A 25-year Patent Landscape of Anti-Biofilm Strategies for Respiratory Tract Infections: Trends, Innovations, and Emerging Clinical Opportunities.

Respiratory tract infections (RTIs) remain a leading cause of global morbidity and mortality, and their clinical management is increasingly undermined by the biofilm mode of growth of major respiratory pathogens. Biofilm formation confers profound tolerance to conventional antibiotics through the extracellular polymeric substance (EPS) matrix, metabolic adaptations, and rapid acquisition of resistance, underscoring the need for new therapeutic strategies. Patents offer a valuable and timely insight into emerging technologies with translational potential. In this review, we analyse patents publ

Shear redistribution in confined biofilm systems: decoupling structure and function in engineered water environments.

Biofilms in engineered water systems often develop within confined, tortuous flow paths where shear stress is spatially heterogeneous. However, biofilm responses are commonly evaluated using bulk metrics that treat shear as uniform and externally imposed, overlooking how geometric confinement redistributes shear during biofilm growth. Here, we examined how confinement-driven shear redistribution governs biofilm-hydrodynamic interactions in labyrinth microchannels under constant pressure. By integrating time-resolved biofilm-altered geometries with flow simulations, we show that localized biofi

Artificial intelligence applications in biofilm research: computational approaches for biofilm analysis and antimicrobial prediction.

Biofilms are organized groups of microbes surrounded by an extracellular polymeric substance (EPS) matrix. This structure helps microbes survive, creates metabolic differences, and makes them less sensitive to antimicrobial treatments. Because biofilms can block antimicrobials and help microbes adapt, they often cause chronic and recurring infections that are hard to treat with standard methods. Most current diagnostic and antimicrobial testing methods focus on free-floating (planktonic) microbes and do not reflect the complex structure and behavior of mature biofilms. This gap often leads to

Biosurfactants as novel antibiofilm agents: mechanisms, biomedical applications, and applications in biofilm-associated infections.

Biofilm-related infections represent a long-standing problem in clinical and industrial environments because of their innate resistance to standard antimicrobials. Biosurfactants, which are amphiphilic and biodegradable compounds made by microorganisms, are showing great promise as anti-biofilm agents due to their wide-ranging antimicrobial effects and minimal toxicity. They function by reducing bacterial adhesion, destabilizing the structural integrity of the extracellular polymeric substance matrix, and enhancing membrane permeability, which may ultimately lead to cell lysis. The multifuncti…

Development of a flow-cell bioreactor for immobilized sulfidogenic sludge characterization using electrochemical H2S microsensors.

The sulfate-reduction process plays a crucial role in the biological valorization of SOx gases. However, a complete understanding of the sulfidogenic process in bioreactors is limited by the lack of technologies for characterizing the sulfate-reducing activity of immobilized biomass. In this work, we propose a flow-cell bioreactor (FCB) for characterizing sulfate-reducing biomass using H2S microsensors to monitor H2S production in real-time within a biofilm. To replace natural immobilization through extracellular polymeric substance production, sulfidogenic sludge was artificially immobilized

Evaluation of quantification methods to determine photodynamic action on mono- and dual-species bacterial biofilms.

The effect of photodynamic inactivation (PDI) sensitized by 5,10,15,20-tetra(4-N,N,N-trimethylammoniophenyl)porphyrin (TMAP4+) on different components of mono- and dual-species biofilms of Staphylococcus aureus and Escherichia coli was determined by different methods. First, the plate count technique showed that TMAP4+-PDI was more effective on S. aureus than E. coli biofilm. However, crystal violet staining revealed no significant differences between before and after PDI biofilms of both bacteria. On the other hand, the 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide method indic

Helicobacter pylori Biofilm and New Strategies to Combat it.

Helicobacter pylori, the most frequent pathogen worldwide that colonizes around 50% of the world's population, causes important diseases such as gastric adenocarcinoma, chronic gastritis, and gastric mucosa-associated lymphoid tissue (MALT) lymphoma. In recent years, various studies have reported that H. pylori biofilm may be one of the critical barriers to the eradication of this bacterial infection. Biofilms inhibit the penetration of antibiotics, increase the expression of efflux pumps and mutations, multiple therapeutic failures, and chronic infections. Nanoparticles and natural products c

Linking oxidative stress defense to biofilm architecture: Ohr mediates strain-dependent persistence in Acinetobacter baumannii.

