Gases in Sepsis: Novel Mediators and Therapeutic Targets.

Sepsis, a potentially lethal condition resulting from failure to control the initial infection, is associated with a dysregulated host defense response to pathogens and their toxins. Sepsis remains a leading cause of morbidity, mortality and disability worldwide. The pathophysiology of sepsis is very complicated and is not yet fully understood. Worse still, the development of effective therapeutic agents is still an unmet need and a great challenge. Gases, including nitric oxide (NO), carbon monoxide (CO) and hydrogen sulfide (H2S), are small-molecule biological mediators that are endogenously

Malproduction of endogenous hydrogen gas in COVID-19.

The molecular footprints of COVID-19 occur everywhere, even reaching the family of biologically active gases and gasotransmitters. Besides nitric oxide and hydrogen sulfide, COVID-19 might also alter the homeostasis of dihydrogen (H2), another gaseous bioactive molecule produced endogenously by the human gut bacteria. Many studies have shown various alterations of the gut microbiota in patients with coronavirus disease 2019, including the lower abundance of hydrogen-producing bacteria that could instigate the shortage of hydrogen output. Since dihydrogen has many important bioactivities, inclu…

Hydrogen sulfide as a neuromodulator of the vascular tone.

Hydrogen sulfide (H2S) is a unique signaling molecule that, along with carbon monoxide and nitric oxide, belongs to the gasotransmitters family. H2S is endogenously synthesized by enzymatic and non-enzymatic pathways. Three enzymatic pathways involving cystathionine-γ-lyase, cystathionine-β-synthetase, and 3-mercaptopyruvate sulfurtransferase are known as endogenous sources of H2S. This gaseous molecule has recently emerged as a regulator of many systems and physiological functions, including the cardiovascular system where it controls the vascular tone of small arteries. In this context, H2S …

Hydrogen sulfide in the regulation of insulin secretion and insulin sensitivity: Implications for the pathogenesis and treatment of diabetes mellitus.

Insulin secretion and sensitivity play an essential role in maintaining normal glucose level and their abnormalities result in diabetes mellitus. H2S-synthesizing enzymes, CBS and/or CSE, are expressed in insulin-secreting pancreatic β cells and H2S inhibits insulin secretion by activating ATP-sensitive K+ channels. In addition, H2S has been reported to have either pro- or antiapoptotic effects on β cells. Studies in the animal models suggest that excess of H2S in pancreatic islets may contribute to both type 1 and type 2 diabetes. H2S has also been demonstrated to regulate insulin sensitivity

A novel treatment for "morning sickness": Nausea of pregnancy could be induced by excess sulfite which molybdenum can help alleviate.

Nausea and vomiting of pregnancy (NVP) remains difficult to treat. Last century, thalidomide was used to alleviate NVP, but it caused teratogenesis by interfering with angiogenesis. The gasotransmitters hydrogen sulfide (H2S) and nitric oxide are mutually dependent on each other for their angiogenesis-related functions. Pregnancy-related requirements for increased endogenous H2S could create a temporary excess of sulfite, an H2S catabolite, which is toxic and can induce nausea. Sulfite oxidase, a molybdenum-containing enzyme, catalyzes oxidation of sulfite to sulfate, which can then be excrete…

Light-triggered hydrogen cyanide (HCN) release from self-immolative cyanohydrin donors.

Hydrogen cyanide (HCN) is an emerging gasotransmitter and protic small molecule bioregulator. We report HCN release from a series of light-triggered, self-immolative cyanohydrin donors built on the 4,5-dimethoxy-o-nitrobenzyl (DMNB) platform. We demonstrate carbonate- and ether-ligated strategies for increasing release efficiency, expanding the chemical space for HCN release.

The Role of Nitric Oxide and Hydrogen Sulfide in Urinary Tract Function.

This MiniReview focuses on the role played by nitric oxide (NO) and hydrogen sulfide (H2 S) in physiology of the upper and lower urinary tract. NO and H2 S, together with carbon monoxide, belong to the group of gaseous autocrine/paracrine messengers or gasotransmitters, which are employed for intra- and intercellular communication in almost all organ systems. Because they are lipid-soluble gases, gaseous transmitters are not constrained by cellular membranes, so that their storage in vesicles for later release is not possible. Gasotransmitter signals are terminated by falling concentrations up…

Hydrogen sulfide in the brain as a silent neuroprotector in Alzheimer's disease.

