To verify whether DNA repair is regulated by FOXO3a, a tet-on flag-h-FOXO3a transgenic mice were used. RT-q-PCR and western blot analysis showed that the mRNA and protein levels of flag-h-FOXO3a, XRCC4, XPC, APE1 and MSH2 increased dose dependently by doxycycline. DNA repair activities like non-homologous end joining (NHEJ), nucleotide excision repair (NER), base excision repair (BER), mismatch repair (MMR) also increased in a doxycycline dose dependent manner. MEF (mouse embryonic fibroblast) cells of the transgenic mouse were transfected with human XRCC4/XPC/APE1/MSH2 promoter-pGL3 basic vec

The high prevalence of malocclusion and dentofacial malformations means that the demand for orthodontic treatments has been increasing rapidly. As the biological basis of orthodontic treatment, the mechanism of mechanical force-induced alveolar bone remodeling during orthodontic tooth movement (OTM) has become the key scientific issue of orthodontics. It has been demonstrated that bone mesenchymal stem cells (BMSCs) are crucial for bone remodeling and exhibit mechanical sensing properties. Mechanical force can promote osteoblastic differentiation of BMSCs and osteogenesis, but the key factor t

Imidacloprid (IMI), a widely used neonicotinoid insecticide, has been frequently detected in drinking water and food; however, whether long-term low-dose exposure to environmentally relevant levels of IMI induces neurotoxic effects in the central nervous system remains unclear. In this study, C57BL/6 N mice were exposed to IMI in drinking water for three, six months at doses derived from human exposure estimates (0, 3 μg/kg, 103 μg/kg, and 6.55 mg/kg). Behavioral tests, including the Morris water maze and novel object recognition, revealed no overt cognitive impairment following IMI exposure.

BACKGROUND: Bone marrow mesenchymal stem cells (BMSCs) exert potent paracrine effects that can reshape the immune microenvironment. Nebulized inhalation enables non-invasive, targeted delivery to the lung, a pivotal immune interface capable of modulating systemic immunity. This study introduces a novel therapeutic strategy using nebulized BMSC supernatant to activate the FOXO3 signaling pathway in pulmonary dendritic cells (DCs), reprogramming them toward a tolerogenic phenotype. This approach suppresses autoreactive T cell infiltration and alleviates central nervous system (CNS) inflammation

Interleukin 13 (IL13), as a cytokine belonging to the chemokine family, exhibits multiple complex physiological effects. Previous studies have consistently observed its significant role in various types of neural injuries and neurodegenerative diseases, including traumatic brain injury (TBI), through immune modulation and influencing neuroinflammation. However, little is known about its other physiological effects following TBI. In this study, we discovered a previously unreported important effect of IL13. In mouse models of TBI, we found that IL13 can protect against the loss of pericytes in

Chronic pelvic pain (CPP) of gynaecological origin encompasses a heterogeneous group of disorders, among which pelvic venous disorders (PeVD) and adenomyosis are recognised contributors. Concomitant PeVD and adenomyosis may potentiate symptom burden and complicate management strategies. A 45-year-old parous woman was referred with a protracted history of CPP manifesting as lower abdominal, lumbar and hip pain, exacerbated by prolonged orthostasis and coital activity. Menstrual symptoms comprised dysmenorrhoea and menorrhagia complicated by iron-deficiency anaemia. Prior conservative measures w

Signals downstream of Akt can either favor or oppose stem cell (SC) maintenance, but how this dual role can be achieved is still undefined. Using human limbal keratinocyte stem cells (LKSCs), a SC type used in transplantation therapies for corneal regeneration, we show that Akt signaling is prominent in SC populations both in vivo and in vitro, and that Akt1 promotes while Akt2 opposes SC self-renewal. Noteworthy, loss of Akt2 signaling enhances LKSC maintenance ex vivo, whereas Akt1 depletion anticipates SC exhaustion. Mechanistically, the antagonistic functions of Akt1 and Akt2 in SC control

