On the background of the neurotoxicity of mercury compounds, the interaction of mercury(II) with the cytoskeleton was investigated in vitro using IMR-32 neuroblastoma cells. Conditions for culture of these cells on microscopic slides and procedures for immunofluorescence staining of the microtubule network were optimised. Both morphology and viability of IMR-32 cells were affected by mercury(II) at concentrations higher than 15 microM. Pronounced disintegration of the microtubule cytoskeleton was detected at 30 microM mercury(II). Compared to previous studies with fibroblasts, the no-observed-…

The cellular mechanisms that configure the cytoskeleton during migration of dendritic cells (DCs) are poorly understood. Immature DCs assemble specialized adhesion structures known as podosomes at their leading edge; these are associated with the localized recruitment of the Wiskott-Aldrich Syndrome protein (WASp) and the actin organizing actin-related protein 2/3 complex. In immature DCs lacking WASp, podosomes are absent, residual dysmorphic lamellipodia and filopodia are nonpolarized, and migration is severely compromised. Microinjection studies indicate that podosome assembly and polarizat…

The main conclusion from the present study is that T. parva sporozoite entry is dependent on a functional host cell actin cytoskeleton and is not driven by the parasite. Treating lymphocytes with cytochalasin D resulted in a dose-dependent reduction in the levels of host cell infection. However, the primary effect was to block sporozoite binding and only at the highest concentration (20 microM) was sporozoite internalization significantly reduced. In fact at lower concentrations (1-10 microM) cytochalasin treatment lead to a relative increase in sporozoite internalization. The results are cons…

Frontal sections of the temporal lobe including the transentorhinal/entorhinal region, amygdala, and/or hippocampus from human adult brains are studied for cytoskeleton changes using immunostaining with the antibodies AT8 and Alz-50 and selective silver impregnation methods for neurofibrillary changes of the Alzheimer type. For the purpose of correlation, the two methods are carried out one after the other on the same section. Layer pre-alpha in the transentorhinal/entorhinal region harbours nerve cells which are among the first nerve cells in the entire brain to show the development of neurof…

Retinal pigment epithelial cells, derived from human donor eyes, have been grown in culture as monolayers on membrane filters or plastic surfaces and shown to possess transferrin receptors with a monomeric molecular mass of 93,000. These receptors internalize 125I-labelled transferrin and recycle it to the surrounding medium in a similar manner to other cell types. Scatchard analyses show that there are about 100,000 high-affinity receptors on the surface of each cell and most of these receptors are associated with the cytoskeleton. In total cell extracts, there are additional low-affinity bin…

Distribution of I, III, IV and V-type collagen, fibronectin, vimentin, filamin, and desmin in myocardial biopsy specimens of 8 patients with alcoholic cardiomyopathy was studied by indirect immunofluorescence. It is found that all types of collagen and fibronectin participate in formation of diffuse cardiosclerosis while small cardiosclerosis is formed by interstitial types of collagen and to a lower extent of fibronectin. Filamin and desmin distribution in cardiomyocytes proves deformation and destruction of Z-lines and intercalated discs.

Methyl mercury (MeHg) disrupts microtubules, but effects on other cytoskeleton components are not well studied. Dose-effect relationships were used to determine the selectivity of MeHg effects on vimentin intermediate filaments, actin microfilaments, and microtubules. These were examined in PtK2 cells by immunofluorescence. At 0.5 microM MeHg the number of microtubules was reduced. After 1.0 or 2.0 microM MeHg, microtubules in most cells were disassembled except for a few "stable" microtubules. No effects on vimentin or actin filaments were observed except as secondary effects at concentration…

Mallory bodies are a morphological key feature of severe alcoholic liver cell injury (alcoholic hepatitis) and the morphological expression of dysregulation and derangement of the intermediate filament cytoskeleton of the hepatocyte. Their pathogenesis is still unclear. Studies on Mallory body formation may not only help to elucidate the mechanisms of liver cell injury associated with alcoholic hepatitis, but may also contribute to our understanding of the regulation and function of the intermediate filament cytoskeleton.

The sialoglycoprotein PAS 2 is present in cytoskeletons generated by Triton X-100 extraction of isolated human erythrocyte stroma. However, removal of the peripheral cytoskeletal proteins by elution with 0.1N NaOH prior to Triton extraction renders PAS 2 Triton-soluble. This suggests association of PAS 2 with the cytoskeletal elements lining the inner surface of the erythrocyte membrane. For this reason, we are proposing the name glycoconnectin for PAS 2, since it is a glycoprotein which connects the core of the cytoskeleton to the membrane bilayer. The cytoskeletal proteins bands 4.1a,b also …

Microtubules play central roles in the organization and morphology of trypanosomatid parasites, forming highly specialized cytoskeletal structures such as the subpellicular corset, the flagellar axoneme, and the mitotic spindle. Functional specialization of MTs is proposed to be regulated by the "tubulin code", which is defined by the combination of different α- and β-tubulin isotypes, a set of post-translational modifications and specific MT-binding proteins. Although multiple tubulin post-translational modifications have been described in trypanosomatids using specific antibodies or mass spe

This review systematically summarizes the central roles and molecular mechanisms of the cytoskeletal system-including actin filaments, microtubules (MT), intermediate filaments, and the Septin family-in the regulation of autophagy. The cytoskeleton not only provides a structural framework and facilitates transport for the autophagic process but also acts as a dynamic signaling hub, participating in every stage from autophagosome formation and cargo recognition to targeted trafficking and autophagosome-lysosome fusion. Actin filaments regulate the initiation of autophagy through dynamic assembl…

Patients with lipopolysaccharide-responsive beige-like anchor protein (LRBA) deficiency typically suffer from severe B cell dysfunction. However, the underlying mechanisms remain incompletely understood. In this study, we identify non-muscle myosin IIA (NMIIA) as an interaction partner of LRBA in B cells, and uncover a role for LRBA in regulating actin cytoskeleton dynamics during B cell activation. LRBA-deficient B cells exhibit abnormal migration, impaired F-actin polymerization, and reduced B cell receptor signalling and polarization upon activation. In addition, LRBA deficiency severely di…

The cytoskeleton plays a fundamental role in orchestrating the intracellular cargo transport, providing a dynamic scaffold that ensures the appropriate sorting, movement, and distribution of membrane-bound compartments. Through the coordinated activity of microtubules and actin filaments, cargo vesicles are driven along defined routes to ensure efficient intracellular trafficking. Among the molecular machineries that interact with the microtubule network, the intraflagellar transport (IFT) system has emerged as a key regulator of vesicular dynamics. Initially identified for its essential role …

Legionella pneumophila is a facultative intracellular bacterial pathogen capable of surviving and replicating within host cells, including macrophages and protozoans. It employs the Dot/Icm type IV secretion system (T4SS) to inject over 330 effector proteins into host cells, manipulating various cellular processes to facilitate infection. Characterizing the functions of these effectors is crucial to deciphering the pathogenesis of L. pneumophila. In this study, we identified SidG as an effector containing a Cys-His-Asp triad, whose functional state is strictly gated by its interaction with the