Hydrogen sulfide (H2S) is an endogenously produced gasotransmitter that has garnered growing attention for its critical roles in cellular signalling and brain function. It regulates NMDA receptors during long-term potentiation, a fundamental mechanism underlying memory consolidation and influences neurotransmission and essential neurophysiological functions. H2S is synthesized by three enzymes: cystathionine γ-lyase (CSE) and cystathionine β-synthase (CBS) and 3-mercaptopyruvate sulfurtransferase (MST) within the cell. CBS is suggested to be the primary source of H2S in the brain parenchyma, w

Systematic analysis of the global characteristics and reciprocal effects of S-nitrosylation and S-persulfidation in the human proteome.

Gasotransmitter-mediated cysteine post-translational modifications, including S-nitrosylation (SNO) and S-persulfidation (SSH), play crucial roles and interact in various biological processes. However, there has been a delay in appreciating the interactional rules between SNO and SSH. Here, all human S-nitrosylated and S-persulfidated proteomic data were curated, and comprehensive analyses from multiple perspectives, including sequence, structure, function, and exact protein impacts (e.g., up-/down-regulation), were performed. Although these two modifications collectively regulated a wide arra

Hydrogen sulfide in health and diseases: cross talk with noncoding RNAs.

Hydrogen sulfide (H2S) is previously described as a potentially lethal toxic gas. However, this gasotransmitter is also endogenously generated by the actions of cystathionine-β-synthase (CBS), cystathionine-γ-lyase (CSE), and 3-mercaptopyruvate sulfurtransferase (3-MST) in mammalian systems, thus belonging to the family of gasotransmitters after nitric oxide (NO) and carbon monoxide (CO). The physiological or pathological significance of H2S has been extensively expanded for decades. Growing evidence has revealed that H2S exerts cytoprotective functions in the cardiovascular, nervous, and gast…

Protein sulfhydration.

Hydrogen sulfide (H2S) is one of the gasotransmitters that modulates various biological processes and participates in multiple signaling pathways. H2S signals by a process termed sulfhydration. Sulfhydration has recently been recognized as a posttranslational modification similar to nitrosylation. Sulfhydration occurs at reactive cysteine residues in proteins and results in the conversion of an -SH group of cysteine to an -SSH or a persulfide group. Sulfhydration is highly prevalent in vivo, and aberrant sulfhydration patterns have been observed under several pathological conditions ranging fr…

A hydrogen sulfide-releasing alginate dressing for effective wound healing.

For wounds with heavy exudate levels, a dressing that can help to absorb wound exudate and improve the wound healing process is highly desired. Hydrogen sulfide (H2S) has been recognized as an important gasotransmitter that can improve angiogenesis which is crucial for wound healing. In this study, a functional sodium alginate (SA) dressing with H2S-releasing property (SA/JK-1) was fabricated by incorporating JK-1 molecule, a pH-dependent H2S donor, into SA sponge. The resultant SA/JK-1 sponge provided a moist and protective healing environment and was capable of releasing H2S consistently und

Dynamics and metabolic effects of intestinal gases in healthy humans.

Many living beings use exogenous and/or endogenous gases to attain evolutionary benefits. We make a comprehensive assessment of one of the m … extracellular, paracrine and distant actions. Further, we highlight the local and systemic roles of gasotransmitters, i.e., signalling gaseous molecules that can freely diffuse through the intestinal cellular membranes. Yet, we provide testable hypotheses concerning the still unknown effects of some intestinal gases on the myenteric and submucosal neurons.

Hydrogen sulfide decreases Cd translocation from root to shoot through increasing Cd accumulation in cell wall and decreasing Cd2+ influx in Isatis indigotica.

Hydrogen sulfide (H2S), a small gaseous signalling molecule, plays a pivotal role in the plant response to heavy metal stress. Here, we revealed a novel mechanism of Isatis indigotica resistance to cadmium (Cd) stress, in which H2S promotes Cd accumulation in the root and decreases the long-distance transport of Cd from the root to shoot. Cd significantly inhibited Isatis indigotica growth and induced the endogenous H2S level. Application of NaHS (a H2S donor) alleviated the effects of Cd. NaHS restriction of the translocation factor of Cd, elevated the Cd content in roots and depressed the Cd
