A Rosella-PLIN2 knock-in mouse reveals lipophagy and immunometabolic interplay in atherosclerosis.

Cytosolic lipid droplets (LDs) regulate lipid homeostasis, with abnormal LD dynamics linked to metabolic diseases like atherosclerosis. In macrophage foam cells, LDs undergo autophagic degradation via lipophagy, but the extent of this process in vascular smooth muscle cell (VSMC) foam cells remains unclear. To track lipophagy in real time, we developed a Rosella-PLIN2 (perilipin 2) biosensor by tagging PLIN2 with the fluorescent pH-biosensor Rosella. We show that proatherogenic lipoproteins and autophagy activators stimulate lipophagy in human macrophages. Targeting LDs with an LC3 fusion prot

The role of hyaluronic acid in polidocanol foam: An in vitro study.

OBJECTIVE: To study the role of hyaluronic acid (HA) in polidocanol (POL) foam. METHODS: The dose-dependent effect of HA-POL on cultured human umbilical vein endothelial cells (HUVECs) as well as foam stability was evaluated by measuring optical density (OD) values and foam half-life time (FHT), respectively. An in vitro model was utilized for estimating the foam blood-displacement capacity by adopting maximum displacement distance (MDD) and displacement time. A comparison of foam viscosity was also carried out. RESULTS: The OD values of HUVECs treated with HA first increased and then decrea

Common Pathogenesis of Acne Vulgaris and Atherosclerosis.

Foam cells are lipid-loaded macrophages and neutrophils that are generated from a massive uptake of oxidized lipid. Foam cells are a pathological hallmark of atherosclerosis, and have also been found in acne lesions. The same pathological changes determine the common pathogenesis. According to the pathological function of foam cells in these lesions, we put forward a viewpoint on the pathogenesis of acne and atherosclerotic plaques.

Atherogenic-like lesions associated with a progressive chronic vascular degeneration in a rat model of repetitive low-level blast brain injury.

Repetitive low-level blast exposures in rats induce the development of a post-traumatic stress disorder (PTSD)-like phenotype and evolving chronic brain vascular alterations. These alterations included atherogenic-like lesions characterized by increased extravasation of blood elements into the arterial subendothelial layers, nuclear expression of c-Fos in endothelial and vascular smooth muscle cells, vascular hyperplasia, foam cell formation, and ultimately vascular rupture. Foam cells were mainly of macrophage/microglial or of vascular smooth muscle cell origin with upregulated expression of

Secondary lipidosis in leukemia.

Foam cells in the spleen, bone marrow, liver and lymph nodes were examined on the 73 reliably recorded and sampled leukemia autopsy cases encountered at Kobe University from 1958 to 1972. Although the substances stored in the foam cells were biochemically unknown, the foam cells in leukemia could be morphologically classified into two types: The one was identified with the Gaucher type, but the other was not identified with the sea-blue type and might be considered as to be the transitional type described in another report. Foam cells could be found in the spleen of 6 out of 12 cases of chroni

Supercritical CO2-Assisted Fabrication of Environmentally Friendly Poly(Propylene Carbonate)/Poly(Butylene Adipate-co-Terephthalate)/Epoxidized Soybean Oil (PPC/PBAT/ESO) Lightweight Open-Cell Foams With Excellent Degradability and Thermal Insulation Properties.

Driven by the increasing severity of plastic pollution, biodegradable polymers such as poly(propylene carbonate) (PPC), owing to their biodegradability, gas barrier properties, and CO2-fixing capability, have attracted considerable attention as alternatives to conventional plastics. In this work, PPC/poly(butylene adipate-co-terephthalate) (PBAT)/epoxidized soybean oil (ESO) composite foams were successfully fabricated via an environmentally friendly supercritical CO2 foaming process. The results demonstrate that ESO, acting as a reactive compatibilizer, forms chemical linkages between PPC and

Hypertension-induced cerebral atherosclerosis in the cholesterol-fed rabbit.

Foam cell lesions were found in cholesterol-fed rabbits with induced hypertension, particularly in intimal cushions at branching sites, where permeability to horseradish peroxidase was enhanced. Permeability to horseradish peroxidase was enhanced at the edge of intimal cushions without foam cell accumulation. This finding suggests that permeability is increased before foam cell infiltration. No foam cell lesions were observed in the intima of cerebral arteries distant from branching sites, but insudation of plasma constituents here caused endothelial cells to separate from the subendothelial m

Simulation of plaque formation in a realistic geometry of a human aorta: effects of endothelial layer properties, heart rate, and hypertension.

