Developmental roles of tyrosine metabolism enzymes in the blood-sucking insect Rhodnius prolixus.

The phenylalanine/tyrosine degradation pathway is frequently described as a catabolic pathway that funnels aromatic amino acids into citric acid cycle intermediates. Previously, we demonstrated that the accumulation of tyrosine generated during the hydrolysis of blood meal proteins in Rhodnius prolixus is potentially toxic, a harmful outcome that is prevented by the action of the first two enzymes in the tyrosine degradation pathway. In this work, we further evaluated the relevance of all other enzymes involved in phenylalanine/tyrosine metabolism in the physiology of this insect. The knockdow

Disturbed ATP and AMPK homeostasis in an Ank F377del mouse model for craniometaphyseal dysplasia.

UNLABELLED: Craniometaphyseal dysplasia (CMD) is a rare genetic disorder characterized by hyperostosis of craniofacial bones and flared metaphyses of long bones. Mutations in ANKH (mouse orthologue ANK), a transmembrane protein mediating ATP and citrate efflux, cause the autosomal dominant form of CMD. How ANK mutations in CMD affect ATP/citrate homeostasis and downstream targets remains unknown. We determined that cellular ATP export, intracellular ATP levels, and plasma citric acid were significantly reduced in ANK F377del knock-in ( Ank KI/KI ) mice. Enrichment and pathway analyses of the p

Combined Analysis of Transcriptome and Metabolome of Large-Size Triploid Rainbow Trout in Different Seasons.

Seasonal temperature variation poses a significant physiological challenge for cold-water aquaculture species; however, the molecular mechanisms underlying seasonal adaptation in large-sized triploid rainbow trout (Oncorhynchus mykiss) remain largely unexplored. In this study, we integrated liver transcriptomics and LC-MS-based untargeted metabolomics across four seasons (February, May, August, November) to investigate this phenomenon. Seasonal comparisons revealed pronounced molecular reprogramming, with the comparison between May and August showing the largest transcriptomic shift (13,359 di

A multi-omic atlas of the maternal-fetal dyad reveals metabolic stratification and prenatal signatures potentially relevant to meat quality development.

Meat quality is influenced by non-lipid small molecules, yet prenatal metabolic organization remains unclear. Here, we constructed a high-resolution atlas of flavor- and color-related metabolism using a maternal-fetal goat dyad. We integrated full-spectrum metabolomics, matrix-assisted laser desorption/ionization mass spectrometry imaging (MALDI-MSI), and full-length transcriptomics to profile longissimus dorsi muscle (LDM) and leg biceps femoris muscle (LM). Multi-omic analysis revealed a dominant two-axis pattern, with many metabolites showing higher abundance in maternal than fetal muscle a

Cold induced pain elicits reproducible breath metabolomic responses across geographically distinct populations.

Observer-independent assessment of nociceptive states is particularly important in non-communicative or vulnerable patient populations, where self-report is unavailable. In this proof-of-concept study, we explored whether exhaled breath metabolomics, measured in real time using secondary electrospray ionization high-resolution mass spectrometry (SESI-HRMS), can capture rapid physiological responses to the cold pressor test (CPT), a standardized model of acute sympathetic and nociceptive activation. Forty healthy adults (n = 19 in Switzerland, n = 21 in China) underwent four breath measurements

Comparative Physiological and Transcriptomic Analyses Revealed Key Metabolic Pathways Involved in Ethanol Fermentation in Nanguo Pears during Ripening.

Nanguo pears produce a distinctive "alcoholic taste" during ripening, while Jingbai pears do not. To better understand the regulatory mechanism of "alcohol taste" formation in Nanguo pears, differences in ethanol fermentation-related metabolisms between the two cultivars were investigated. Compared to Jingbai pears, Nanguo pears had higher activities of pyruvate decarboxylase (PDC) and alcohol dehydrogenase (ADH) and contents of pyruvate, acetaldehyde, and ethanol during ripening. Nanguo pears generally showed higher starch degradation and higher levels of acetyl-CoA and citric acid but lower

Mitochondrial dysfunction results in enhanced adrenal androgen production in H295R cells.

The role of mitochondria in steroidogenesis is well established. However, the specific effects of mitochondrial dysfunction on androgen synthesis are not fully understood. In this study, we investigate the effects of various mitochondrial and metabolic inhibitors in H295R adrenal cells and perform a comprehensive analysis of steroid and metabolite profiling. We report that mitochondrial complex I inhibition by rotenone shifts cells toward anaerobic metabolism with a concomitant hyperandrogenic phenotype characterized by rapid stimulation of dehydroepiandrosterone (DHEA, 2 h) and slower accumul

Hyperpolarized [1-13 C] pyruvate as a possible diagnostic tool in liver disease.

Introduction of hyperpolarized magnetic resonance in preclinical studies and lately translation to patients provides new detailed in vivo information of metabolic flux in organs. Hyperpolarized magnetic resonance based on 13 C enriched pyruvate is performed without ionizing radiation and allows quantification of the pyruvate conversion products: alanine, lactate and bicarbonate in real time. Thus, this methodology has a promising potential for in vivo monitoring of energetic alterations in hepatic diseases. Using 13 C pyruvate, we investigated the metabolism in the porcine liver before and aft

α-Ketoglutarate enhances bovine oocyte maturation and embryo development: insights from metabolomic and transcriptomic profiling.

