Differences between the fecal microbiota of coeliac infants and healthy controls.

Coeliac disease (CD) is an immune-mediated enteropathy with a multifactorial aetiology, characterized by chronic inflammation of the small intestinal mucosa. Although evidence suggests that the gut microbiota contributes to other chronic inflammatory disorders, its possible role in CD has not been determined. In this study, the composition of the fecal microbiota of coeliac children and age-matched controls was investigated by culture-dependent and -independent methodologies, using fluorescent in situ hybridization (FISH). The levels of Bacteroides, Clostridium and Staphylococcus were signific

The presence of antibody-coated anaerobic bacteria in asymptomatic bacteriuria during pregnancy.

Quantitative anaerobic culture of urine samples obtained from 593 pregnant women by suprapubic bladder aspiration was performed to establish the involvement of anaerobic bacteria in asymptomatic urinary tract infections. The fluorescent antibody (FA) test was applied to the sediments of bladder aspirates to determine the site of infection. Anaerobic bacteriuria (greater than or equal to 10(4) microorganisms/ml of urine) was found in 34 patients, of whom five were FA-positive. These anaerobes were identified as Lactobacillus minutus, Veillonella parvula (two patients). Clostridium putrefaciens,

Dietary advanced glycation end products induced anxiety- and depression-like behaviors in female C57BL/6 mice and the ameliorative effects of quercetin.

Quercetin is a flavonoid bioactive compound with potential anti-depression effect. Dietary advanced glycation end-products (AGEs) might be critically associated with depression. We aimed to explore whether quercetin ameliorates dietary AGEs-induced anxiety and depression-like behaviors in female mice, with a focus on hypothalamic-pituitary-adrenal (HPA) axis regulation and gut microbiota composition. Mice were divided into three groups: control, dietary AGEs, and AGEs plus quercetin. Dietary AGEs induced anxiety and depression-like behavioral effects, reduced brain-derived neurotrophic factor,

Ferulic acid ameliorates metabolic dysfunction associated with fatty liver disease by regulating macrophage polarization through spermidine derived from gut microbiota.

OBJECTIVES: This study aimed to investigate the efficacy and mechanism of Ferulic acid (FA) in a high-fat diet (HFD)-induced murine model of Metabolic dysfunction-associated steatotic liver disease (MASLD), focusing on the gut microbiota-polyamine-macrophage polarization axis. METHODS: A murine MASLD model was induced with an HFD and treated with FA. We evaluated its efficacy through serological and histopathological analyses. Key mechanisms, including macrophage polarization and gut microbiota, were investigated using molecular analyses and 16S rRNA gene sequencing. The direct effect of sper

Characterization of oral microbiome alterations in thalassemia patients using 16S rRNA gene sequencing.

BACKGROUND: This study investigates the composition of the oral microbiota in thalassemia patients and compares it with healthy individuals from Saudi Arabia. Thalassemia is an inherited blood disorder characterized by reduced or absent production of globin chains, resulting in hypochromic microcytic anemia and multiple complications, such as iron overload resulting from frequent blood transfusions. METHODS: Using 16S rRNA gene sequencing, 28 saliva samples (14 thalassemia patients and 14 healthy controls) were analyzed to assess changes in the richness and diversity of the oral microbiota.

Butyrate-Producing Bacteria in Intestinal Disease Therapy: Potential and Challenges.

Butyrate-producing bacteria have emerged as keystone species whose metabolic activity orchestrates host-microbial homeostasis in the human gut. This review synthesizes current understanding of how these anaerobic Firmicutes, including Faecalibacterium prausnitzii, Roseburia spp., and Eubacterium rectale, function as key contributors to intestinal health through convergent mechanisms: serving as the primary energy source for colonocytes, enforcing mucosal hypoxia that excludes facultative pathogens, and modulating immunity via histone deacetylase inhibition and G-protein-coupled receptor signal

Mucosal Colonized Engineered Nissle 1917 for Targeted Gut Delivery and Restored Microbiota Homeostasis in Inflammatory Bowel Disease.

Inflammatory bowel disease (IBD) is a serious health problem with an increasing disease burden. Oral probiotics are a potential treatment for IBD. However, the limited colonization and lack of sustained activity in the inflamed colon are challenging. In this research, mucosal colonized engineered Nissle 1917 (EcN) was designed for targeted gut delivery and restored microbiota homeostasis in IBD. 2-iminothiolane thiolation was modified on the surface of EcN, thus the employed disulfide bonds can targeted bind with intestinal mucus layer, enhancing EcN activity and retention in the inflamed colo

Association between Gut Microbiota and Biological Aging: A Two-Sample Mendelian Randomization Study.

Recent observational studies revealed an association between gut microbiota and aging, but whether gut microbiota are causally associated with the aging process remains unknown. We used a two-sample Mendelian randomization approach to investigate the causal association between gut microbiota and biological age acceleration using the largest available gut microbiota GWAS summary data from the MiBioGen consortium and GWAS data on biological age acceleration. We further conducted sensitivity analysis using MR-PRESSO, MR-Egger regression, Cochran Q test, and reverse MR analysis. Streptococcus (IVW

Bidirectional Mendelian randomization to explore the causal relationships between the gut microbiota and male reproductive diseases.

