Ophthalmoplegia due to mitochondrial DNA disease: the need for genetic diagnosis.

We describe a patient with chronic progressive external ophthalmoplegia (CPEO) who underwent muscle biopsy for suspected mitochondrial disease. In spite of normal histocytochemical cytochrome c oxidase (COX) activity and respiratory chain enzyme measurements in muscle, subsequent molecular genetic analysis revealed the presence of a single, large-scale deletion of mitochondrial DNA (mtDNA). The case serves to illustrate the importance of pursuing the proposed mitochondrial genetic abnormality, even in patients with normal biopsy findings.

Effect of changing the detergent bound to bovine cytochrome c oxidase upon its individual electron-transfer steps.

The influence of the detergent environment upon individual electron-transfer rates of cytochrome c oxidase was investigated by stopped-flow spectrophotometry. The effects of three detergents were studied: lauryl maltoside, which supports a high turnover number (TN = 350 s-1), n-dodecyl octaethylene glycol monoether (C12E8), which supports an intermediate TN (150 s-1), and Triton X-100 in which oxidase is nearly inactive (TN = 2-3 s-1). Under limited turnover conditions (cytochrome c:cytochrome c oxidase ratio = 1:1 to 8:1), the rate of oxidation of cytochrome c was measured and compared with t

Polymyositis with cytochrome C oxidase negative fibers--a pathological and clinical challenge.

Polymyositis (PM) with cytochrome C oxidase negative fibers also referred to as PM with mitochondrial pathology (PM-Mito) is characterized by the symptoms of inclusion body myositis (IBM) and by the myopathological findings of PM except for an increase of muscle fibers with insufficient mitochondrial cytochrome C oxidase activity. Few PM-Mito cases are published; mitochondrial ultrastructure has not been studied in these patients. We report 2 PM-Mito patients with later onset than usually seen in IBM and poor responsiveness to glucocorticoids. Electron microscopy of muscle fibers showed irregu

The reaction of cytochrome oxidase with cyanide. Preparation of the rapidly reacting form and its conversion to the slowly reacting form.

The magnitude of the slow phase of reaction of cytochrome oxidase with cyanide has been correlated with the size of the epr signal at g' = 12. This epr signal was not found in submitochondrial particles, and significant g' = 12 epr was only observed late in the purification of solubilized enzyme. The Hartzell-Beinert procedure for the purification of cytochrome oxidase (Hartzell, C.R., and Beinert, H. (1974) Biochim. Biophys. Acta 368, 318-338) has been modified so that the purified enzyme reacts in a single rapid phase with potassium cyanide and lacks the g' = 12 epr signal. This enzyme could

Integrated 'omics analysis reveals new drug-induced mitochondrial perturbations in human hepatocytes.

We performed a multiple 'omics study by integrating data on epigenomic, transcriptomic, and proteomic perturbations associated with mitochondrial dysfunction in primary human hepatocytes caused by the liver toxicant valproic acid (VPA), to deeper understand downstream events following epigenetic alterations in the mitochondrial genome. Furthermore, we investigated persistence of cross-omics changes after terminating drug treatment. Upon transient methylation changes of mitochondrial genes during VPA-treatment, increasing complexities of gene-interaction networks across time were demonstrated,

The southward expansion of Anopheles farauti (Diptera: Culicidae) in Australia: a genetic and ecological niche modeling investigation.

The mosquito Anopheles farauti Laveran (1902) is a coastal malaria vector with a wide distribution throughout the Southwest Pacific. This species shows strong population genetic structure across Australia, New Guinea, and the Solomon Archipelago. In Queensland, Australia, its southern range limit has been the 21° latitude but, in 2023, it was detected in mosquito surveys 300 km further south, at 23° latitude. Additional collections undertaken in 2024 suggested that a population of An. farauti had established at this new southern limit. Sequencing of the mitochondrial cytochrome oxidase I (COI)

Genetic divergence and signatures of historical isolation of the traditionally harvested Amolops marmoratus from the Darjeeling Hill Region in the Himalaya Biodiversity Hotspot.

BACKGROUND: The Darjeeling Hill Region (DHR) has numerous freshwater bodies, such as hill streams, springs, and temporary water pools. It receives abundant rainfall and has extensive forest cover, which together play vital roles in sustaining frog diversity. Amolops marmoratus is among the notable hillstream frogs, known for its wide geographic distribution and status as a local delicacy. Thus, A. marmoratus is an important subject for studying genetic divergence and population structure along with anthropogenic pressure. This research investigates the standard morphological characteristics an

Getting to the Core of the Matter-Assessing the Role of Replication in Metabarcoding-Based sedaDNA.

