess the effects of brief HFD exposure on hippocampal cholinergic function and neurobeha

[Anticholinergic action of clonidine on cardiovascular effects of the central cholinergic estimulation].

A study was made relating the anticholinergic action of alpha 2-adrenoceptor agonist clonidine on the cardiovascular responses to i.c.v. injection of neostigmine, a quaternary anticholinesterase, in conscious rats. The cholinergic agent neostigmine (0.1-1 microgram i.c.v.) induced a dose-dependent pressor and bradycardic responses in the rats. Clonidine (10 micrograms.kg-1 i.v.), given 30 min previous the anticholinesterase, failed to modify the pressor effect of neostigmine (0.1-1 microgram i.c.v.) but prevented the bradycardia. On the other side, the i.c.v. administration of clonidine (3 mic

Different types of blockade of crustacean acetylcholine-induced currents.

The voltage dependence, concentration dependence, and agonist dependence of blocking and unblocking produced by anticholinergic agents on the ionophoretically activated cholinergic currents of the lobster gastric mill 1 (g.m.1) muscle were examined. Although the ionophoretic technique provides only qualitative information as to blocking mechanisms it is useful in revealing slow components of the blocking action of some drugs. At least two qualitatively different types of voltage-dependent block of the crustacean cholinergic currents were observed. For pempidine, mecamylamine and decamethonium

Serotonin 5-HT1A and 5-HT1B receptor-mediated inhibition of glutamatergic transmission onto rat basal forebrain cholinergic neurones.

Cholinergic neurones in the basal forebrain (BF) project into various brain regions and receive excitatory inputs from the cortex and brain stem. These cholinergic neurones receive serotonergic fibres from the dorsal raphe nuclei. This study was aimed to elucidate serotonin (5-HT)-induced modulation of glutamatergic transmission onto rat BF cholinergic neurones identified with Cy3-192IgG. Excitatory postsynaptic currents (EPSCs) were evoked by focal stimulation. Bath application of either 5-HT, the 5-HT1A receptor agonist 8-OH-DPAT (DPAT), or the 5-HT1B receptor agonist CP93129 (CP), inhibited

Rostral ventromedial medulla modulates nociception and tonic immobility behavior through connections with the A7 catecholaminergic region.

Cholinergic stimulation of the rostral ventromedial medulla (RVM) produces antinociception and reduces the duration of tonic immobility (TI) behavior in guinea pigs. Previous studies indicated that cholinergic antinociception in the RVM is mediated through connections with the A7 catecholaminergic cell group (A7). In the current study, we tested the role of the A7 in both the antinociception and reduction of TI duration mediated by cholinergic stimulation of the RVM. In addition, we used biotinylated dextran amines (BDA) to evaluate the connections between the RVM and A7. The microinjection of

Autonomic control of cardiovascular functions during neonatal development and in adult sheep.

We studied the autonomic control of resting heart rate of systemic and pulmonary vascular blood pressures (BP) in chronically instrumented neonatal lambs 1-8 weeks of age. The maximum response to ganglionic blockade and sympathetic and parasympathetic antagonists was taken as an index of the magnitude of the total neural, adrenergic, and cholinergic tones. The reactivity of the circulatory parameters to adrenergic and cholinergic agonists also was investigated. All findings were compared with those in adult nonpregnant sheep studied concomitantly and with data previously obtained from term fet…

Muscle atonia is triggered by cholinergic stimulation of the basal forebrain: implication for the pathophysiology of canine narcolepsy.

Narcolepsy is a sleep disorder characterized by excessive daytime sleepiness and rapid eye movement (REM) sleep-related symptoms, such as cataplexy. The exact pathophysiology underlying the disease is unknown but may involve central cholinergic systems. It is known that the brainstem cholinergic system is activated during REM sleep. Furthermore, REM sleep and REM sleep atonia similar to cataplexy can be triggered in normal and narcoleptic dogs by stimulating cholinergic receptors within the pontine brainstem. The pontine cholinergic system is, therefore, likely to play a role in triggering cat

Cholinergic receptors on intestine cells of Ascaris suum and activation of nAChRs by levamisole.

Cholinergic agonists, like levamisole, are a major class of anthelmintic drugs that are known to act selectively on nicotinic acetylcholine receptors (nAChRs) on the somatic muscle and nerves of nematode parasites to produce their contraction and spastic paralysis. Previous studies have suggested that in addition to the nAChRs found on muscle and nerves, there are nAChRs on non-excitable tissues of nematode parasites. We looked for evidence of nAChRs expression in the cells of the intestine of the large pig nematode, Ascaris suum, using RT-PCR and RNAscope in situ hybridization and detected mR…

Synaptic block of a transmitter-induced potassium conductance in Aplysia neurones.

A voltage-clamp study was made of a slow excitatory post-synaptic potential (slow e.p.s.p.) that can be elicited in the medial cells of the left pleural ganglion of Aplysia californica by the firing of at least three different presynaptic neurones (labelled I, II and III). Each of these three neurones elicits other permeability changes in addition to the slow e.p.s.p., and all elements of these synaptic responses were shown to be mediated monosynaptically. The slow e.p.s.p., associated with an increase in membrane resistance, was shown to be due to a decrease in K permeability. When the slow e