Gut bacteria might play an important role in male reproductive disorders, such as male infertility and sperm abnormalities; however, their causal role is unclear. Herein, Mendelian randomization (MR)-Egger, weighted median, inverse variance weighting, Simple mode, and Weighted mode were used to test the causal relationship between gut microbes and male reproductive diseases. The MR results were validated using various metrics. The MR results were also consolidated using reverse causality speculation, conducted using two-way MR analysis and Steiger filtering. Biological function was analysed us

Comparison of gut microbiota compositions and corresponding genetic and metabolic features between guttate and plaque psoriasis by metagenomic sequencing.

BACKGROUND: Guttate psoriasis (GP) and psoriasis plaques (PP) are common subtypes of psoriasis. Previous studies have fully researched the association between psoriasis and gut microbiota. However, the differences in gut microbiota between GPs and PPs are still unknown. METHODS: Fecal samples were collected from 30 psoriatic patients (15 GP and 15 PP) and 15 healthy subjects. Metagenomic sequencing was then used to compare gut microbiota compositions and corresponding genetic and metabolic features between GP and PP. RESULTS: We found that the genus Megamonas was increased in PP and reduced

Influence of Short-Term Consumption of Hericium erinaceus on Serum Biochemical Markers and the Changes of the Gut Microbiota: A Pilot Study.

Hericium erinaceus (H. erinaceus) is widely studied as a medicinal and edible fungus. Recent studies have shown that H. erinaceus has protective effects for diseases, such as inflammatory bowel disease and cancer, which are related to gut microbiota. To investigate the benefits of H. erinaceus intake on gut microbiota and blood indices in adulthood, we recruited 13 healthy adults to consume H. erinaceus powder as a dietary supplement. Blood changes due to H. erinaceus consumption were determined by routine hematological examination and characterized by serum biochemical markers. Microbiota com

Molecular profiling of oral microbiota in jawbone samples of bisphosphonate-related osteonecrosis of the jaw.

OBJECTIVE: Infection has been hypothesized as a contributing factor to bisphosphonate (BP)-related osteonecrosis of the jaw (BRONJ). The objective of this study was to determine the bacterial colonization of jawbone and identify the bacterial phylotypes associated with BRONJ. MATERIALS AND METHODS: Culture-independent 16S rRNA gene-based molecular techniques were used to determine and compare the total bacterial diversity in bone samples collected from 12 patients with cancer (six, BRONJ with history of BP; six, controls without BRONJ, no history of BP but have infection). RESULTS: Denaturin

Bacteremia due to anaerobic bacteria in newborns.

Awareness of the role of anaerobic bacteria in neonatal bacteremia has increased in recent years. The incidence of recovery of anaerobes in neonatal bacteremia varies, according to different studies, between 1.8% and 12.5%. Of the 178 cases reported in the literature, 73 were due to Bacteroides species (69 were the Bacteroides fragilis group), 57 Clostridium species (mostly Clostridium perfringens), 35 Peptostreptococci, 5 Propionibacterium acnes, 3 Veillonella species, 3 Fusobacterium species, and 2 Eubacterium species. Predisposing factors were perinatal maternal complications (especially pr

Phylogeny of the Whipple's-disease-associated bacterium.

Efforts to culture and identify the intracellular bacteria associated with Whipple's disease have been unsuccessful. Nucleotide sequencing and amplification by the polymerase chain reaction was done on the bacterial 16 S ribosomal DNA present in a small-bowel biopsy specimen taken from a patient with Whipple's disease. A search by computer for similar rRNA sequences filed in databases showed the Whipple's-associated organism to be most similar to bacteria of the Rhodococcus, Streptomyces, and Arthrobacter genera, and more weakly related to mycobacteria. The biopsy specimen was estimated to con

[Current aspects of gas gangrene, apropos of 47 cases collected over a 3-year period (1974-1976)].

On the basis of 47 cases of gas gangrene collected over the three year period between 1974 and 1976, the authors review the circumstances surrounding its development, the clinical features and the prognosis of the disorder which remains grave despite a well-defined therapeutic protocol combining surgery, antibiotics and hyperbaric oxygen. There would appear to be a real resurgence of the disease at the present time. Post-traumatic and surgical aetiologies predominate, giving rise to two types of gangrene: clostridial gas gangrene secondary to contamined wounds, with a quasi-constant vascular e

Inulin enhanced rifaximin-inhibited colon cancer pulmonary metastasis by flora-regulated bile acid pathway.

Inulin as a natural polysaccharide regulates intestinal microorganisms, and improves the immune and gastrointestinal function. In order to explore the effect of inulin on pulmonary metastasis of colon cancer, we set up a CT26 injected pulmonary metastatic model. The results showed that inulin used alone did not improve pulmonary metastasis of colon cancer, while inulin combined with rifaximin significantly prolonged the survival time of mice, and inhibited pulmonary metastasis compared with model and inulin groups. Inulin treatment increased the abundance of harmful bacteria such as Proteobact

Microbial alpha diversity in the intestine negatively correlated with disease duration in patients with Meniere's disease.

Ménière's disease (MD) is characterized by loss of balance and hearing disorders. Although there is known to be endolymphatic hydrops involved in the pathological process, the pathogenesis of the disease is still largely unclear. Approximately half of patients with MD suffer from depressive symptoms and high levels of several stress hormones were observed in MD and depression, simultaneously. Recently, gut microbes have been shown to influence the function of the central nervous system in humans through their metabolites. We studied the intestinal microbiota of 10 patients with MD and 11 healt
