P. aeruginosa (PA) continues to pose a threat to global public health due to its high levels of antimicrobial resistance (AMR). The ongoing AMR crisis has led to an alarming shortage of effective treatments for resistant microbes, and hence there is a pressing demand for the development of novel antimicrobial interventions. The potential use of antivirulence therapeutics to tackle bacterial infections has attracted considerable attention over the past decades as they hamper the pathogenicity of target microbes with reduced selective pressure, minimizing the emergence of resistance. One such ap

Upon biofilm formation, production of extracellular matrix components and alteration in physiology and metabolism allows bacteria to build up multicellular communities which can facilitate nutrient acquisition during unfavorable conditions and provide protection toward various forms of environmental stresses to individual cells. Thus, bacterial cells within biofilms become tolerant against antimicrobials and the immune system. In the present study, we evaluated the antibiofilm activity of the macrolides clarithromycin and azithromycin. Clarithromycin showed antibiofilm activity against rdar (r

The potato common scab pathogen Streptomyces scabies produces N-coronafacoyl-l-isoleucine (CFA-Ile), which is a member of the coronafacoyl family of phytotoxins that are synthesized by multiple plant pathogenic bacteria. The CFA-Ile biosynthetic gene cluster contains a regulatory gene, cfaR, which directly controls the expression of the phytotoxin structural genes. In addition, a gene designated orf1 encodes a predicted ThiF family protein and is cotranscribed with cfaR, suggesting that it also plays a role in the regulation of CFA-Ile production. In this study, we demonstrated that CfaR is an

UNLABELLED: Methylobacterium strains can use one-carbon compounds, such as methanol, for methylotrophic growth. In addition to methanol, a few strains also utilize dichloromethane, a major industrial chlorinated solvent pollutant. With a fully assembled and annotated genome, M. extorquens DM4 is the reference bacterium for aerobic dichloromethane degradation. The doublet N-terminal oriented proteomics (dN-TOP) strategy was applied to further improve its genome annotation and a differential proteomics approach was performed to compare M. extorquens DM4 grown either with methanol or dichlorometh

Leptospira interrogans is the agent of leptospirosis, a reemerging zoonotic disease. It is transmitted to humans through environmental surface waters contaminated by the urine of mammals chronically infected by pathogenic strains able to survive in water for long periods. Little is known about the regulatory pathways underlying environmental sensing and host adaptation of L. interrogans during its enzootic cycle. This study identifies the EbpA-RpoN regulatory pathway in L. interrogans In this pathway, EbpA, a σ54 activator and putative prokaryotic enhancer-binding protein (EBP), and the altern

Actinobacillus suis is an opportunistic pathogen that resides in the tonsils of the soft palate of swine. Unknown stimuli can cause this organism to invade the host, resulting in septicaemia and sequelae including death. To better understand its pathogenesis, the expression of several adhesin genes was evaluated by semi-quantitative real-time PCR in A. suis grown in conditions that mimic the host environment, including different nutrient and oxygen levels, exponential and stationary phases of growth, and in the presence of the stress hormone epinephrine. Fifty micromolar epinephrine did not af

Brucellosis, the most common widespread zoonotic disease, is caused by Brucella spp., which are facultative, intracellular, Gram-negative bacteria. With the development of molecular biology techniques, more and more virulence-associated factors have been identified in Brucella spp. A suitable plasmid system is an important tool to study virulence genes in Brucella. In this study, we constructed three constitutive replication plasmids (pTM1-Cm, pTM2-Amp, and pTM3-Km) using the replication origin (rep) region derived from the pBBR1-MCS vector. Also, a DNA fragment containing multiple cloning sit

Micrococcus luteus (NCTC2665, "Fleming strain") has one of the smallest genomes of free-living actinobacteria sequenced to date, comprising a single circular chromosome of 2,501,097 bp (G+C content, 73%) predicted to encode 2,403 proteins. The genome shows extensive synteny with that of the closely related organism, Kocuria rhizophila, from which it was taxonomically separated relatively recently. Despite its small size, the genome harbors 73 insertion sequence (IS) elements, almost all of which are closely related to elements found in other actinobacteria. An IS element is inserted into the r

A large number of bacteria are able to degrade aromatic carbon sources employing different strategies. All these pathways are objects of regulatory control at the level of gene expression. This includes specific control in response to the availability of the respective substrate and in many cases global control responding to other available carbon sources or to the metabolic status of the cell. Here, the regulatory proteins responsible for gene regulation are reviewed in particular in correlation to other proteins with a similar primary structure. Most common is the appearance of regulators of

Certain hydrogenases are capable of converting the trace amounts of molecular hydrogen constantly present in Earth's atmosphere to supply cells with a little energy during periods of starvation. Recently, Kropp and colleagues investigated the regulation of one of these "high-affinity" hydrogenases in response to different growth conditions for the ubiquitous soil bacterium Mycobacterium smegmatis (A. Kropp, J. D. Archer, M. Jespersen, T. D. Watts, et al., mSystems 11:e01678-25, 2026, https://doi.org/10.1128/msystems.01678-25). A mutation in the gylR gene, whose product acts as a positive trans

