The skin test with hay extract or with Micropolyspora faeni (MF), is not commonly used in the diagnosis of Farmer's lung (FL), as it is not considered specific. In our study, we have applied the intracutaneous test with these antigens in 26 patients affected with FL; 18 of them were still in contact with the antigen and the remaining eight had not been in contact with the hay during the previous year. Twenty-five asymptomatic farmers (AF) served as a control group. In the first group and with hay extract, the immediate reading (I) was positive in 15 of 18 (83.3 percent), the late reaction (L) …

A cross-sectional epidemiological study was carried out on 2932 farmers (response rate 92%) living in Northern Italy. A questionnaire on respiratory symptoms was supplied, chest X-ray taken and simple spirometry tests made. Thirty-nine subjects reported attacks of breathlessness associated with fever after exposure to mouldy hay; thirty-six had a clinical history of farmer's lung and X-ray and/or respiratory function changes indicative of chronic farmer's lung; 113 were classified as having bronchial asthma. These 118 subjects and a random sample of 131 non-symptomatic subjects were examined b…

The leukocyte migration inhibition test (LMIT) was studied in 20 patients with farmer's lung (FL) and in 24 asymptomatic farmers. In the first group, the test proved to be 95% positive (19/20), whereas it was positive in only 44% (11/25) of the control group; this difference was statistically significant (p less than 0.005). The LMIT was also studied in eight patients with FL who had not been in contact with the antigen during the year prior to testing and was found to be 87% positive (7/8). The test therefore proved to be more effective in the diagnosis of FL compared with tests for specific …

The seasonal variation in the incidence of 78 consecutive cases of farmer's lung was investigated according to a statistical method described by Roger. The cyclic trend was statistically highly significant (P less than or equal to 0.0001) and the centre of gravity of incidence appeared in the latter half of April. The reasons for the variation are discussed.

Several methods were used to obtain serologically active materials from cultures of Micropolyspora faeni. From the results of immunodiffusion and immunoelectrophoresis tests on these materials it is suggested that preparations for the laboratory diagnosis of farmer's lung disease (FLD) should contain concentrated culture supernatant (CS) and extracts of mycelium obtained by ultrasonic treatment (MU). Although CS and MU have many serological activities in common they also possess activities unique to each.Extraction of mycelium with trichloracetic acid, boiling water or methanol yielded a produ

The isolation and physicochemical characterization of three of the `major' farmer's lung antigens derived from Micropolyspora faeni is recorded. Antigen 1, the major antigen in the C region is a glycopeptide with an average molecular weight of 85,000 and exhibits extreme heat stability being unaffected by 120°C for 7 days. Antigens 2 and 3, the major antigens in the A and B regions respectively are proteins whose molecular weights are 44,000 and 77,000 respectively and are thermolabile with half lives of 25 days and 20 days at 50°C respectively. The extreme thermolability of certain other anti
