Activity of plant flavonoids against antibiotic-resistant bacteria.

Thirty eight plant-derived flavonoids representing seven different structural groups were tested for activities against antibiotic-resistant bacteria using the disc-diffusion assay and broth dilution assay. Among the flavonoids examined, four flavonols (myricetin, datiscetin, kaempferol and quercetin) and two -flavones (flavone and luteolin) exhibited inhibitory activity against methicillin-resistant Staphylococcus aureus (MRSA). Myricetin was also found to inhibit the growth of multidrug-resistant Burkholderia -cepacia, vancomycin-resistant enterococci (VRE) and other medically important orga

Assessment of luteolin (3',4',5,7-tetrahydroxyflavone) neuropharmacological activity.

Since the discovery that certain flavonoids (namely flavones) specifically recognise the central BDZ receptors, several efforts have been made to identify naturally occurring GABA(A) receptor benzodiazepine binding site ligands. Flavonoid derivatives with a flavone-like structure such as apigenin, chrysin and wogonin have been reported for their anxiolytic-like activity in different animal models of anxiety. Luteolin (3',4',5,7-tetrahydroxyflavone) is a widespread flavonoid aglycon that was reported as devoid of specific affinity for benzodiazepine receptor (BDZ-R) binding site, but its psycho

Rosemary Diterpenes and Flavanone Aglycones Provide Improved Genoprotection against UV-Induced DNA Damage in a Human Skin Cell Model.

Overexposure to solar ultraviolet (UV) radiation is the major cause of a variety of cutaneous disorders, including sunburn, photoaging, and skin cancers. UVB radiation (290-320 nm) causes multiple forms of DNA damage, p53 induction, protein and lipid oxidation, and the generation of harmful reactive oxygen species (ROS). In recent years, botanicals containing polyphenols with antioxidant and anti-inflammatory properties as skin photoprotective agents have emerged. This study evaluated the protective effects of two formulations against UVB-induced damage in a skin cell model. One of the formula

Flavonoid modulation of murine neutrophil cytokinesis.

Flavonoids are known to produce a wide range of immunomodulatory effects. In this study, flavone and six hydroxylated analogues were examined for their effect on the FMLP-directed and the random migration of murine peritoneal exudate neutrophils. Flavone significantly (p less than 0.01) inhibits both directed and random migration at an assay concentration of 100 microM. In contrast, fisetin, kaempferol, chrysin, flavonol, morin and quercetin (in decreasing order of activity) significantly (p less than 0.05 to less than 0.01) enhance both directed and random migration at concentrations of 1.0 t

The thiocarbonyl analog of chrysin induces apoptosis and oxidative stress-mediated unfolded protein response in breast cancer cells.

Natural flavones such as chrysin and baicalein were characterized by a wide range of biological activities including, among others, antioxidant and anticancer action. Nevertheless, their use in biomedical applications is restricted due to limited bioavailability. In the present study, C4 thionation-based approach was considered to produce thiochrysin and thiobaicalein with improved anticancer activity. The thiocarbonyl analogs were tested against breast cancer cells with different receptor status (MCF-7 ER-positive, MDA-MB-231 TNBC, and SK-BR-3 HER2-positive) along with the corresponding norma

Nitric oxide suppresses apoptosis in human colon cancer cells by scavenging mitochondrial superoxide anions.

In cells lacking a functional p53 tumor suppressor protein, the endogenous free radical nitric oxide (NO) appears to inhibit apoptosis, and thereby promotes growth of cancer cells. In order to elucidate the underlying mechanisms on a molecular basis, we used HT-29 human colon carcinoma cells, carrying a p53 loss-of-function mutation, and examined the effects of NO on apoptosis when induced either by the flavonoid flavone or by the chemotherapeutic drug camptothecin (CPT), which is able to scavenge NO by superoxide anion production. Caspase-3 activation as well as nuclear fragmentation, both in

Transcriptome and Metabolome-Based Analysis Reveals the Molecular Mechanisms Underlying the Differences in Tanshinone and Salvianolic Acid Content Between Salvia miltiorrhiza Roots and Leaves.

Background/Objectives: Salvia miltiorrhiza (Danshen) is a well-known medicinal herb in traditional Chinese medicine. It produces tanshinones and salvianolic acids as key bioactive constituents in its roots, yet the molecular basis for their tissue-specific accumulation is still poorly understood. This study aims to identify candidate functional genes involved in the biosynthesis of tanshinones and salvianolic acids, and to reveal the molecular basis underlying their tissue-specific accumulation in S. miltiorrhiza. Methods: For this purpose, we compared transcriptomic and metabolomic difference

Striking Diversification of Exudate Profiles in Selected Primula Lineages.

In continuation of previous studies on glandular exudates of Primula, we analyzed eleven so far unstudied species and several populations for exudate composition. Unsubstituted flavone and unusually substituted flavones, normally predominant in Primula exudates, were not detected in all of the analyzed samples. Instead, some species exhibited regular substituted flavonoids, and in some cases, no flavonoids could be detected at all. The detection of a diterpene (1) in P. minima exudates is new to Primula. On basis of MS and NMR, 1 was structurally characterized as ent-kaur-16-en-19-oic acid. Co

Role of reactive oxygen intermediates in Japanese encephalitis virus infection in murine neuroblastoma cells.

Infection with Japanese encephalitis virus (JEV), a mosquito-borne, neurotropic flavivirus, may cause acute encephalitis in humans and induce severe cytopathic effects in various types of cultured cells. This study attempted to determine whether JEV infection induces free radical generation and whether oxidative stress contributes to virus-induced cell death in neuroblastoma cells. A rise in the intracellular level of free radicals indicated by the 2',7'-dichlorofluorescein fluorescence was observed in N18 cells following JEV infection. Cellular flavon-containing enzymes were involved in JEV-i

Flavanoids induce expression of the suppressor of cytokine signalling 3 (SOCS3) gene and suppress IL-6-activated signal transducer and activator of transcription 3 (STAT3) activation in vascular endothelial cells.

The atherogenic cytokine IL-6 (interleukin-6) induces pro-inflammatory gene expression in VECs (vascular endothelial cells) by activating the JAK (Janus kinase)/STAT3 (signal transducer and activator of transcription 3) signalling pathway, which is normally down-regulated by the STAT3-dependent induction of the E3 ubiquitin ligase component SOCS3 (suppressor of cytokine signalling 3). Novel treatments based on the regulation of SOCS3 protein levels could therefore have value in the treatment of diseases with an inflammatory component, such as atherosclerosis. To this end we carried out a scree
