To study the role of viruses of the California encephalitis virus complex (the family Bunyaviridae) in infectious pathology, 187 fever patients admitted to the Clinical Infectious Hospital in May-September 1986 were examined. In 10 of these patients the neutralization test revealed the presence of diagnostically significant changes in neutralizing antibodies (neutralization indices), which was indicative of the role played by Tahyna virus or other related viruses belonging to the California encephalitis virus complex in the etiology of the diseases. The analysis of the clinical picture showed

Thirty-four strains of Asian and Pacific mosquitoes belonging to 22 species of 7 genera were compared for oral and/or parenteral susceptibility to infection with 1 or more strains of each of the 4 dengue serotypes. Surprisingly, several species of common man-biting Aedes were much more susceptible to oral infection with each of the 4 dengue serotypes than was Aedes aegypti. These species included Aedes albopictus and members of the scutellaris group of the subgenus Stegomyia found on South Pacific islands. Mosquito strains and species relatively susceptible to 1 dengue serotype usually were re

The relationships among virus-specified proteins of Murray Valley encephalitis (MVE), Kunjin (KUN) and West Nile (WN) viruses were investigated by peptide mapping of exhaustive proteolytic digests of radioactively labelled polypeptides. Maps of the three structural proteins (E, C and M) derived from purified virions and of two non-structural proteins (NV5 and NV4) obtained from infected cells were compared. For each polypeptide considered, the peptide maps of the KUN and WN virus-specified proteins were more similar to each other than either was to the map of the corresponding MVE virus-specif

The results of virological and morphological study of the pathogenesis of the infection induced by viruses of the tick-borne encephalitis complex (attenuated TP-21 and E-30 strains) in Syrian hamsters with normal and suppressed immune response are presented. The data obtained indicate long-term persistence of the virus-specific antigen: sufficiently high titres of specific antibodies in the blood serum of the animals at late periods after virus inoculation, positive immunoenzyme tests for virus antigen in the meninges of the hamsters inoculated with the TP-21 strain. Features of the pathogenes

Studies on cloning of continuous HEp-2 and SPEV cell lines were carried out. The sensitivity of the resulting clones to tick-borne encephalitis virus was determined and the clone lines were shown to be heterologous in their sensitivity to TBE virus by means of the immunofluorescence and virological methods. Among 47 clones of HEp-2 line virus reproduction was observed in 30 clones, no reproduction was demonstrated in 17 clones. The maximum number of cells simultaneously synthesizing virus antigen did not exceed 35% of the population which was conformity with the results obtained in the study o

As arthropod-borne viruses continue to threaten populations globally, there is a pressing need for experimental systems that enable rapid antiviral discovery. Reverse-genetics platforms producing recombinant reporter orthoflaviviruses have been developed to address this gap. Here, we present two new recombinant tick-borne encephalitis viruses (TBEVrec) generated on a European-subtype Haselmühl Tiho1 isolate backbone. A reporter gene, either eGFP or Nluc, was inserted in the capsid-coding region of the genome downstream of the capsid RNA regulatory signal and separated from the complete viral p…

A diverse group of RNA viruses including Rabies, Polio, La Crosse, West Nile, Zika, Nipah, Eastern and Western equine encephalitis, Venezuelan equine encephalitis, Japanese encephalitis, and tick-borne encephalitis viruses have the ability to gain access to and replicate in the central nervous system (CNS), causing severe neurological disease. Current treatment for these patients is generally limited to supportive care. To address the need for a generalizable antiviral, we utilized a strategy of mutagenesis to limit virus replication. We evaluated ribavirin (RBV), favipiravir (FAV) and N 4 -hy…

Alphavirus replicons were evaluated as potential vaccine candidates for Venezuelan equine encephalitis virus (VEEV), western equine encephalitis virus (WEEV), or eastern equine encephalitis virus (EEEV) when given individually or in combination (V/W/E) to mice or cynomolgus macaques. Individual replicon vaccines or the combination V/W/E replicon vaccine elicited strong neutralizing antibodies in mice to their respective alphavirus. Protection from either subcutaneous or aerosol challenge with VEEV, WEEV, or EEEV was demonstrated out to 12 months after vaccination in mice. Individual replicon v

OBJECTIVE: To explore on the immunity of live attenuated Japanese Encephalitis (JE) vaccine (SA14-14-2) to different wild JE virus (JEV) strains.
METHODS: The neutralizing effect of the vaccine against different wild JE virus strains was detected by plaque reduction neutralization test (PRNT), and the immunogenicity was studied on mice by vaccination -challenge protection test. In the PRNT, pooled sera from vaccinated human were tested against 10 strains of JEV, one isolated in Taiwan and 9 from other Asian countries. In the vaccination challenge test, mice received one dose of the live vacci

The Japanese encephalitis vaccine virus (JE 2-8) and its parent virus (JE SA 14) strains were compared for their ability to infect, to replicate in, and to be transmitted by Culex tritaeniorhynchus mosquitoes. Both viruses replicated in the mosquitoes after intrathoracic infection. The JE 2-8 vaccine was transmitted to 1/36 (3%) of suckling mice bitten by intrathoracically infected mosquitoes; however, the parent JE SA 14 strain was transmitted to 46/46 (100%) of the mice bitten. In oral infection trials, only 4/36 (11%) of the Cx. tritaeniorhynchus mosquitoes ingesting the vaccine JE 2-8 stra

We determined the nucleotide and deduced amino acid sequences of the complete envelope (E) protein gene of the five tick-borne encephalitis virus (TBEV) strains KrM 93, KrM 213, KrM 215, KrM 216, and KrM 219, isolated from wild rodents in South Korea. We analyzed genetic variability within the isolates and compared them with 13 other TBEV strains. The complete E protein genes were amplified by reverse transcription polymerase chain reaction (RT-PCR), cloned into pGEM-T vectors, and sequenced. The five isolates were similar to the Western subtype in nucleotide and deduced amino acid sequences (

The significant antigenic crossovers between West Nile virus (WNV) and tick-borne encephalitis virus (TBEV) make the immunological diagnosis of these diseases difficult. The avidicity index of virus-specific class G immunoglobulins (IgG) was used as a criterion for the differentiation of an immune response to WNV or TBEV in patients and convalescents. The panels of the sera sampled from patients with tick-borne encephalitis and convalescents in the Novosibirsk and Tomsk Regions and in the Primorye Territory and from those with West Nile fever and convalescents in the Volgograd Region. The dete

House finches, Carpodacus mexicanus, were experimentally infected with high and standard doses of western equine encephalomyelitis virus (WEEV) or St. Louis encephalitis virus (SLEV) to determine whether high doses would produce an elevated viremia response and a high frequency of chronic infections. Finches inoculated with approximately100,000 plaque forming units (PFU) of WEEV or SLEV produced viremia and antibody responses similar to those in finches inoculated with approximately 100 PFU of WEEV or 1000 PFU of SLEV, the approximate quantities of virus expectorated by blood-feeding Culex tar
