Does silvering or 11-ketotestosterone affect osmoregulatory ability in the New Zealand short-finned eel (Anguilla australis)?

Silvering has been associated with advancing osmoregulatory ability. Given the demonstrated role of 11-ketotestosterone (11KT) in mediating many of the silvering-related changes, we investigated the role of 11KT in driving this advanced osmoregulatory ability in the New Zealand short-finned eel (Anguilla australis). Yellow (non-migratory) eels with or without 11KT implants and blank-implanted silver (migratory) eels, either held in freshwater or subjected to seawater challenge, were sampled to determine serum [Na+] and [Cl-], pituitary prolactin mRNA levels, gill Na+/K+-ATPase activity and gil

Interactive effects of dietary composition and hormonal treatment on reproductive development of cultured female European eel, Anguilla anguilla.

Farmed female eels were fed two experimental diets with similar proximate composition but different n-3 polyunsaturated fatty acid (PUFA) levels. Both diets had similar levels of arachidonic acid (ARA), while levels of eicosapentaenoic acid (EPA) and docosahexaenoic acid (DHA) in one diet were approximately 4.5 and 2.6 times higher compared to the other diet, respectively. After the feeding period, each diet group was divided into two and each half received one of two hormonal treatments using salmon pituitary extract (SPE) for 13 weeks: i) a constant hormone dose of 18.75mg SPE/kg initial bod

Post- and pre-synaptic action of isotocin in the upper esophageal sphincter muscle of the eel: its role in water drinking.

Isotocin is a fish analogue of the mammalian hormone oxytocin. To elucidate sites of action of isotocin (IT) in the upper esophageal sphincter (UES) muscle, a key muscle in swallowing, IT was applied after treatment with tetrodotoxin (TTX). Even after blocking nerve activity with TTX, IT relaxes the UES muscle in a concentration-dependent manner, suggesting that IT receptor(s) is present on the muscle cells. Similar relaxation was also obtained by application of 3-isobutyl-1-methylxanthine (IBMX), forskolin (FSK) and 8-bromo-adenosine, 3',5'-cyclic monophosphate (8BrcAMP) after pretreatment wi

Bioaccumulation and the effects of organochlorine pesticides, PAH and heavy metals in the Eel (Anguilla anguilla) at the Camargue Nature Reserve, France.

Pesticides (organochlorines-OC), polycyclic aromatic hydrocarbons (PAH) and heavy metals are toxic to fish and may be taken in through gills, skin and contaminated foods. Here we measure concentrations of OC, PAH and heavy metals, and their effects in the eel Anguilla anguilla from three locations in the Camargue Reserve in southern France. The Camargue Biosphere Reserve is the largest coastal wetland in Western Europe, and A. anguilla is a common predator at the top of the food chain. Livers and spleens were analyzed for histopathological, chemical and organo-somatic (HSI and SSI) effects. Gi

Pseudocapillaria (Ichthyocapillaria) ophisterni sp. n. (Nematoda : Capillariidae) from the swamp-eel Ophisternon aenigmaticum (Pisces) in Mexico.

A new nematode species, Pseudocapillaria ophisterni sp. n., is described from the intestine and rarely from the stomach of the swamp-eel, Ophisternon aenigmaticum Rosen et Greenwood, from Catemaco Lake, Veracruz, Mexico. In having both caudal lobes in the male interconnected by a distinct dorsal membrane, it belongs to the subgenus Ichthyocapillaria. It differs from the three species in this subgenus mainly in possessing either a distinctly longer spicule or a smaller length of oesophagus relative to body length. It also differs in host type and geographical distribution. P. ophisterni is the

Bicarbonate absorption in eel intestine: evidence for the presence of membrane-bound carbonic anhydrase on the brush border membranes of the enterocyte.

Bicarbonate absorptive fluxes through the isolated intestine of the European eel (Anguilla anguilla) were evaluated by the pH-stat method under short-circuited conditions. It was found that bicarbonate absorptive flux was dependent on the luminal Na+ and was inhibited by luminal 4-acetamido-4' stilbene-2-2' disulfonic acid (SITS; 2.5 x 10(-4) M) and luminal acetazolamide (10(-4) M), while luminal amiloride (1 mM) was without effect. Furthermore, by using brush border membrane vesicles (BBMV) isolated from eel intestine, the existence of two carbonic anhydrase (CA) isoforms, one tightly associa

Porrocaecum muraenesoxi n. sp. (Nematoda: Heterocheilidae) from the intestine of marine eel fish Muraenesox talabonoides (Bleeker).

