Studies on fluoride concentrations in human submandibular/sublingual saliva and their relation to flow rate and plasma fluoride levels.

Submandibular/sublingual saliva and blood were collected from five subjects after ingestion of 1 mg fluoride as NaF. An individual collection device, made from a silicone impression material, was used to collect the saliva in 10-minute samples, before and during 2 hr after the fluoride intake. In two separate experiments on each individual, submandibular/sublingual saliva was collected continuously at different flow rates: without stimulation and with gustatory stimulation. Blood was also collected at intervals throughout the experiments. The concentration of fluoride in the submandibular/subl

Fluoride content in human iliac bone: results in controls, patients with fluorosis, and osteoporotic patients treated with fluoride.

The major part of fluoride ingested is fixed on calcified tissues, mainly in bone tissue, and then is progressively but slowly recycled during bone remodeling. Thus, the measurement of bone fluoride content allows the determination of the extent of bone fluoride retention, and this parameter constitutes a useful complement to bone histology for the diagnosis of skeletal fluorosis and could also be used for the management of fluoride treatment of osteoporosis. A simple method is described to measure the fluoride content in calcined human iliac bone samples. Bone ashes were diluted in perchloric

Surface enamel fluoride concentrations of different types of human teeth in a high- and near-optimal-fluoride area.

The surface enamel (approximately 4 micron) fluoride concentrations of different tooth types (11, 21, 22, 23, 24) were determined in 12-13-year-old children who were life-long residents in a naturally high (3.70 ppm) and in a near optimal (0.62 ppm) fluoride area. Enamel biopsies were taken by means of an acid-etch procedure, and the fluoride levels were measured with an adapted fluoride-ion-selective electrode. The enamel fluoride concentrations were related to the fluoride levels of the drinking water, and it appeared that the enamel fluoride concentrations of the different tooth types (exce

Relationship between plasma, dentin and bone fluoride concentrations in rats following long-term fluoride administration.

A detailed knowledge of fluoride (F) pharmacokinetics is necessary if its pharmacologic and toxicologic effects are to be adequately understood. In the present study plasma F concentrations have been analysed using repeated blood samples taken from the ethmoidal sinus of rats given 25, 50, 100 and 150 p.p.m. F in drinking water ad libitum for 79 days. A close relationship between the F-concentration in the drinking water and plasma F concentrations was found. When the administration of F was stopped, a rapid decline in plasma F concentration was noted during 48 hrs in all groups. The plasma F

Fluoridation and county-level secondary bone cancer among cancer patients 18 years or older in New York State.

The decision whether to fluoridate drinking water continues to be controversial in some communities. Dental and skeletal fluorosis in response to chronic fluoride overexposure are cited as reasons to avoid community water fluoridation in spite of evidence of the oral and skeletal health benefits fluoridation confers. Community fluoridation of ~ 1 mg/L fluoride has not been found to be associated with primary bone cancer but is associated with improved bone strength. No studies have examined fluoride exposure and secondary bone cancer, a common metastasis with significant morbidity. We hypothes…

Effect of silver diamine fluoride versus fluoride varnish in arresting early childhood caries: A clinical study.

The comparative effectiveness of silver diamine fluoride and fluoride varnish in arresting early childhood caries remains insufficiently established. Therefore, it is of interest to evaluate caries arrest among 90 children treated with either silver diamine fluoride or fluoride varnish. Hence, participants were divided into two groups, with clinical assessment of caries arrest at baseline and follow-up using standardized criteria. Silver diamine fluoride showed a higher caries arrest rate than fluoride varnish, reflecting its combined antimicrobial and remineralization effects. Thus, we show c

Evaluation of Fluoride Uptake of Two Fluoride Varnishes into and onto the Enamel Surface at Different Temperatures: An In Vitro Study.

BACKGROUND: Fluoride (F) is an effective anticaries agent and can be delivered through various mediums at different concentrations. The ability to increase the enamel resistance to acid by decreasing solubility through the incorporation of F into the enamel apatite structure is the primary function of these agents. The efficacy of topical F can be determined by measuring the amount of F incorporated in and on human enamel. AIM: To compare the F uptake into and onto the enamel surface of two different F varnishes at different temperatures. MATERIALS AND METHODS: In this study, 96 teeth were d

Amine fluoride/stannous fluoride and chlorhexidine mouthwashes as adjuncts to single-stage dental implants: a comparative study.

BACKGROUND: The growing popularity of non-submerged dental implants in recent years requires a greater emphasis on microbial plaque control. Chlorhexidine (CHX), the most commonly used mouthwash in implant surgery, is sometimes associated with tooth staining and alteration in taste perception. Amine fluoride/stannous fluoride (AmF/SnF2) mouthwash has been shown to have anti-infective properties; however, it has not been tested as an adjunct anti-infective means in non-submerged dental implants. The purpose of this trial was to compare AmF/SnF2 and CHX mouthwashes as adjuncts to single-stage de

The prevalence of dental fluorosis in a school-based program of fluoride mouthrinsing, fluoride tablets, and both procedures combined.