Replication is central to most experimental and sampling designs, increasing inferential power and capturing fine-scale data heterogeneity. However, its importance remains poorly evaluated in some ecological and evolutionary settings. This is the case of metabarcoding studies using DNA recovered from sedimentary archives, in which biological signals integrate ecological information through depositional and burial processes, yet are commonly inferred from a single sediment core per site. Here, we evaluated the effect of different types of replication using sedimentary DNA metabarcoding data fro

Dirofilaria immitis in a cat and update on the geographic distribution of the infection in domestic felines in Brazil.

Feline dirofilariosis caused by Dirofilaria immitis represents a diagnostic and clinical challenge in veterinary medicine. Heartworm-associated respiratory disease (HARD) is one of the clinical manifestations observed in infected cats. In many cases, cats may die suddenly without suggestive clinical signs. This report describes the first natural D. immitis infection in a cat from the state of Pernambuco, and provides an update on the geographic distribution of this infection in domestic felines in Brazil. In June 2024, the body of a cat that had died suddenly was submitted for necropsy. The ma

Application of COI DNA barcode to identify phlebotomine sand flies (Diptera: Psychodidae) from Maranhão in leishmaniasis-endemic areas, Northeast Brazil.

This study evaluated the effectiveness of DNA barcoding, based on the mitochondrial COI gene, in delimiting species of the subfamily Phlebotominae from Maranhão, in addition to generating new molecular information and detecting possible cryptic species. Fragments of the COI gene were sequenced and integrated into public GenBank data. The species was delimited by means of species delimitation algorithms based on genetic distance (ABGD and ASAP) and phylogenetic gene trees (PTP), making it possible to identify patterns of cryptic diversity. In total, 54 consensus sequences were obtained, represe

Taxonomy and systematics of the Australasian gum nut orb-weaving spider genus Carepalxis (Araneae, Araneidae).

We revised the orb-weaving spider genus Carepalxis L. Koch, 1872 and tested its monophyly using Maximum Likelihood and Bayesian Inference phylogenetic analyses, comparing our results to a previously published family-level dataset on world-wide Araneidae. We studied the placement of the genus and the classification of the informally termed clade 'backobourkiines' using phylogenetic analyses based on two mitochondrial genes, cytochrome c oxidase subunit 1 (COI ) and 16S rRNA (16S ), and two nuclear genes, 28S rRNA (28S ) and 18S rRNA (18S ). The Canine Hookworm Ancylostoma Caninum: First Confirmed Evidence in a Dog in Central Europe: Epidemiological Relevance or Coincidence?

Canine hookworms represent some of the most globally prevalent parasitic nematodes affecting dogs and pose a significant zoonotic risk to humans, in whom they can induce cutaneous larva migrans. Infection with these parasites may lead to blood loss, anaemia, and, in severe cases, mortality-particularly in young puppies. The present study reports a confirmed case of Ancylostoma caninum infection in a 15-month-old dog in the Slovak Republic. The infected dog presented with severe, persistent diarrhoea, and haematological examination revealed a decrease in erythrocyte and haemoglobin levels, as w

Integrative morphological, cytogenetic and molecular characterization of the Andean climbing catfish Astroblepus mindoensis (Regan, 1916) (Siluriformes:Astroblepidae).

Astroblepus species, commonly known as Andean climbing catfish, exhibit a unique challenge in species delimitation, leading to ongoing taxonomic debates. Here we report data on Astroblepus mindoensis, a vulnerable species endemic to Ecuador, obtained by an integrative approach that includes cytogenetic analysis, molecular identification of the specimens, and recording of morphological and morphometric characters useful for species diagnosis. Thus, this study aimed to associate the karyotype data of the specimens analyzed with morphological and molecular characters, improving and expanding the

A new species of lungworm from the Atlantic Forest: Rhabdias megacephala n. sp. parasite of the endemic anuran Proceratophrys boiei.

Rhabdias are lung-dwelling parasites of anurans and some reptiles. Currently, 93 species are known to exist worldwide. The identification of Rhabdias species is based mainly on morphological traits of hermaphroditic females that generally have a very conserved morphology. However, different approaches, such as the combination of morphological, molecular, and ecological data, have provided advances in identifying and delimiting rhabdiasid species. Here, we describe a new species of Rhabdias from the south of Brazil, with morphological and molecular data. The new species is distinguished from it

Effect of the mitochondrial membrane potential on the absorbance of the reduced form of cytochrome c oxidase.

The effect of mitochondrial membrane potential (ΔΨm) on the absorbance of the reduced cytochrome c oxidase (COX) was evaluated in isolated rabbit heart mitochondria using integrating sphere optical spectroscopy. Maximal reduction of the mitochondrial cytochromes was achieved by either blowing nitrogen to remove oxygen, or by adding cyanide. Gradual depolarization of ΔΨm by adding increasing concentrations of uncoupler resulted in an increase of up to 50 % in the absorbance of cytochrome aa3 under nitrogen saturation, and of 25 % with cyanide. Cytochrome aa3 absorbance increases were also obser