Nowadays, cardiovascular diseases are the most common cause of death worldwide. Besides, atherosclerosis is a cardiovascular disease that occurs with persistent narrowing of arteries, especially medium and large-sized arteries. Atherosclerosis begins with a local elevation in the permeability of the arterial wall as a result of endothelial inflammation. Subsequently, excess LDL permeates into the arterial wall. Then, through several chemical responses and reactions, foam cells are produced. These foam cells serve as a crucial indicator for assessing the development of atherosclerosis within th

Reactive Oxygen Species-Responsive Sequentially Targeted AIE Fluorescent Probe for Precisely Identifying the Atherosclerotic Plaques.

The formation of atherosclerosis is the root cause of various cardiovascular diseases (CVDs). Therefore, effective CVD interventions call for precise identification of the plaques to aid in clinical treatment of such diseases. Herein, a reactive oxygen species (ROS)-responsive sequentially targeted fluorescent probe is developed for atherosclerotic plaque recognition. An aggregation-induced emission active fluorophore is linked to maleimide (polyethylene glycol) hydroxyl with a ROS-responsive cleavable bond, which is further functionalized with CLIKKPF peptide (TPAMCF) for specifically binding

Facile Synthesis of Formaldehyde-Free Bio-Based Thermoset Resins for Fabrication of Highly Efficient Foams.

Bio-based biodegradable foams were formulated from a crosslinkable network structure combining starch, furfuryl alcohol, glyoxal, and condensed tannin in the presence of p-toluenesulfonic acid (pTSA) and azodicarbonamide (AC) as a foaming agent. More importantly, the reinforcement of gelatinized starch-furanic foam using tannin, originating from forestry, resulted in an excellent compressive strength and lower pulverization ratio. Moreover, the addition of tannin guaranteed a low thermal conductivity and moderate flame retardancy. Fourier transform infrared (FTIR) spectroscopy approved the suc

A Myelin Debris Cleaner for Spinal Cord Injury Recovery: Polycaprolactone / Cell Membrane Assembled Scaffolds.

After spinal cord injury (SCI), a mass of myelin debris derived from injured myelin sheath will be consistently generated and induce macrophages to be foam cells. It has been established that myelin debris and foam cells are negative on SCI recovery through direct and indirect neurotoxicity. Different from previous studies, the present research utilized efficient biological composite materials to adsorb myelin debris, exploring new avenues for solving foam cells and myelin debris following SCI. To achieve the strategy, the present author team has developed the biomaterial composed of polycapro

Enhancing the Sound and Thermal Insulation Properties of Polypropylene Foam by Preparing High Melt Strength Polypropylene.

High-performance polypropylene (PP) foam is a vital polymer product in industrial areas. However, the poor melt strength of ordinary PP homopolymer limits its foaming molding. In this work, high melt strength polypropylene (HMSPP) is prepared by using styrene (St) and tripropylene glycol diacrylate (TPGDA) as comonomers, and then PP foams are prepared by mold foaming method. The results show that adding St in the grafting process of TPGDA will obviously improve the melt strength of the PP matrix, and its melt strength (28 184 Pa.s) is 7.4 times higher than that of pure PP. HMSPP foam has more

Anti-atherosclerosis effect of H2S donors based on nicotinic acid and chlorfibrate structures.

Based on the structures of nicotinic acid and chlorfibrate, a series of new H2S donors were synthesized and their anti-atherosclerosis activities using Ox-LDL RAW 264.6 cells as model were evaluated. The release test showed that all the compounds could release H2S effectively and showed low cytotoxicity. In the bioactivity experiments, compounds 1, 3, 9 and 14 increased the survival rate of HUVEC cells treated by ox-LDL; among four compounds, compounds 1 and 3 displayed higher activity than the others. In the foam cell model, compounds 1 and 3 were found to inhibit the formation of foam cells

Effective Halogen- and Phosphorus-Free Polyphenylene Ether Resin-Based Flame-Retardant Foam.