OBJECTIVES: The objective of the study was to describe and compare the prevalence and severity of dental fluorosis in children who participated in an eight-year clinical trial of the effectiveness of school-based fluoride procedures according to three treatment regimens and age of regimen initiation. METHODS: At baseline in 1981, 1,640 kindergarten and first grade children residing in a fluoride-deficient community (Springfield, OH) were assigned randomly to a group that (1) rinsed once a week with a 0.2 percent neutral NaF solution; (2) chewed, rinsed, and swallowed daily a neutral 2.2 mg Na

Characterization of stable beryllium fluoride, aluminum fluoride, and vanadate containing myosin subfragment 1-nucleotide complexes.

Beryllium and aluminum fluorides are good phosphate analogues. These compounds, like orthovanadate, form stable complexes with myosin subfragment 1 (S1) in the presence of MgADP. The formation of the stable S1-nucleotide complexes is characterized by the loss of ATPase activity. For the complete loss of ATPase activity there was necessary a higher concentration of aluminum than of beryllium or vanadate. In the presence of MgATP the onset of the inhibition is delayed, which indicates that stable complexes cannot form when a specific site is occupied by the gamma-phosphate of ATP or by P(i) deri

Otospongiosis and sodium fluoride. A clinical double-blind, placebo-controlled study on sodium fluoride treatment in otospongiosis.

The effect of sodium fluoride treatment in patients with otospongiosis has been evaluated in a prospective clinical double-blind, placebo-controlled study of 95 patients. The results showed a statistically significant greater deterioration of hearing loss in the placebo group than in the group actively treated with 40 mg of sodium fluoride daily. These results support the view that sodium fluoride can change otospongiotic, active lesions to more dense, inactive otosclerotic lesions.

The prevalence of fluorosis-like lesions associated with topical and systemic fluoride usage in an area of optimal water fluoridation.

The frequency of using self-applied (over-the-counter) fluoride products and prescribed supplemental fluorides, and the prevalence of fluorosis-like enamel lesions in the permanent dentitions of children living in an optimally fluoridated urban community were investigated. Subjects were selected randomly, and consent forms and questionnaires concerning fluoride usage and residency status were provided to guardians prior to examination. After a supervised tooth brushing event, 300 consenting subjects were examined by the principal investigator using Dean's index to assess levels of fluorosis-li

Otospongiosis and sodium fluoride. A blind experimental and clinical evaluation of the effect of sodium fluoride treatment in patients with otospongiosis.

The effect of sodium fluoride treatment in patients with otospongiosis has been evaluated blindly in a morphological and microchemical element analysis of otospongiotic specimens together with a prospective clinical double-blind, placebo-controlled study. The results show that using the calcium/phosphorus ratio as an indication for bone maturity, the sodium fluoride treatment can stabilize otospongiotic lesions in retaining calcium relative to phosphorus. The clinical double-blind, placebo-controlled study of 95 patients showed a statistically significant worse deterioration of the hearing los

From Evidence to Engagement: Understanding Online Resistance to Community Water Fluoridation for Public Health Communication-A Qualitative Study.

OBJECTIVES: This study explored how individuals in an online anti-CWF community construct and sustain opposition narratives to inform evidence-based communication and policy engagement. METHODS: A qualitative study was undertaken with adults who self-identified as not supportive of CWF. Fourteen participants from the United Kingdom, Australia, and the United States of America were recruited via the Fluoride Action Network Facebook page, a prominent global hub for anti-fluoridation discourse. Semi-structured interviews were conducted via Zoom between January and July 2025. Data were analyzed u…

Comparison of effects of diode laser and CO2 laser on human teeth and their usefulness in topical fluoridation.

Various authors have reported more effective fluoridation from the use of lasers combined with topical fluoride than from conventional topical fluoridation. Besides the beneficial effect of lasers in reducing the acid solubility of an enamel surface, they can also increase the uptake of fluoride. The study objectives were to compare the action of CO(2) and GaAlAs diode lasers on dental enamel and their effects on pulp temperature and enamel fluoride uptake. Different groups of selected enamel surfaces were treated with amine fluoride and irradiated with CO(2) laser at an energy power of 1 or 2…

[Is the reason for a decrease in dental caries after fluoridation in Basle the delay in tooth eruption?].

In several previous reports it was shown that water fluoridation in Basel has significantly reduced caries frequency in 7--15 year-old schoolchildren. This reduction is only for a small part caused by certain time differences in the eruption of the teeth. Neither can it be explained by instruction and motivation for better nutrition and home care, because mouth hygiene of the Basel schoolchildren has not improved over the last decade. Also, a reduction in sugar consumption could not be observed. Therefore, the conclusion seems justified that water fluoridation is mainly responsible for the red…

The Cariostatic Mechanisms of Fluoride-An Updated Review.

Fluoride remains the keystone of evidence-based caries prevention by stabilizing the mineral balance at the tooth-biofilm-saliva interface. Contemporary understanding emphasizes a predominantly post-eruptive, topical mode of action where fluoride inhibits demineralization and accelerates remineralization. This interfacial catalysis is reinforced by pH-responsive calcium-fluoride-like reservoirs that release fluoride during acid challenges. While community water fluoridation confers population-level reductions, the most effective approach is sustaining low-level fluoride in the biofilm environm…
