The role of the endoplasmic reticulum stress response following cerebral ischemia.

Background Cornu ammonis 3 (CA3) hippocampal neurons are resistant to global ischemia, whereas cornu ammonis (CA1) 1 neurons are vulnerable. Hamartin expression in CA3 neurons mediates this endogenous resistance via productive autophagy. Neurons lacking hamartin demonstrate exacerbated endoplasmic reticulum stress and increased cell death. We investigated endoplasmic reticulum stress responses in CA1 and CA3 regions following global cerebral ischemia, and whether pharmacological modulation of endoplasmic reticulum stress or autophagy altered neuronal viability . Methods In vivo: male Wistar ra

The Roles of Endoplasmic Reticulum Stress in the Pathophysiological Development of Cartilage and Chondrocytes.

Endoplasmic reticulum (ER) has evolved an adaptive mechanism called unfolded protein response (UPR) at the initial stage to restore cellular homeostasis. The three ER transmembrane sensors, such as IRE1α, PERK, and ATF6, are the key factors to decide cell fates. They exhibit both advantageous and disadvantageous effects, depending on the micro-environmental state of cells. ER stress has been implicated in chondrocytes proliferation, differentiation, and hypertrophy through regulating transcriptional factors SOX9, Ihh, BMP-2, RUNX, and HIF1/2α. In addition, the chronic ER stress induced by the

Endoplasmic reticulum stress and the related signaling networks in severe asthma.

The endoplasmic reticulum (ER) is a specialized organelle that plays a central role in biosynthesis, correct protein folding, and posttranslational modifications of secretory and membrane proteins. Loss of homeostasis in ER functions triggers the ER stress response, resulting in activation of unfolded protein response (UPR), a hallmark of many inflammatory diseases. These pathways have been reported as critical players in the pathogenesis of various pulmonary disorders, including pulmonary fibrosis, lung injury, and chronic airway disorders. More interestingly, ER stress and the related signal

Vesicularization of the endoplasmic reticulum is a fast response to plasma membrane injury.

The endoplasmic reticulum of most cell types mainly consists of an extensive network of narrow sheets and tubules. It is well known that an excessive increase of the cytosolic Ca(2+) concentration induces a slow but extensive swelling of the endoplasmic reticulum into a vesicular morphology. We observed that a similar extensive transition to a vesicular morphology may also occur independently of a change of cytosolic Ca(2+) and that the change may occur at a time scale of seconds. Exposure of various types of cultured cells to saponin selectively permeabilized the plasma membrane and resulted

Endoplasmic reticulum stress in the pathogenesis of early-onset pre-eclampsia.

Recent data have provided molecular evidence of high levels of endoplasmic reticulum stress in non-laboured placentas from cases of early-onset pre-eclampsia. Endoplasmic reticulum stress is intricately linked to oxidative stress, and the two often share the same aetiology. In the case of pre-eclampsia this is likely to be placental malperfusion, secondary to deficient conversion of the spiral arteries. Endoplasmic reticulum stress activates a number of signalling pathways aimed at restoring homeostasis, but if these attempts fail then the apoptotic machinery may be activated. The potential co

Further assembly required: construction and dynamics of the endoplasmic reticulum network.

The endoplasmic reticulum (ER) is a continuous membrane system comprising the nuclear envelope, ribosome-studded peripheral sheets and an interconnected network of smooth tubules extending throughout the cell. Although protein biosynthesis, transport and quality control in the ER have been studied extensively, mechanisms underlying the notably diverse architecture of the ER have only emerged recently; this review highlights these new findings and how they relate to ER functional specializations. Several protein families, including reticulons and DP1/REEPs/Yop1, harbour hydrophobic hairpin doma

Endoplasmic reticulum overcrowding as a mechanism of beta-cell dysfunction in diabetes.

This study suggests a molecular mechanism that explains the accumulation of denaturated proinsulin in the endoplasmic reticulum (ER) of beta-cells. Such states were frequently observed in beta-cells experiencing increased demand for insulin production and were shown to lead to secretory dysfunction and diabetes. Here, a self-consistent kinetic model is used to investigate changes in protein translation due to ER overloading. The model is based on a molecular theory that relates the molecular composition and level of molecular crowding in the ER to the kinetic rates of protein folding/misfoldin

Rrs1 is involved in endoplasmic reticulum stress response in Huntington disease.

The induction of Rrs1 expression is one of the earliest events detected in a presymptomatic knock-in mouse model of Huntington disease (HD). Rrs1 up-regulation fulfills the HD criteria of dominance, striatal specificity, and polyglutamine dependence. Here we show that mammalian Rrs1 is localized both in the nucleolus as well as in the endoplasmic reticulum (ER) of neurons. This dual localization is shared with its newly identified molecular partner 3D3/lyric. We then show that both genes are induced by ER stress in neurons. Interestingly, we demonstrate that ER stress is an early event in a pr

The endoplasmic reticulum is a key component of the plasma cell death pathway.

Plasma cells (PC) are the effector cells of the humoral Ab response. Unlike other dedicated secretory cells, they exist as two populations with opposite cell fates: short-lived and long-lived PC. Upon transformation they lead to an incurable neoplasia called multiple myeloma. In this study we have explored the molecular mechanism of PC death. Our data show that their apoptotic pathway is unique among other hemopoietic cells inasmuch as neither the death receptors nor the mitochondria play the central role. PC apoptosis is initiated by activation of Bax at the endoplasmic reticulum membrane and

Imaging endoplasmic reticulum calcium with a fluorescent biosensor in transgenic mice.

