Case report: Rethinking NGS analysis in diagnosing Diamond-Blackfan anemia syndrome.

Diamond-Blackfan anemia syndrome (DBAS) is a rare inherited bone marrow failure (BMF) syndrome characterized by erythroid aplasia, congenital malformations, and cancer predisposition. With its genetic heterogeneity, variable penetrance and expressivity, DBAS poses significant diagnostic challenges, necessitating advancements in genetic testing for improved accuracy. Here, we present the case of an 18-year-old male with a long-standing macrocytic anemia that remained undiagnosed despite standard whole exome sequencing (WES). Revisiting a family-trio WES analysis with clinical insight led to the…

Copper-Based Diamond-like Thermoelectric Compounds: Looking Back and Stepping Forward.

The research on thermoelectric (TE) materials has a long history. Holding the advantages of high elemental abundance, lead-free and easily tunable transport properties, copper-based diamond-like (CBDL) thermoelectric compounds have attracted extensive attention from the thermoelectric community. The CBDL compounds contain a large number of representative candidates for thermoelectric applications, such as CuInGa2, Cu2GeSe3, Cu3SbSe4, Cu12SbSe13, etc. In this study, the structure characteristics and TE performances of typical CBDLs were briefly summarized. Several common synthesis technologies …

Adult-Onset Diamond-Blackfan Anemia with RPL11 Gene Variation Case Report.

BACKGROUND Diamond-Blackfan anemia (DBA) is a rare genetic disorder associated with macrocytic anemia and reticulocytopenia, with patients usually transfusion-dependent in the first years of life. The disease inheritance is predominantly autosomal dominant, but varying presentations have been described owing to incomplete penetrance and widely variable expression. De novo mutations have been reported in about 55% of cases. This pediatric disease is commonly characterized by malformation of the extremities as well as craniofacial abnormalities and cardiac and urogenital defects. There have been…

Radiosensitivity in lymphoblastoid cell lines derived from Shwachman-Diamond syndrome patients.

Shwachman-Diamond syndrome is an autosomal-recessive disorder characterised by bone marrow failure and a cumulative risk of progression to acute myeloid leukaemia. The Shwachman-Bodian-Diamond syndrome (SBDS) gene, the only gene known to be causative of the pathology, is involved in ribosomal biogenesis, stress responses and DNA repair, and the lack of SBDS sensitises cells to many stressors and leads to mitotic spindle destabilisation. The effect of ionising radiation on SBDS-deficient cells was investigated using immortalised lymphocytes from SDS patients in comparison with positive and nega…

Growth hormone improves short stature in children with Diamond-Blackfan anemia.

BACKGROUND: Diamond-Blackfan anemia (DBA), an inherited marrow failure syndrome, has severe hypoplastic anemia in infancy and association with aplastic anemia, MDS/leukemia, and other malignancies. Short stature is present in most patients. Isolated cases have demonstrated improved growth on growth hormone (GH) therapy. PROCEDURES: GH treatment data were obtained from 19 children with DBA (6 at our site and 13 from Genentech). Control data from 44 non-GH treated children were provided by Diamond Blackfan Anemia Registry. Annual growth velocity (GV) and height-for-age Z-scores (HAZ) were compa…

Using chemical vapor deposition diamond finishing burs for conservative esthetic procedures.

The article demonstrates how chemical vapor deposition (CVD) diamond burs were used in a simple esthetic and cosmetic procedure to treat discolored anterior teeth. A patient who experienced discoloration after bleaching was treated with direct resin composite veneers. Excess restorative material close to the periodontium was removed with a CVD diamond bur attached to an ultrasonic handpiece. The results indicate that CVD diamond burs are appropriate for removing excess material at the gingival margins of resin composite restorations without damaging the periodontium.

Understanding reductive dechlorination of trichloroethene on boron-doped diamond film electrodes.

This research investigated reduction of trichloroethylene (TCE) at boron-doped diamond (BDD) film cathodes using a rotating disk electrode reactor. Rates of TCE reduction were determined as functions of the electrode potential and TCE concentration over a temperature range between 2 and 32 degrees C. Reduction of TCE resulted in production of acetate and chloride ions with no detectable intermediate products. At a current density of 15 mA/cm2 and concentrations below 0.75 mM, reaction rates were first order with respect to TCE concentration, with surface area normalized rate constants 2 orders…

Hematologic abnormalities in Shwachman Diamond syndrome: lack of genotype-phenotype relationship.

Shwachman-Diamond syndrome (SDS) is an autosomal-recessive disorder characterized by short stature, exocrine pancreatic insufficiency, and hematologic defects. The causative SBDS gene was sequenced in 20 of 23 unrelated patients with clinical SDS. Mutations in the SBDS gene were found in 75%, being identical in 11 patients. Hematologic parameters for all 3 lineages were determined over time such as absolute neutrophil counts (ANCs), granulocyte functions, and erythroid and myeloid colony formation (erythroid burst-forming unit [BFU-E] and granulocyte-monocyte colony-forming unit [CFU-GM]) from…

Chronic parvovirus infection and G6PD deficiency masquerading as Diamond-Blackfan anemia.

Bone marrow aplasia due to parvovirus B19 infections is usually mild and self-limited. In patients with hereditary or acquired hemolytic anemias, B19 infection can cause a severe and life-threatening anemia due to the shortened half-life of red cells, but here, too, the transient nature of the infection soon remits the symptoms. Chronic infections with parvovirus are more characteristically associated with immunodeficiency states. We report here a case of B19-induced anemia in a patient with G6PD deficiency and hypogammaglobulinemia which was mislabeled as Diamond-Blackfan anemia until the use…

Normal erythropoietic helper T cells in congenital hypoplastic (Diamond-Blackfan) anemia.

To examine the erythropoietic function of T and null cells in congenital hypoplastic (Diamond-Blackfan) anemia, we fractionated the peripheral blood of three normal subjects and three affected patients into subclasses of null, T and B cells. Mixtures of these cells were co-cultured in plasma clots in the presence of erythropoietin. Erythroid colonies grew in cultures of normal null cells if either normal or patient T cells were co-cultured with them. Null cells of patients with hypoplastic anemia did not produce erythroid colonies under any culture conditions. We conclude that in this disorder…

Experimental Study on Slicing Sapphire Crystal with Ultrasonic-Assisted Diamond Wire Saw.

Sapphire crystal, owing to its high hardness, chemical inertness, thermal stability, optical transparency, and superior dielectric strength, as well as resistance to scratching, abrasion, friction, and wear, is widely utilized in a broad range of engineering applications. Slicing is the most critical step in sapphire industry processing, as it largely dictates the final surface quality and morphology. Conventional wire sawing methods often lead to undesirable surface defects, while ultrasonic-assisted diamond wire sawing (UADWS) offers potential advantages through enhanced abrasive self-sharpe…

Diamond Flap Anoplasty for Severe Post-hemorrhoidectomy Anal Stenosis: A Case Report.

Anal stenosis is an uncommon but important complication of anorectal surgery, most frequently occurring after hemorrhoidectomy. While mild cases may be managed conservatively, severe stenosis typically fails to respond to non-surgical interventions and requires operative correction. The diamond flap technique has increasingly been recognized as a valuable option, providing both optimal tissue perfusion and good functional outcomes. We report the case of a 60-year-old man with a history of hemorrhoidectomy performed approximately 20 years earlier who presented with long-standing symptoms of obs…
