Non-alcoholic fatty liver disease is characterised by a reduced polyunsaturated fatty acid transport via free fatty acids and high-density lipoproteins (HDL).

BACKGROUND AND OBJECTIVES: Non-alcoholic fatty liver disease (NAFLD) develops due to impaired hepatic lipid fluxes and is a risk factor for chronic liver disease and atherosclerosis. Lipidomic studies consistently reported characteristic hepatic/VLDL "lipid signatures" in NAFLD; whole plasma traits are more debated. Surprisingly, the HDL lipid composition by mass spectrometry has not been characterised across the NAFLD spectrum, despite HDL being a possible source of hepatic lipids delivered from peripheral tissues alongside free fatty acids (FFA). This study characterises the HDL lipidomic si

The Protective Effect of Camellia Bee Pollen on Alcoholic Fatty Liver in Zebrafish.

Background/Objectives: Camellia bee pollen refers to pollen pellets collected by bees from plant stamens and mixed with salivary secretions. Alcoholic fatty liver disease (AFLD), as the initial phase within the spectrum of alcohol-induced liver diseases, has resulted in a rising global incidence rate and treatment burden of such liver ailments. Methods: This study employs acute zebrafish juvenile and adult zebrafish chronic alcoholic liver models to explore the protective effects of camellia bee pollen as well as its ethanol and water extracts on zebrafish alcoholic fatty liver. Results: The r

Identification of Lactate-related Biomarkers in Non-alcoholic Fatty Liver Disease.

INTRODUCTION: Nonalcoholic fatty liver disease (NAFLD) is characterized by profound metabolic reprogramming. Recent evidence suggests that lactate, beyond its role as a metabolic waste product, may modulate histone lactylation, linking metabolic stress to the epigenetic regulation of disease progression. However, the specific lactate-related gene (LRG) signatures driving NAFLD remain to be elucidated. METHODS: We integrated transcriptomic mining of the GSE164760 dataset with in vivo validation to decode the LRG landscape. Differentially expressed LRGs were identified using strict thresholds a

Herbal medicine as a potential treatment for non-alcoholic fatty liver disease.

Non-alcoholic fatty liver disease (NAFLD) has emerged as a global public health concern, affecting over one-quarter of the global population. It is closely associated with the prevalence of obesity and metabolic syndrome. Current treatment options for NAFLD are limited and often have side effects. Traditional Chinese medicine (TCM) offers a promising alternative with its holistic approach and use of multi-component herbal formulations. A recent study explored the potential of the TCM formula, "Fanlian Huazhuo Formula (FLHZF)", to alleviate high-fat diet-induced NAFLD by regulating autophagy an

Correlation of organelle interactions in the development of non-alcoholic fatty liver disease.

Organelles, despite having distinct functions, interact with each other. Interactions between organelles typically occur at membrane contact sites (MCSs) to maintain cellular homeostasis, allowing the exchange of metabolites and other pieces of information required for normal cellular physiology. Imbalances in organelle interactions may lead to various pathological processes. Increasing evidence suggests that abnormalorganelle interactions contribute to the pathogenesis of non-alcoholic fatty liver disease (NAFLD). However, the key role of organelle interactions in NAFLD has not been fully eva

Cell and gene therapeutic approaches in non-alcoholic fatty liver disease.

Non-Alcoholic Fatty Liver Disease (NAFLD) refers to a range of conditions marked by the buildup of triglycerides in liver cells, accompanied by inflammation, which contributes to liver damage, clinical symptoms, and histopathological alterations. Multiple molecular pathways contribute to NAFLD pathogenesis, including immune dysregulation, endoplasmic reticulum stress, and tissue injury. Both the innate and adaptive immune systems play crucial roles in disease progression, with intricate crosstalk between liver and immune cells driving NAFLD development. Among emerging therapeutic strategies, c

The role of ubiquitination and deubiquitination in the pathogenesis of non-alcoholic fatty liver disease.

Nonalcoholic fatty liver disease (NAFLD) is one of the most common chronic liver diseases and is closely associated with metabolic abnormalities. The causes of NAFLD are exceedingly complicated, and it is known that a variety of signaling pathways, endoplasmic reticulum stress, and mitochondrial dysfunction play a role in the pathogenesis of NAFLD. Recent studies have shown that ubiquitination and deubiquitination are involved in the regulation of the NAFLD pathophysiology. Protein ubiquitination is a dynamic and diverse post-translational alteration that affects various cellular biological pr

Diagnosis and Management of Pediatric Non-Alcoholic Fatty Liver Disease: An Overview.

Non-alcoholic fatty liver disease (NAFLD) is the most common pediatric chronic liver disease worldwide, with an increasing prevalence, mainly due to the increase in childhood obesity and sedentary lifestyle. The pathogenesis of NAFLD is multifactorial, but the mechanisms by which the factors involved, namely the genetic, intrauterine and environmental factors responsible for its onset and progression to NASH, are not fully known. Children with NAFLD are usually asymptomatic or show nonspecific symptoms, and NAFLD is generally diagnosed incidentally by screening tests in overweight or obese chi

Management of Cardiovascular Risk in the Non-alcoholic Fatty Liver Disease Setting.

Non-alcoholic fatty liver disease (NAFLD) is an overlooked and undetected pathology, which affects more than 32% of adults worldwide. NAFLD is becoming more common in Western industrialised countries, particularly in patients with central obesity, type 2 diabetes, dyslipidaemia and metabolic syndrome. Although NAFLD has traditionally been interpreted as a liver disease with a high risk of liver-related complications, NAFLD is an underappreciated and independent risk factor for atherosclerotic cardiovascular disease, which is the principal cause of death in patients with NAFLD. Treatment option

Targeting histone methylation and demethylation for non-alcoholic fatty liver disease.

Non-alcoholic fatty liver disease (NAFLD) is the leading chronic liver disease worldwide, facing increasing challenges in terms of prevention and treatment. The methylation of lysine and arginine residues on histone proteins is dynamically controlled by histone methyltransferases (HMTs) and histone demethylases (HDMs), regulating chromatin structure and gene transcription. Mutations, genetic translocations, and altered gene expression involving HMTs and HDMs are frequently observed in NAFLD. HMTs and HDMs are receiving increasing attention in regulating NALFD. Targeting specific HMTs and HDMs