A novel halogen- and phosphorus-free intrinsic flame-retardant foam is fabricated from curable phenol-terminated polyphenylene ether resin with a high molecular weight using phenol, formaldehyde, and diphenyl ether as starting materials. The limiting oxygen index (LOI) of the pure foam is 24.90% ± 0.28. When 0.5 wt % silica sol is added, the LOI of the foam (SPF-0.5) is up to 28.5% ± 0.15 and both the combustion heat release rate and total combustion heat are low. Moreover, the SPF-0.5 foam exhibits high carbon residue, high compressive strength, and low pulverization rate and is superior to s

Regional Segregated Architecture Strategy for High-Efficiency Microwave Absorption of Polyimide Foam.

Polyimide (PI) microwave absorption (MA) foam characterized by its superior electromagnetic protection performance and adjustable mechanical properties, is considered a quintessential structure-function integrated material. Currently, the expressed low compressive strength (∼kPa) of PI MA foam matrix limits application as a direct load-bearing component, despite potential enhancements through chemical modification and process optimization. Herein, leveraging the higher compressive strength of aramid honeycomb, a foam precursor solution with a more controllable foaming process is integrated int

Hypersensitivity pneumonitis secondary to foam exposure in mattress and pillows.

Fibrotic hypersensitivity pneumonitis (HP) has a poor prognosis when no antigen is identified, which occurs in many cases. We present a case of HP due to foam exposure in bedding, an unrecognised cause of HP. A woman was referred for dyspnoea and cough. High-resolution chest computed tomography (HRCT) showed a three-density pattern with gas trapping. Pulmonary function tests (PFTs) revealed restriction and reduced diffusing capacity. Bronchoalveolar lavage showed lymphocytosis (43%) and lung cryobiopsy showed fibrosis, lymphocytic infiltration and multinucleated giant cells. She had foam in ma

Preparation of chain-extended poly(lactic acid)/poly(vinyl acetate) composite nanofoams via supercritical CO2 foaming with DFT approach.

Biodegradable nanocellular poly(lactic acid) (PLA) foams have emerged as a key alternative to traditional petroleum-based foams due to their environmental friendliness, lightweight properties, and thermal insulation capabilities. In this study, PLA was chain-extended using ethylene-acrylate-glycidyl methacrylate terpolymer as a chain extender, and poly(vinyl acetate) (PVAc) was introduced to prepare CEPLA/PVAc blends. Supercritical carbon dioxide (CO2) was used to prepare nanobimodal cellular foams via a solid-state foaming method. In parallel, density functional theory was employed to quantit

Planar network statistics for two-dimensional rupturing foams.

We conduct experiments on a class of two-dimensional semiwet foams generated through compressing a three-dimensional soap foam between two glass plates. To induce a spatially uniform rupturing process on foam boundaries, an additional plate is heated and placed on top of the unheated plates. For 30 separate foam samples, we record network statistics related to cell side numbers and areas as the foam coarsens over a half-minute. We find that the Aboav law and a quadratic Lewis Law, two commonly used relations between network topology and geometry, hold well for preheated foams. To track how wel

Lysosomes, cholesterol and atherosclerosis.

Cholesterol-engorged macrophage foam cells are a critical component of the atherosclerotic lesion. Reducing the sterol deposits in lesions reduces clinical events. Sterol accumulations within lysosomes have proven to be particularly hard to mobilize out of foam cells. Moreover, excess sterol accumulation in lysosomes has untoward effects, including a complete disruption of lysosome function. Recently, we demonstrated that treatment of sterol-engorged macrophages in culture with triglyceride-containing particles can reverse many of the effects of cholesterol on lysosomes and dramatically reduce

High Performance Attapulgite/Polypyrrole Nanocomposite Reinforced Polystyrene (PS) Foam Based on Supercritical CO2 Foaming.

CO2 has been regarded as one of the most promising blowing agents for polystyrene (PS) foam due to its non-flammability, low price, nontoxicity, and eco-friendliness. However, the low solubility and fast diffusivity of CO2 in PS hinder its potential applications. In this study, an attapulgite (ATP)/polypyrrole (PPy) nanocomposite was developed using the in situ polymerization method to generate the hierarchical cell texture for the PS foam based on the supercritical CO2 foaming. The results demonstrated that the nanocomposite could act as an efficient CO2 capturer enabling the random release o