Oocyte quality, governed by metabolic and epigenetic regulation, is essential for fertilization and embryogenesis. This study aimed to elucidate the dose-dependent and multifaceted roles of α-ketoglutarate (α-KG) in modulating the molecular and metabolic dynamics of cumulus-oocyte complexes (COCs) during in vitro maturation (IVM). Notably, supplementation with 100 μM α-KG resulted in the most pronounced improvement in maturation and developmental outcomes. α-KG remolds energy metabolism in both oocytes and their cumulus cells (CCs) during maturation via suppression of the citric cycle (TCA cyc

Estrogen-related receptor alpha (ERRα) controls the stemness and cellular energetics of prostate cancer cells via its direct regulation of citrate metabolism and zinc transportation.

Compared to most tumors that are more glycolytic, primary prostate cancer is less glycolytic but more dependent on TCA cycle coupled with OXPHOS for its energy demand. This unique metabolic energetic feature is attributed to activation of mitochondrial m-aconitase in TCA caused by decreased cellular Zn level. Evidence suggests that a small subpopulation of cancer cells within prostate tumors, designated as prostate cancer stem cells (PCSCs), play significant roles in advanced prostate cancer progression. However, their cellular energetics status is still poorly understood. Nuclear receptor ERR

Comparative metabolomics of MCF-7 and MCF-7/TAMR identifies potential metabolic pathways in tamoxifen resistant breast cancer cells.

OBJECTIVES: Breast cancer is the most common cancer and the leading cause of cancer-related death among women. An Estrogen Receptor (ER) antagonist called tamoxifen is used as an adjuvant therapy for ER-positive breast cancers. Approximately 40% of patients develop tamoxifen resistance (TAMR) while receiving treatment. Cancer cells can rewire their metabolism to develop resistant phenotypes, and their metabolic state determines how receptive they are to chemotherapy. METHODS: Metabolite extraction from human MCF-7 and MCF-7/TAMR cells was done using the methanol-methanol-water extraction meth

Discovery of myristic acid, from chick early amniotic fluid for the treatment of acute gastric ulcers by targeting pyruvate carboxylase to improve TCA cycle.

Gastric ulcer (GU) is a common digestive disease. We recently identified early chicken embryo amniotic fluid (ceAF) as a potent antioxidant, which exerts effects via its active components. This study demonstrates the potential of ceAF and one of its active components, myristic acid (MA), as GU protectants. UPLC-MS and GC-MS identified the key components and fatty acid contents in ceAF. In absolute ethanol-induced acute gastric ulcer (AGU) mice, ceAF and MA treatment significantly reduced ulcer area, while improving histopathology and inflammatory factor infiltration. Further, proteomics was us

Pivotal role of exogenous pyruvate in human natural killer cell metabolism.

Resting natural killer (NK) cells display immediate effector functions after recognizing transformed or infected cells. The environmental nutrients and metabolic requirements to sustain these functions are not fully understood. Here, we show that NK cells rely on the use of extracellular pyruvate to support effector functions, signal transduction and cell viability. Glucose-derived carbons do not generate endogenous pyruvate. Consequently, NK cells import extracellular pyruvate that is reduced to lactate to regenerate glycolytic NAD+ and is oxidized in the tricarboxylic acid (TCA) cycle to pro

Perturbations in mitochondrial metabolism associated with defective cardiolipin biosynthesis: An in-organello real-time NMR study.

Mitochondria are central to cellular metabolism; hence, their dysfunction contributes to a wide array of human diseases. Cardiolipin, the signature phospholipid of the mitochondrion, affects proper cristae morphology, bioenergetic functions, and metabolic reactions carried out in mitochondrial membranes. To match tissue-specific metabolic demands, cardiolipin typically undergoes an acyl tail remodeling process with the final step carried out by the phospholipid-lysophospholipid transacylase tafazzin. Mutations in tafazzin are the primary cause of Barth syndrome. Here, we investigated how defec

Isocitrate dehydrogenase 3b is required for spermiogenesis but dispensable for retinal viability.

Isocitrate dehydrogenase 3 (IDH3) is a key enzyme in the mitochondrial tricarboxylic acid (TCA) cycle, which catalyzes the decarboxylation of isocitrate into α-ketoglutarate and concurrently converts NAD+ into NADH. Dysfunction of IDH3B, the β subunit of IDH3, has been previously correlated with retinal degeneration and male infertility in humans, but tissue-specific effects of IDH3 dysfunction are unclear. Here, we generated Idh3b-KO mice and found that IDH3B is essential for IDH3 activity in multiple tissues. We determined that loss of Idh3b in mice causes substantial accumulation of isocitr

Inhibition of brain energy metabolism by the branched-chain amino acids accumulating in maple syrup urine disease.

In the present work we investigated the in vitro effect of the branched-chain amino acids (BCAA) accumulating in maple syrup urine disease (MSUD) on some parameters of energy metabolism in cerebral cortex of rats. 14CO2 production from [1-14C]acetate, [1-5-14C]citrate and [U-14C]glucose, as well as glucose uptake by the brain were evaluated by incubating cortical prisms from 30-day-old rats in the absence (controls) or presence of leucine (Leu), valine (Val) or isoleucine (Ile). All amino acids significantly reduced 14CO2 production by around 20-55%, in contrast to glucose utilization, which w