Intervertebral disc degeneration (IVDD) is the leading cause of low back pain and associated disability worldwide. The cartilage endplate (CEP) is a critical structure in maintaining the homeostasis of the intervertebral disc, by exosomes (Exos)-mediated intracellular communication between cartilage endplate stem cells (CESCs) and nucleus pulposus cells (NPCs). However, whether the senescence of CESCs influences the functionality of CESCs-derived Exos (CESCs-Exos) and participates in the progress of IVDD remains unclear. In this study, we explored the role and mechanism of the Exos-based intra

Copper (Cu) and zinc (Zn) are essential trace elements for terrestrial animals. Studies suggested that single dietary Cu- or Zn- deficiency lead to liver lipid deposition and cause metabolic dysfunction-associated steatotic liver disease (MASLD). However, the mechanisms by which dietary Cu and Zn co-deficiency promote hepatic lipid deposition and metabolism remain unknown. Our study found that, compared to single Cu- or Zn- deficiency, Cu and Zn co-deficiency significantly increased lipid deposition, induced mitochondrial oxidative stress, disrupted mitochondrial structure and function, and in

OBJECTIVES: Perinatal hypoxia causes premature activation and initiation of growth in dormant follicles, leading to diminished ovarian reserve. An indirect mechanism such as the release of stress-related hormones may influence ovarian follicle recruitment under hypoxic conditions. We wanted to determine whether hypoxic ovarian damage results from increased follicle growth and "burnout" or from increased apoptosis and whether this damage is age-dependent.
DESIGN: Animal study was conducted.
PARTICIPANTS/MATERIALS, SETTING, METHODS: Using adult 6-week-old (n = 8) and one-day-old newborn (n = 2

Hypoxic injury of cardiac microvascular endothelial cells (CMECs) is an important pathophysiological event in myocardial infarction, whereas, the underlying mechanism is still poorly understood. Autophagy, a highly conserved process of cellular degradation, is required for normal cardiac function and also has been implicated in various cardiovascular diseases. Here we investigated the potential role of autophagy in CMEC dysfunction under hypoxia. CMECs were isolated from SD rats. Hypoxia (6-24h, 1% O2) induced autophagy in CMECs as evidenced by formation of punctate LC3, increased conversion o

Atherosclerosis (AS) is a chronic progressive disease caused by various factors and causes various cerebrovascular and cardiovascular diseases (CVDs). Reducing the plasma levels of low-density lipoprotein cholesterol is the primary goal in preventing and treating AS. Proprotein convertase subtilisin/kexin type 9 (PCSK9) plays a crucial role in regulating low-density lipoprotein cholesterol metabolism. Panax notoginseng has potent lipid-reducing effects and protects against CVDs, and its saponins induce vascular dilatation, inhibit thrombus formation, and are used in treating CVDs. However, the

Enterovirus 71 (EV71), the major cause of hand-foot-and-mouth disease (HFMD), has evolved diverse strategies to counter the type I interferon (IFN-I) response during infection. Recently, microRNAs have regulatory roles in host innate immune responses to viral infections; however, whether EV71 escapes the IFN-I antiviral response through regulation of miRNAs remains unclear. Using a microarray assay, microRNA-155-5p (miR-155-5p) was found to be significantly up-regulated in serum from patients with EV71 infection and the increased expression of miR-155-5p was further confirmed in vivo and in vi

Non-alcoholic fatty liver disease (NAFLD) is a globally prevalent chronic liver condition, primarily characterized by excessive accumulation of fat within the liver. A pivotal factor in the progression of NAFLD is cholesterol deposition, which significantly exacerbates liver cell damage through the induction of endoplasmic reticulum (ER) stress. At the heart of this process is sterol regulatory element-binding protein 2 (SREBP2), a crucial transcription factor in cholesterol synthesis. The expression levels of SREBP2 are closely associated with the severity of NAFLD, marking it as a potential

The presence of Helicobacter pylori (H. pylori) infection poses a substantial risk for the development of gastric adenocarcinoma. The primary mechanism through which H. pylori exerts its bacterial virulence is the cytotoxin CagA. This cytotoxin has the potential to induce inter-epithelial mesenchymal transition, proliferation, metastasis, and the acquisition of stem cell-like properties in gastric cancer (GC) cells infected with CagA-positive H. pylori. Cancer stem cells (CSCs) represent a distinct population of cells capable of self-renewal and generating heterogeneous tumor cells. Despite ev
