Jingfang Baidu San Alleviates Feng-Han (Wind-Cold) Syndrome-Associated Lung Injury in Mice Through Macrophage Functional Regulation and Dectin-1-Associated Immune Remodeling.

ETHNOPHARMACOLOGICAL RELEVANCE: Jingfang Baidu San (JF) is a classic traditional Chinese medicine(TCM)formula that has been widely used in clinical practice for hundreds of years. While its efficacy in treating Feng Han (wind-cold, FH) common cold has garnered increasing attention, its underlying mechanism of action remain incompletely understood. AIM: This study aimed to evaluate the therapeutic effect of JF on wind-cold common cold and to elucidate its molecular mechanism. METHODS: A mouse model of wind-cold syndrome (FH) was established by exposing mice to wind and cold stress for 7 conse

Multimethod-guided discovery of novel α-pyrone dimers with cardiomyocyte-protective activity against cold ischemia injury from the coral-derived Talaromyces sp. TJ403-AL05.

Guided by molecular networking, HSQC-based DeepSAT, and SMART analyses, a chemical investigation of the coral-derived fungus Talaromyces sp. TJ403-AL05 led to the isolation and identification of taladipyrones A-I (1-9), a series of new α-pyrone dimers with four distinct skeletons. Among these, compounds 1 and 9 represented two unprecedented classes of rearranged α-pyrone dimers, and compounds 5-7 were the first talarolactone-type dimers obtained as racemates. Extensive spectroscopic analysis, quantum chemical calculations, and X-ray diffraction analysis were used to determine their structures,

Microfluidic transection injury and high-shear thrombus formation demonstrate increased hemostatic efficacy of cold-stored platelets and in vitro resuscitation in induced coagulopathy models.

Hemostatic resuscitation is an essential aspect of treating traumatic bleeding. Trauma-induced coagulopathy is a multifactorial disorder that can lead to increased transfusion requirements. However, little is known about the interplay between coagulopathies and stored blood products used for hemostatic resuscitation, which themselves acquire dysfunction in the form of a storage lesion. Physiologically relevant models can aid in the study of trauma and hemostatic resuscitation by incorporating important aspects such as biological surfaces and flow regimes that mimic injury. This study aims to e

Air-ventilated normothermic mechanical perfusion improves susceptibility to donation after circulatory death and cold preservation-induced cholestatic liver injury through PPAR-γ/UGT1A1 axis.

End-ischemic normothermic mechanical perfusion (NMP) could provide a curative treatment to reduce cholestatic liver injury from donation after circulatory death (DCD) in donors. However, the underlying mechanism remains elusive. Our previous study demonstrated that air-ventilated NMP could improve functional recovery of DCD in a preclinical NMP rat model. Here, metabolomics analysis revealed that air-ventilated NMP alleviated DCD- and cold preservation-induced cholestatic liver injury, as shown by the elevated release of alanine aminotransferase (ALT), aspartate aminotransferase (AST), bilirub

Expeditionary Immersion Circulating Heating Device: A Promising Technique for Treating Frostbite Injuries and Warming Intravenous Fluids in a Forward Deployed Cold Weather Environment.

INTRODUCTION: Cold weather injuries require prompt warm water immersion therapy, which proves to be a difficult task in the cold austere environment. Current guidelines recommend 104 °F water immersion, but producing and maintaining large volumes of warm water is challenging in sub-freezing temperatures. We describe a novel process of utilizing a sous vide immersion circulator to maintain warm fluids for immersion therapy and efficient fluid rewarming in a cold forward-deployed setting for the treatment of cold weather injuries in an effort to bridge the gap between current medical guidelines

Ginger-Processed American Ginseng Alleviates Myocardial Injury Induced by Acute Cold Exposure/Rewarming by Remodeling Gut Microbiota to Activate the AMPK/PGC-1α/PPARα Pathway.

Acute cold exposure/rewarming (ACE/R) disrupts cardiovascular homeostasis and causes severe myocardial injury, yet effective countermeasures are scarce. Ginger-processed American ginseng (GPAG), a medicine-food homologous herb, exerts anti-inflammatory, antioxidant, and metabolic-regulatory effects; however, its cardioprotective mechanisms against ACE/R remain unclear. An ACE/R rat model was established by exposing rats to -15 °C for 6 h followed by 24 °C rewarming for 12 h, with 7-day GPAG gavage (1 and 2 g/kg) prior to cold exposure. Integrated multiomics, fecal microbiota transplantation (F

Randomized evaluation of comparative effects of optimized static cold storage, hypothermic and normothermic machine perfusion, and colchicine pretreatment on ischemia-reperfusion injury in a porcine model.

BACKGROUND: Heart transplantation remains limited by ischemia-reperfusion injury (IRI). Optimizing graft preservation and modulating inflammation may improve early graft quality. We compared optimized static cold storage (SCS), hypothermic machine perfusion (HMP), and normothermic machine perfusion (NMP), and evaluated colchicine pretreatment as an adjunct anti-inflammatory strategy. METHODS: Thirty-six pigs were randomized to colchicine or placebo (n=18 each) and subsequently assigned to SCS, HMP, or NMP (n=12 per group). After 4 h of preservation, all hearts underwent 1 h of normothermic re

Finger- and toe-temperature responses to local cooling and rewarming have limited predictive value identifying susceptibility to local cold injury-a cohort study in military cadets.