The use of biosynthetic fluorescent sensors is an important new approach for imaging Ca(2+) in cells. Genetically encoded indicators based on green fluorescent protein, calmodulin, and fluorescence resonance energy transfer (FRET) have been utilized to measure Ca(2+) in nonmammalian transgenic organisms and provide information about the organization and regulation of Ca(2+) signaling events in vivo. However, expression of biosynthetic FRET-based Ca(2+) indicators in transgenic mammals has proven to be problematic. Here, we report transgenic expression of an endoplasmic reticulum (ER) Ca(2+) bi

Rubella virus glycoprotein interaction with the endoplasmic reticulum calreticulin and calnexin.

Very little is known about the cellular factors that are required for the maturation of rubella virus glycoproteins (E2 and E1) in the endoplasmic reticulum of the infected cell. In the present study, we established the interaction of the ER chaperone proteins, calreticulin and calnexin, with the RV E1 and E2 proteins in cells stably expressing the viral proteins. The interaction between E2 and calnexin was significantly higher than with calreticulin. In pulse-chase experiments, the half-life of the E2-calnexin was >45 min, whereas the half-life of the calreticulin-E2 interaction was approxima

Warfarin causes the degradation of protein C precursor in the endoplasmic reticulum.

Warfarin, an antagonist of vitamin K, is known to disrupt the microsomal vitamin K cycle, which results in a decrease in the plasma level of protein C, an anticoagulant factor, as well as some other vitamin K-dependent coagulation factors. Here, we examined the effect of warfarin on the secretion of recombinant protein C expressed in human kidney 293 or BHK cells. In transient expression, warfarin caused a 2-4-fold decrease in the quantity of protein C secreted, compared to findings with vitamin K-treated cells. Pulse-chase experiments using stable cells showed that, although recombinant prote

The hidden hand of endoplasmic reticulum stress in anticancer drug resistance.

Endoplasmic reticulum stress (ERS) and the unfolded protein response (UPR) play pivotal roles in cancer adaptation and drug resistance. Under stress, dissociation of BiP from ER stress sensors activates three pathways such as PERK, IRE1α, and ATF6, which together promotes tumor cell survival. The PERK-eIF2α-ATF4 axis also suppresses global protein synthesis by inhibiting cyclin D, induces G1 arrest, and selectively enhances pro-survival gene translation. Simultaneously, IRE1α splices XBP1 mRNA to generate XBP1s, augmenting chaperone production, endoplasmic-reticulum-associated degradation (ERA

The endoplasmic reticulum protein FAM134B acts as a regulator of mitochondrial morphology.

The endoplasmic reticulum (ER) and mitochondria are known to affect myriad cellular mechanisms processes. More recently, dynamic association between them has been identified in different eukaryotes; these interactions vary in their composition and involvement in regulation of intracellular machineries. FAM134B (also known as RETREG1), originally identified as an oncogene, regulates ER membrane shape and curvature. It is a key ER-phagy or reticulophagy receptor, which promotes autophagy of not only the ER but also simultaneous dual autophagy of ER and mitochondria. Although it is known that FAM

The role of LECT2 in kidney fibrosis progression and endoplasmic reticulum stress.

AIMS: Our previous studies showed that Leukocyte Cell-Derived Chemotaxin 2 (LECT2), as a ligand for Tie1, modulates vascular endothelial cell function, promoting hepatic fibrosis. These findings prompted an investigation into whether LECT2 effects kidney fibrosis. This study aimed to elucidate the role of LECT2 in kidney fibrosis and its regulation of C/EBP homologous protein (CHOP) expression. MATERIALS AND METHODS: We utilized Lect2-KO mice, Lect2-2 A-Cre-Rosa26-LSL-tdTomato reporter mice, and EA.hy926 cells overexpressing LECT2 to establish models of kidney fibrosis and endoplasmic reticul

Endoplasmic reticulum in the axon: Insights into structural dynamics and implications in neurodegeneration.

The endoplasmic reticulum (ER) is an interconnected and highly dynamic organelle essential for multiple cellular functions. In neurons, the ER extends into axons, where it plays a pivotal role in maintaining neuronal polarity. The unique structural and dynamic adaptations of the axonal ER enable it to meet the specialized demands of neurons, ranging from compartmentalized physiological regulation to long-distance intracellular communication. Recent studies have shown that axonal ER supports the regulation of organelle remodeling and trafficking in a spatiotemporal manner, processes that become

The UPRising connection between endoplasmic reticulum stress and the tumor microenvironment.

The tumor microenvironment (TME) represents a dynamic network of cancer cells, stromal cells, immune mediators, and extracellular matrix components, crucial for cancer progression. Stress conditions such as oncogene activation, nutrient deprivation, and hypoxia disrupt the endoplasmic reticulum (ER), activating the unfolded protein response (UPR), the main adaptive mechanism to restore ER function. The UPR regulates cancer progression by engaging cell-autonomous and cell-non-autonomous mechanisms, reprogramming the stroma and promoting immune evasion, angiogenesis, and invasion. This review ex