UNLABELLED: Methanotrophs primarily use methane as a carbon and energy source, but they can also grow on methanol. However, methanol assimilation in the absence of methane oxidation results in a redox imbalance due to the accumulation of NADH. In Methylomonas sp. DH-1, a type I methanotroph, we show that this methanol-induced redox stress activates the RegB/RegA two-component system, a global redox sensor. Methanol triggers the RegB-to-RegA phosphotransfer cascade, resulting in RegA-dependent expression of the feo operon and increased ferrous iron uptake. This iron acquisition is essential for

Thermoregulatory systems are rarely utilized in the synthetic biology of Streptomyces. In Streptomyces hygroscopicus TL01, the previous finding of enhanced validamycin biosynthesis at 37°C indicated the involvement of a thermoregulatory system. In this study, the PvalA (PKA) promoter was thermoregulated in Streptomyces albus J1074, and its regulation was determined by a GC-rich DNA clamp. PKA-binding regulatory proteins were identified through DNA affinity chromatography. Overexpression of SHJG_5852 and SHJG_8253 in S. albus resulted in the PKA strength ratios of 2.61 and 4.34 at 37°C and 30°C

Lactate is a key metabolite that is used as a carbon and energy source. It can also be generated as a metabolic end product, through reduction of pyruvate. Bacterial enzymes involved in lactate generation are classified as NAD+-dependent lactate dehydrogenases and are generally involved in production of lactate during fermentation, while NAD+-independent lactate dehydrogenases are involved in oxidation of lactate that is linked to reduction of quinone in respiratory or photosynthetic electron transport pathways, or in anaerobic lactate oxidation linked to electron bifurcation during heterotrop

UNLABELLED: The quorum-sensing (QS) system in Methylobacterium oryzae CBMB20, an endophytic bacterium associated with rice (Oryza sativa L.), plays a critical role in regulating bacterial behaviors essential for plant growth promotion and adaptation. This study aimed to elucidate the functional role of the MorI/MorR QS system in M. oryzae CBMB20 and its potential application as a bioinoculant. We identified and characterized two QS signals, 3-OH-C12-HSL and 3-oxo-C12-HSL, synthesized by the MorI enzyme. The MorR receptor was found to preferentially respond to 3-OH-C12-HSL, indicating a high de

Syringic acid (SA) is a key intermediate in the bacterial catabolism of syringyl lignin-derived aromatic compounds. However, bacterial SA catabolism remains largely unknown. Here, we investigated the SA catabolic system in Pseudomonas sp. NGC7, which catabolizes various lignin-derived aromatic monomers. Pathway analysis and gene disruption studies revealed that SA undergoes O demethylation by VanA1B1 to 3-O-methylgallic acid, which is subsequently catabolized through a linear pathway, involving MgaAB, MgaC, MgaD, GalD, GalB, and GalC into the TCA cycle, unlike the branching pathway found in sp

The mammalian cell entry (Mce) proteins are critical for the entry and persistence of Mycobacterium tuberculosis (Mtb) within alveolar macrophages. These proteins assemble into an ATP-binding cassette transporter comprising six substrate-binding proteins (MceA-F), two transmembrane domains (YrbEAB), and two nucleotide-binding domains (MceG2). Although Mtb harbours four distinct mce operons, the regulatory mechanisms governing differential operon expression remain insufficiently understood. In this study, we analyzed the genetic organization, regulatory features, and functional roles of operons

UNLABELLED: Mycobacterium tuberculosis is well adapted to survive and persist in the infected host, escaping the host's immune response. Since polyamines such as spermine, which are synthesized by infected macrophages, are able to inhibit the growth of M. tuberculosis, the pathogen needs strategies to cope with these toxic metabolites. The actinomycete Streptomyces coelicolor, a close relative of M. tuberculosis, makes use of a gamma-glutamylation pathway to functionally neutralize spermine. We therefore considered whether a similar pathway would be functional in M. tuberculosis. In the curren

Bacteria use two-component regulatory systems (TCSs) to adapt to changes in their environment by changing their gene expression. In this study, we show that the EnvZ/OmpR TCS of the clinically relevant opportunistic pathogen Klebsiella pneumoniae plays an important role in successfully establishing lung infection and virulence. In addition, we elucidate the K. pneumoniae OmpR regulon within the host. This work suggests that K. pneumoniae OmpR might be a promising target for innovative anti-infectives.

Adaptation to various stresses during infection is important for Salmonella Typhimurium virulence, while the fitness determinants under infection-relevant stress conditions remain unknown. Here, we simulated conditions Salmonella encountered within the host or in the environment by 15 individual stresses as well as two model cell lines (epithelium and macrophage) to decipher the genes and pathways required for fitness. By high-resolution Tn-seq analysis, a total of 1242 genes were identified as essential for fitness under at least one stress condition. The comparative analysis of fitness deter

Cholera is a severe small intestine bacterial disease caused by consumption of water and food contaminated with Vibrio cholera. The disease causes watery diarrhea leading to severe dehydration and even death if left untreated. In the past few decades, V. cholerae has emerged as multidrug-resistant enteric pathogen due to its rapid ability to adapt in detrimental environmental conditions. This research study aimed to design inhibitors of a master virulence gene expression regulator, HapR. HapR is critical in regulating the expression of several set of V. cholera virulence genes, quorum-sensing