The purpose was to evaluate whether a cold-water immersion test could be used to identify individuals susceptible to local cold injuries (LCI). Sixty-five healthy non-injured (N-I) subjects, and fifteen subjects, who were tested either prior to or after a LCI, sequentially immersed one hand and one foot, in 8 °C water for 30 min (CWI phase); this was followed by 15 min of spontaneous rewarming (RW phase). The LCI group showed a lower toe temperature during the CWI phase, and a lower maximum RW temperature of the fingers than the N-I group. However, digit temperatures during the CWI and RW phas

[Studies on ultrastructural changes in perivascular cells (F.G.P.) of small cerebral vessels at the initial stage after cold injury--chronological observations on F.G.P. within 24 hours].

As the potentiated cells in reaction to cerebral edema and damage, microglia, astrocytes and macrophages had been listed. Recent neuropathological study elucidated that most of cerebral macrophages were derived from blood leucocytes including monocytes. Several years ago, one of the authors reported the presence of histiocytic cells in perivascular space and named them as fluorescent granular perithelial (F.G.P.) cells from their morphological characteristics. The present paper is concerned with the response of the F.G.P. cells to the vasogenic edema induced with cold injury. The animals emplo…

Elucidation of the Role of TRPV1, VEGF-A, TXA2, Redox Homeostasis, and Inflammatory Cascades in Protection against Cold Injuries by Herbosomal-Loaded PEG-Poloxamer Topical Formulation.

High-altitude regions, cold deserts, permafrost regions, and the polar region have some of the severest cold conditions on earth and pose immense perils of cold injuries to exposed individuals. Accidental and unintended exposures to severe cold, either unintentionally or due to occupational risks, can greatly increase the risk of serious conditions including hypothermia, trench foot, and cold injuries like frostbite. Cold-induced vasoconstriction and intracellular/intravascular ice crystal formation lead to hypoxic conditions at the cellular level. The condition is exacerbated in individuals h

Conditioned Medium from Human Amnion-Derived Mesenchymal Stromal/Stem Cells Attenuating the Effects of Cold Ischemia-Reperfusion Injury in an In Vitro Model Using Human Alveolar Epithelial Cells.

The clinical results of lung transplantation (LTx) are still less favorable than other solid organ transplants in both the early and long term. The fragility of the lungs limits the procurement rate and can favor the occurrence of ischemia-reperfusion injury (IRI). Ex vivo lung perfusion (EVLP) with Steen SolutionTM (SS) aims to address problems, and the implementation of EVLP to alleviate the activation of IRI-mediated processes has been achieved using mesenchymal stromal/stem cell (MSC)-based treatments. In this study, we investigated the paracrine effects of human amnion-derived MSCs (hAMSC

A Dose-Response Study on Human FGF21 to Inhibit Necrosis, Spectrin Breakdown, and to Increase Intracellular Cold Shock Proteins in Cultured Cortical Neurons Subjected to Oxygen-Glucose Deprivation Injury.

The cold-stress hormone fibroblast growth factor 21 (FGF21) induces a broad spectrum of neuroprotective effects, including a blunting effect on α-II-spectrin breakdown product 145 (SBDP145) levels in models of hypoxia-ischemia (HI). Here, we studied the impact of FGF21 dosing on cell survival and on SBDP145 levels in normothermic male/female cortical neurons in an oxygen-glucose deprivation (OGD) injury model of necrosis. We also explored the impact of FGF21 dosing on cold-shock proteins RNA-binding motif 3 (RBM3) and cold-induced RNA-binding protein (CIRBP) in the OGD injury model. FGF21 conc

Blockade of fibronectin-alpha4beta1 adhesive interactions down-regulates cyclooxygenase-2 inducible nitric oxide synthase and prolongs recipient survival in a 24-hour model of cold hepatic ischemia-reperfusion injury.

We investigated the effects of the connecting segment-1 (CS1) peptide, which blocks fibronectin (FN)-alpha4beta1 integrin interactions, upon recipient survival and extent of tissue injury in a well-established rat liver model of ex vivo 24-hour cold ischemia followed by isotransplantation. In this model, CS1 peptides were administered through the portal vein of rat livers prior to and after cold ischemic storage. In addition, recipients of orthotopic liver transplants (OLT) received a dose of CS1 peptides 1 hour post-OLT. CS1 peptide therapy significantly inhibited the intragraft recruitment o

Sperm membrane fatty acid composition in the Eastern grey kangaroo (Macropus giganteus), koala (Phascolarctos cinereus), and common wombat (Vombatus ursinus) and its relationship to cold shock injury and cryopreservation success.

Marsupial spermatozoa tolerate cold shock well, but differ in cryopreservation tolerance. In an attempt to explain these phenomena, the fatty acid composition of the sperm membrane from caput and cauda epididymides of the Eastern grey kangaroo, koala, and common wombat was measured and membrane sterol levels were measured in cauda epididymidal spermatozoa. While species-related differences in the levels of linolenic acid (18:3, n-6) and arachidonic acid (20:4, n-6) were observed in caput epididymal spermatozoa, these differences failed to significantly alter the ratio of unsaturated/saturated

Inhibition of a triggering receptor expressed on myeloid cells-1 (TREM-1) with an extracellular cold-inducible RNA-binding protein (eCIRP)-derived peptide protects mice from intestinal ischemia-reperfusion injury.

BACKGROUND: Intestinal ischemia-reperfusion injury results in morbidity and mortality from both local injury and systemic inflammation and acute lung injury. Extracellular cold-inducible RNA-binding protein is a damage associated molecular pattern that fuels systemic inflammation and potentiates acute lung injury. We recently discovered a triggering receptor expressed on myeloid cells-1 serves as a novel receptor for extracellular cold-inducible RNA-binding protein. We developed a 7-aa peptide, named M3, derived from the cold-inducible RNA-binding protein, which interferes with cold-inducible