Biofilm formation is critical for the persistence of Acinetobacter baumannii, yet its directional correlation with antimicrobial resistance remains paradoxical. Here, we confirmed our prior finding that drug-sensitive and multidrug-resistant (MDR) strains employ distinct, temporally regulated biofilm developmental programs: susceptible strains excel at rapid biomass accumulation but undergo early collapse, while MDR strains delay biofilm initiation and optimize for the maintenance and reinforcement of mature biofilms. Transcriptomic profiling identified the organic hydroperoxide resistance pro

Robust HPLC-MS/MS quantification of N-acyl homoserine lactones (AHLs) in biomass-associated solid matrices reveals their aqueous-solid distribution in wastewater treatment systems.

N-acyl homoserine lactones (AHLs) are quorum-sensing signals that regulate biofilm formation, extracellular polymeric substance production, and other collective microbial behaviors in biological wastewater treatment systems. However, reliable AHL quantification in biomass-associated solid matrices remains analytically challenging because sludge and biofilm extracts contain co-extracted constituents that cause severe matrix effects and chain-length-dependent recovery losses. Here, an ultrasonic extraction-liquid-liquid extraction (UE-LLE) workflow coupled with HPLC-MS/MS was developed and valid

Use of Micro/Nanorobots In Vivo for the Eradication of Bacterial Biofilm: A Review of Challenges and Strategies.

The term bacterial biofilm refers to a complex community of microorganisms embedded within a self-produced matrix of extracellular polymeric substances. This structural organization creates an environment that, when present in an infectious context within a living organism, limits the effectiveness of conventional antibiotic therapy. Consequently, such conditions substantially promote the development of antibiotic resistance. The decline in the discovery of novel antibiotic agents, coupled with a concurrent increase in the prevalence of multidrug-resistant microorganisms, has intensified the s

Biofilm modifiers: The disparity in paradigm of oral biofilm ecosystem.

A biofilm is a population of sessile microorganisms that has a distinct organized structure and characteristics like channels and projections. Good oral hygiene and reduction in the prevalence of periodontal diseases arise from minimal biofilm accumulation in the mouth, however, studies focusing on modifying the ecology of oral biofilms have not yet been consistently effective. The self-produced matrix of extracellular polymeric substances and greater antibiotic resistance make it difficult to target and eliminate biofilm infections, which lead to serious clinical consequences that are often l

Development and validation of a colorimetric assay for simultaneous quantification of neutral and uronic sugars.

A colorimetric assay based on the conventional anthrone reaction was investigated for specific quantification of uronic acids (UA) in the presence of neutral sugars and/or proteins. Scanning of glucose (Glu) and glucuronic acid (GlA) was performed after the reaction with anthrone and a double absorbance reading was made, at 560 nm and at 620 nm, in order to quantify the UA and neutral sugars separately. The assay was implemented on binary or ternary solutions containing Glu, GlA and bovine serum albumin (BSA) in order to validate its specificity towards sugars and check possible interference w

Candida albicans biofilm-induced vesicles confer drug resistance through matrix biogenesis.

Cells from all kingdoms of life produce extracellular vesicles (EVs). Their cargo is protected from the environment by the surrounding lipid bilayer. EVs from many organisms have been shown to function in cell-cell communication, relaying signals that impact metazoan development, microbial quorum sensing, and pathogenic host-microbe interactions. Here, we have investigated the production and functional activities of EVs in a surface-associated microbial community or biofilm of the fungal pathogen Candida albicans. Crowded communities like biofilms are a context in which EVs are likely to funct

Interface-dependent V. parahaemolyticus biofilm under varying temperatures, media, and oxygen conditions: implications for seafood safety.

