Optimized psilocybin production in tryptophan catabolism-repressed fungi.

The high therapeutic potential of psilocybin, a prodrug of the psychotropic psilocin, holds great promise for the treatment of mental disorders such as therapy-refractory depression, alcohol use disorder and anorexia nervosa. Psilocybin has been designated a 'Breakthrough Therapy' by the US Food and Drug Administration, and therefore a sustainable production process must be established to meet future market demands. Here, we present the development of an in vivo psilocybin production chassis based on repression of l-tryptophan catabolism. We demonstrate the proof of principle in Saccharomyces

Mucorales fungi suppress nitric oxide production by macrophages.

In October 2022, Mucorales fungi were listed in the "High Priority Group" on the first-ever list of fungal priority pathogens by the World Health Organization. As the causative agent of mucormycosis, Mucorales have become of great clinical and public health importance with growing mucormycosis numbers, notably with the exponential rise of COVID-19-associated mucormycosis cases. Despite the dire need, there are limited therapeutic options to treat mucormycosis. Our research fills in critical gaps of knowledge about how Mucorales fungi evade the host immune system. Specifically, we offer evidenc…

Versatile effectors of phytopathogenic fungi target host immunity.

Phytopathogenic fungi secrete a large arsenal of effector molecules, including proteinaceous effectors, small RNAs, phytohormones and derivatives thereof. The pathogenicity of fungal pathogens is primarily determined by these effectors that are secreted into host cells to undermine innate immunity, as well as to facilitate the acquisition of nutrients for their in planta growth and proliferation. After conventional and non-conventional secretion, fungal effectors are translocated into different subcellular compartments of the host cells to interfere with various biological processes. In extrac…

PTP1B Inhibitors from the Entomogenous Fungi Isaria fumosorosea.

Protein tyrosine phosphatase 1B (PTP1B) is implicated as a negative regulator of insulin receptor (IR) signaling and a potential drug target for the treatment of type II diabetes and other associated metabolic syndromes. Thus, small molecule inhibitors of PTP1B can be considered as an attractive approach for the design of new therapeutic agents of type II diabetes and cancer diseases. In a continuing search for new PTP1B inhibitors, a new tetramic acid possessing a rare pyrrolidinedione skeleton named fumosorinone A (1), together with five known ones 2-6 were isolated from the entomogenous fun

Tipranavir exhibits different effects on opportunistic pathogenic fungi.

OBJECTIVES: Tipranavir (TPV) is a non-peptidic protease inhibitor (PI) that represents one of the latest options approved in the salvage setting for HIV-infected multi-drug resistant patients. In this study, we explored whether TPV affects virulence of opportunistic fungi such as Cryptococcus neoformans and Candida albicans. METHODS: C. neoformans and C. albicans were cocultured in the presence or absence of TPV for various time periods. Subsequently, growth inhibition, phospholipases, proteases and capsule size were examined. In selected in vivo experiments, TPV was administered in immunocom…

Evolution of Dermatophytic Fungi: Anthropogenic and Biogenic Environmental Factors.

Climate change presents new challenges to dermatologic care, ranging from anthropogenic factors, such as pollution that select for extremotolerant and potentially antifungal-resistant fungi, to biogenic factors that can drive zoonotic outbreaks, which have received increased attention since the SARS-CoV-2 pandemic. Through this lens, we examine the evolution of dermatophytic fungi.

Checklist of Lichens and Lichenicolous Fungi from Mainland Portugal.

Mainland Portugal has a lichenological tradition spanning more than three centuries, yet it still lacks a modern, critically revised checklist of its lichen biota. This gap is particularly relevant given the increasing role of species checklists as authoritative references for DNA metabarcoding applications and the expansion of large biodiversity datasets, including those derived from citizen science initiatives. Here we present the first comprehensive checklist of lichens and lichenicolous fungi recorded from Mainland Portugal, based on the critical assessment of 43,718 bibliographic records …

Antibacterial Diphenyl Ethers from Fungi Aspergillus versicolor YC-27.

Microbial metabolites serve as a vital source for drug discovery, with diphenyl ether derivatives from Aspergillus sp. garnering significant attention due to their antimicrobial activities. This work focused on the antibacterial compounds from the fungi Aspergillus versicolor YC-27, which led to the isolation of one new diphenyl ether derivative, aspesicol (1), together with eight known compounds (2-9). The structure of the new compound was elucidated by spectroscopic methods, high-resolution electrospray ionization mass spectrometry, and nuclear magnetic resonance calculations with DP4+ metho…

Antifouling Lipids from Marine Fungi of the Beibu Gulf.

Background: The search for environmentally friendly antifouling agents has led to an increased focus on marine natural products. Methods: This study investigated the antifouling potential of lipid fractions extracted from ten marine fungal strains isolated from the Beibu Gulf, China. The lipids were evaluated through a multi-level bioassay approach, including the inhibition of microfouling (against four fouling bacteria: Marinobacterium jannaschii, Vibrio pelagius, Vibrio rotiferianus, and Alteromonas macleodii), the prevention of macrofouling (inhibition of barnacle Amphibalanus reticulatus c

Tackling multi-drug resistant fungi by efflux pump inhibitors.

The emergence of multidrug-resistant fungi is of grave concern, and its infections are responsible for significant deaths among immunocompromised patients. The treatment of fungal infections primarily relies on a clinical class of antibiotics, including azoles, polyenes, echinocandins, polyketides, and a nucleotide analogue. However, the incidence of fungal infections is increasing as the treatment for human and plant fungal infections overlaps with antifungal drugs. The need for new antifungal agents acting on different targets than known targets is undeniable. Also, the pace at which loss of…

Highly Reactive Group I Introns Ubiquitous in Pathogenic Fungi.

Systemic fungal infections are a growing public health threat, and yet viable antifungal drug targets are limited as fungi share a similar proteome with humans. However, features of RNA metabolism and the noncoding transcriptomes in fungi are distinctive. For example, fungi harbor highly structured RNA elements that humans lack, such as self-splicing introns within key housekeeping genes in the mitochondria. However, the location and function of these mitochondrial riboregulatory elements has largely eluded characterization. Here we used an RNA-structure-based bioinformatics pipeline to identi…

Curcumin-based molecular probes for fluorescence imaging of fungi.

Fluorescence imaging is a powerful and widely used method to visualize and study living organisms. However, fungi are notoriously difficult to visualize using fluorescence microscopy, given that their cell wall represents a diffusion barrier, and the synthetic organic dyes available are very limited when compared to molecular probes available for other organisms. Moreover, these dyes are usually available in only one colour, preventing co-staining experiments. To fill this gap, curcumin-based molecular probes were designed based on the rationale that curcumin is fluorescent and has moderate to…

Biocontrol of almond canker diseases caused by Botryosphaeriaceae fungi.

BACKGROUND: Botryosphaeria dieback is a canker disease caused by fungal species of the Botryosphaeriaceae family that threatens almond productivity. The most common control measure to prevent canker development is the application of fungicides which are being phased out by European Union regulations. In the present study, two sets of bacterial strains were evaluated for their antifungal activity against pathogenic Botryosphaeriaceae species through in vitro and in vivo antagonism assays. RESULTS: The rhizospheric bacteria Pseudomonas aeruginosa AC17 and Bacillus velezensis ACH16, as well as t…