A new species of the nematode genus Porrocaecum Railliet et Henry, 1912 obtained from the marine eel fish Muraenesox talabonoides (Bleeker) from Kakinada (Andhra Pradesh) is described. The specimens do not agree with the description of known species of the genus Porrocaecum, hence a new species Porrocaecum muraenesoxi has been established to describe these nematodes. The new species is distinguished by body size, location of nerve ring, length of intestinal caecum, number and arrangement of caudal papillae, spicules length, and tail conical in both sexes. Rectal glands present.

Treatment of fish parasites. 3. Effects of levamisole HCl, metrifonate, fenbendazole, mebendazole, and ivermectin on Anguillicola crassus (nematodes) pathogenic in the air bladder of eels.

The effects of five nematocidal drugs (levamisole HCl, metrifonate, fenbendazole, mebendazole, and ivermectin) on the nematode Anguillicola crassus, pathogenic in eels and recently introduced in Europe, were tested under in vivo conditions. The resulting tissue alterations were studied by means of light and electron microscopy. It was found that levamisole and metrifonate were most effective in a freshwater bath (1 mg/l) for 24 h. The 50% lethal dose of levamisole was 250 mg/l per 24 hours, whereas that of metrifonate was only 10 mg/l per 2 hours. Morphological studies of the nematodes showed

Adjuvanted Edwardsiella anguillarum vaccine confers protection and cross-protection against E. piscicida in Japanese eel (Anguilla japonica).

Edwardsiellosis, caused by Edwardsiella species, is a major bacterial disease in eel aquaculture in East Asia. However, effective vaccines for Japanese eel (Anguilla japonica) remain unavailable. In this study, a formalin-killed Edwardsiella anguillarum vaccine (FKC) was evaluated with two adjuvants: squalene-aluminum hydroxide (Squ) and Montanide™ ISA 760 VG (ISA). Protective efficacy varied among formulations, with survival increasing from FKC (26.7%-65%) to FKC-Squ (55%-85%) and highest in FKC-ISA (80%-95%). FKC-Squ induced early protection, whereas FKC-ISA provided sustained protection up

Effects of cold seawater pre-treatments on induction of early sexual maturation and sperm production in European eel (Anguilla anguilla).

To induce sexual maturation in captivity, eels rely on hormonal treatments, but this process is costly and time-consuming. As an alternative, different types of conditioning, also referred as pre-treatment, have been assessed to ease hormonal treatment response. Recent studies have shown that migrating eels experience a wide range of temperatures, varying from 12 °C at night to as low as to 8 °C during the day. Therefore, this study evaluates the effects of low-temperature (10 °C) seawater pre-treatments of different durations (2 and 4 weeks) on male eel reproduction. The eye, gonadosomatic an

Transcriptome Analysis of Host Anti-Vibrio harveyi Infection Revealed the Pathogenicity of V. harveyi to American Eel (Anguilla rostrata).

Vibrio harveyi, a recently discovered pathogenic bacterium isolated from American eels (Anguilla rostrata), poses uncertainties regarding its pathogenesis in American eel and the molecular mechanisms underlying host defense against V. harveyi infection. This study aimed to determine the LD50 of V. harveyi in American eel and assess the bacterial load in the liver, spleen, and kidney post-infection with the LD50 dose. The results showed that the LD50 of V. harveyi via intraperitoneal injection in American eels over a 14d period was determined to be 1.24 × 103 cfu/g body weight (6.2 × 104 cfu/fi

Immune responses and protective efficacy of American eel (Anguilla rostrata) immunized with a formalin-inactivated vaccine against Anguillid herpesvirus.

Anguillid herpesvirus 1 (AngHV), the causative agent of "mucus sloughing and hemorrhagic septicemia disease", causes serious infectious diseases in farmed eel. Among the effective prevention and control strategies, vaccination is one of the most effective approaches. However, no vaccine for AngHV is available. Our study developed a formalin-inactivated AngHV vaccine and evaluated its performance in American eels. Initially, AngHV-FJ, a strain of AngHV, was inactivated completely by 0.1 % formaldehyde, mixed with adjuvant Montanide ISA 763 A VG (763A). Then, vaccines containing different amount

Type of hormonal treatment administered to induce vitellogenesis in European eel influences biochemical composition of eggs and yolk-sac larvae.