Vibrio parahaemolyticus biofilms play a critical role in pathogen persistence in marine and seafood-processing environments, where oxygen availability, temperature, and surface interfaces vary widely. This study investigated biofilm development by three strains on partially submerged stainless-steel coupons under gas-liquid-wall (GLW) and fully submerged (SM) interfaces. Viable cell counts (log₁₀CFU/cm2) along with normalized protein concentration per viable cell (nProt) and normalized polysaccharide concentration per viable cell (nPol) were measured, under aerobic and anaerobic conditions acr

Transport and retention of biogenic selenium nanoparticles in biofilm-coated quartz sand porous media and consequence for elemental mercury immobilization.

Bacterial biofilms are structured cell communities embedded in a matrix of extracellular polymeric substances (EPS) and a ubiquitous growth form of bacteria in the environment. A wide range of interactions between biofilms and nanoparticles have been reported. In the present study, the influence of a mixed bacterial biofilm on retention of biogenic selenium nanoparticles (BioSeNPs) and consequences for immobilization of elemental mercury (Hg0) in a porous quartz sand system were examined. BioSeNPs were significantly retained in the presence of a biofilm through electrical double layer effects,

Fungal co-inoculation enhances drought tolerance and soil functioning during cyanobacterial biological soil crust development.

BACKGROUND: Soil moisture availability strongly constrains the establishment of cyanobacteria-based biological soil crusts (BSCs). This study investigated whether fungal co-inoculation could improve drought tolerance during biocrust development. METHODS: Cyanobacterial crusts (C) and cyanobacterial-fungal composite crusts (CF) were cultivated for 60 days under five soil-moisture levels (0-20%) using sand from the Hobq Desert, China. Chlorophyll a, chlorophyll fluorescence, soil nutrients, enzyme activities, EPS fractions, and GC-MS-based exometabolomics were measured. RESULTS: CF crusts main

Efficient nitrate removal by photo-assisted microbial denitrification using FexOy/g-C3N4-biofilm hybrid in a coiled flow inverter.

Nitrate contamination in aquatic systems poses serious environmental and public health challenge, necessitating efficient and sustainable denitrification technologies. In this study, a photo-assisted microbial denitrification system was developed by integrating an S-scheme FexOy/g-CN (FCN) photocatalyst with an in situ grown microbial biofilm in a coiled flow inverter (CFI) reactor. The FCN heterojunction enhanced visible-light absorption and charge separation, enabling efficient generation of photoinduced electrons for microbial nitrate reduction. The biofilm matrix provided a protective micr

Human Corneal Lenticules Decorated with Mesenchymal-Stem-Cells-Encapsulated Microneedles for Cornea Stroma Regeneration.

Human corneal stromal lenticules obtained from refractive surgeries have merged as a promising alternative for corneal regeneration, although their applications face challenges such as rejection reactions, inflammatory responses, and graft instability. To overcome these challenges, we developed decellularized human stromal lenticules integrated with microneedles, designed as a delivery matrix for mesenchymal stem cells (MSCs) to facilitate corneal regeneration. The microneedle patches consist of MSC-encapsulated methacrylated hyaluronic acid (HAMA) hydrogel needle tips with a corneal lenticule

Synergistic bactericidal effect of hot water with citric acid against Escherichia coli O157:H7 biofilm formed on stainless steel.

This study investigated the antimicrobial effect of hot water with citric acid against Escherichia coli O157:H7 biofilm on stainless steel (SS). Hot water (50, 60, or 70 °C) with 2% citric acid exhibited a synergistic bactericidal effect on the pathogen biofilm. It was revealed that hot water and citric acid combination induced sub-lethally injured cells. Additionally, mechanisms of the synergistic bactericidal effects of hot water with citric acid were identified through several approaches. In terms of biofilm matrix, hot water removes exopolysaccharides, a major component of extracellular po

Stiffness signatures along early stages of Xylella fastidiosa biofilm formation.

The pathogenicity of Xylella fastidiosa is associated with its systematic colonization of the plant xylem, forming bacterial biofilms. Mechanisms of bacterial transport among different xylem vessels, however, are not completely understood yet, but are strongly influenced by the presence of extracellular polymeric substances (EPS), which surrounds the assembly of cells forming the biofilm. In this work, we show quantitative measurements on the elastic properties of the system composed by EPS and bacterial cell. In order to investigate the mechanical properties of this system, force spectroscopy