Egg biochemical composition is among the main factors affecting offspring quality and survival during the yolk-sac stage, when larvae depend exclusively on yolk nutrients. These nutrients are primarily embedded in the developing oocytes during vitellogenesis. In aquaculture, assisted reproduction procedures may be applied enabling gamete production. For the European eel (Anguilla anguilla), reproductive treatment involves administration of pituitary extracts from carp (CPE) or salmon (SPE) to induce and sustain vitellogenesis. In the present study, we compared the influence of CPE and SPE trea

Daily variation of D2 dopamine receptor transcription in the brain of the Japanese eel Anguilla japonica and its regulation with dopamine and melatonin.

Dopamine plays a crucial role in controlling reproduction in eels, and its action is mediated through D2-type dopamine receptors. D2A and D2B receptors in the Japanese eel Anguilla japonica were cloned and characterized in the present study. Attention (daily expression patterns in the brain and endogenous regulation) was paid to D2B receptor because it is considered to play a crucial role in eel reproduction. The cDNAs of D2A and D2B receptors had open reading frames comprising 456 and 454 amino acid residues, respectively, which were phylogenetically clustered with those of other teleost spec

Molecular characterization, expression pattern and evolution of nine suppressors of cytokine signaling (SOCS) gene in the swamp eel (Monopterus albus).

Suppressors of cytokine signaling (SOCS) family members have negative effects on cytokine signaling pathways involved in immunity, growth and development. Owing to their typical feature, they have been extensively studied in mammalians, but they have not offered systematic studies among teleosts. In the present study, nine SOCS family genes were identified in the swamp eel genome and analyzed regulation mechanisms of SOCS family members in swamp eels. The open reading frames of MaSOCS1a, MaSOCS1b, MaSOCS2, MaSOCS3a, MaSOCS3b, MaSOCS4, MaSOCS5, MaSOCS6 and MaSOCS7 were 663 bp, 603 bp, 717 bp, 6

Branchial expression of an aquaporin 3 (AQP-3) homologue is downregulated in the European eel Anguilla anguilla following seawater acclimation.

A cDNA encoding the homologue of mammalian aquaporin 3 (AQP-3) was isolated by reverse transcription-polymerase chain reaction from the gill of the European eel. The derived amino acid sequence shares 67-70% homology with other vertebrate AQP-3 homologues. Northern blot analysis revealed two AQP-3-specific mRNA species of 2.4 kb and 7 kb. AQP-3 mRNA is expressed predominantly in the eye, oesophagus, intestine (as found in mammals) and the gill; no expression could be demonstrated in the stomach and only low and sporadic levels in the kidney. Quantitative studies demonstrated that, following th

Neuromuscular control of anguilliform locomotion: patterns of red and white muscle activity during swimming in the american eel anguilla rostrata.

Two areas that have received substantial attention in investigations of muscle activity during fish swimming are (1) patterns of fiber type recruitment with swimming speed and (2) the timing of muscle activation in relation to muscle strain. Currently, very little is known about either of these areas in eels, which represent an extreme body form among fishes and utilize a mode of locomotion found at one end of the undulatory spectrum (anguilliform locomotion). To assess how this swimming mode and body form influence the neuromuscular control of swimming, I recorded electromyographic data from

Prioritization based on risk assessment to study the bioconcentration and biotransformation of pharmaceuticals in glass eels (Anguilla anguilla) from the Adour estuary (Basque Country, France).

The presence of contaminants of emerging concern in the aquatic environment directly impacts water-living organisms and can alter their living functions. These compounds are often metabolized and excreted, but they can also be accumulated and spread through the food chain. The metabolized contaminants can also lead to the formation of new compounds with unknown toxicity and bioaccumulation potential. In this work, we have studied the occurrence, bioconcentration, and biotransformation of CECs in glass eels (Anguilla anguilla) using UHPLC-HRMS. To select the target CECs, we first carried out an

Japanese eel SOCS1 suppresses MyD88-mediated NF-κB activation: implications for negative regulation of innate immunity in teleosts.

Suppressor of cytokine signaling 1 (SOCS1) is a pivotal negative regulator of cytokine-mediated signaling in mammals, yet its role in teleost fish immunity remains poorly understood. In this study, we cloned and characterized the full-length SOCS1 cDNA from Japanese eel (Anguilla japonica), which encodes a 203-amino acid protein containing a conserved SH2 domain and SOCS box. Phylogenetic analysis placed AjSOCS1 within the teleost clade, showing high conservation with other vertebrate SOCS1 homologs.
