Presentation of Localized Aggressive Periodontitis in Monozygotic Twins: Case Report.

INTRODUCTION: Localized aggressive periodontal disease is an uncommon finding observed in adolescents and children. Studies have been conducted to elucidate the etiology, contributing factors, and genetic roles in the phenomena. In particular, twin studies have shown genetic makeup to be a substantial risk factor for periodontitis. This case is, to the best of the authors' knowledge, the first to be presented in literature of a same dwelling, set of monozygotic twins, displaying similar presentation of localized aggressive periodontitis, however with different microbial profiles. CASE PRESENT

Genotype-Specific Interactions Between Wheat and Rhizosphere Microbiome Under Drought Stress.

Drought severely limits global wheat production, and plants can mitigate this stress by recruiting beneficial rhizosphere microbiomes. However, the role of host genotype in shaping this recruitment and influencing microbial inoculant efficacy is poorly understood. Here, we aimed to elucidate the interplay among host genotype, drought stress, and inoculation with Trichoderma citrinoviride. A factorial pot experiment investigated the responses of two wheat genotypes (JN 14-95, constitutively drought-tolerant; JN 72, drought-responsive) to water regimes and inoculation with T. citrinoviride. Modulating the Gut Microbiome in Type 2 Diabetes: Nutritional and Therapeutic Strategies.

Type 2 diabetes mellitus (T2DM) is a complex metabolic disorder influenced not only by genetics, diet, and lifestyle but also by the gut microbiota. Dysbiosis (imbalances in microbial composition) can disrupt gut barrier integrity, alter microbial metabolites, and trigger low-grade inflammation, contributing to insulin resistance and β-cell dysfunction. Nutritional interventions, such as probiotics, prebiotics, synbiotics, postbiotics, and bioactive food components, represent potential therapeutic approaches for ameliorating gut eubiosis and advancing glycemic regulation. This narrative review

Comparative Study on Jejunal Immunity and Microbial Composition of Growing-Period Tibetan Pigs and Duroc × (Landrace × Yorkshire) Pigs.

The gut microbiota plays vital roles in metabolizing nutrient, maintaining the intestinal epithelial barrier but also in modulating immunity. Host genetics and the pig breed are implicated in shaping gut microbiota. Tibetan pig is a unique native Chinese breed and has evolved to manifest a strong disease resistance. However, the immunity and microbiota of growing Tibetan (TP) pigs were still rarely understood. The jejunal immunity phenotype and microbial composition of TP and Duroc × (Landrace × Yorkshire) (DLY) pigs were explored through immunohistochemistry and 16S rRNA sequencing. Higher sc

Advances in Microbiome Detection Technologies and Application in Antirheumatic Drug Design.

Rheumatic diseases are a kind of chronic inflammatory and autoimmune disease affecting the connection or supporting structures of the human body, such as the most common diseases Ankylosing spondylitis (AS), gout and Systemic lupus erythematosus (SLE). Although the precise etiology and pathogenesis of the different types of rheumatic diseases remain mostly unknown, it is now commonly believed that these diseases are attributed to some complex interactions between genetics and environmental factors, especially the gut microbiome. Altered microbiome showed clinical improvement in disease symptom

Prehistoric global migration of vanishing gut microbes with humans.

The gut microbiome is crucial for health and is affected strongly by lifestyle1. Many microorganisms commonly found in non-industrialized populations are disappearing or have become extinct in industrialized populations2-6. Studying which microorganisms have been long-term residents of the human gut and may have co-evolved with humans2,7,8 could provide insights into how microbial biodiversity loss affects human health. However, the genetic complexities of microbial evolution and the plasticity of gut microbiome composition have made it challenging to resolve the evolutionary history of these

Genomic Map of Escherichia coli and Single Nucleotide Polymorphism Markers in Colorectal Cancer.

Gut microbial single nucleotide polymorphisms (SNPs) offer stable, specific genetic markers for disease diagnosis. Escherichia coli (E. coli), a dominant gut bacterium, is associated with colorectal cancer (CRC), but limited enteric reference genomes hinder SNP annotation in intestinal strains. Metagenomic sequencing profiled gut microbiota in 200 CRC patients and 200 healthy controls. The E. coli strain WDP was fully sequenced via PacBio single-molecule technology for genome assembly and functional annotation. Wilcoxon tests identified differentially abundant microbes, while Lasso regression

Hadal topography incubates hidden microbial hotspots in the deepest ocean.

Plate subduction creates unique topographic features in hadal trenches, yet their influence on microbial ecosystems and the global ocean remains unclear. Here, we conducted a topography-targeted investigation across 6-11 km of water depth within the Mariana Trench, integrating metagenomic, metaproteomic, and geochemical analyses. Coupled with high-resolution topographic mapping, our analyses reveal topography as an overlooked determinant of hadal geochemical and microbial heterogeneity. Convex areas exhibit classical sediment-depth-decay patterns with sparse, cooperative microbial communities.

Voltage Imaging with Engineered Proton-Pumping Rhodopsins: Insights from the Proton Transfer Pathway.

Voltage imaging using genetically encoded voltage indicators (GEVIs) has taken the field of neuroscience by storm in the past decade. Its ability to create subcellular and network level readouts of electrical dynamics depends critically on the kinetics of the response to voltage of the indicator used. Engineered microbial rhodopsins form a GEVI subclass known for their high voltage sensitivity and fast response kinetics. Here we review the essential aspects of microbial rhodopsin photocycles that are critical to understanding the mechanisms of voltage sensitivity in these proteins and link the

Early detection of pulmonary mucormycosis using microbial cell-free DNA sequencing of bronchoalveolar lavage fluid in neutropenic Hosts: A report of two cases.

Mucormycosis is an aggressive fungal infection characterized by angioinvasion, tissue necrosis, and high mortality rates. Microbial cell-free DNA (mcfDNA) sequencing provides rapid, non-invasive detection of microbial genetic material to guide diagnosis and treatment. We describe 2 cases of immunocompromised patients with invasive mucormycosis wherein mcfDNA sequencing significantly expedited diagnosis, allowing early surgical intervention and antimicrobial treatment.

CRISPR-Based Approaches to Engineer Nonmodel Bacteria for Bioproduction and Biotherapeutics.

Microbial diversity encompasses vast genetic and functional capacities, with immense potential for biotechnological applications. Yet, most biotechnological advances have been confined to a narrow set of model organisms, leaving the broader repertoire of nonmodel microbes largely untapped due to species-specific barriers that hinder genetic manipulation. Over the past decade, the advent of CRISPR (clustered regularly interspaced short palindromic repeats)-Cas (CRISPR-associated protein) systems has transformed microbial engineering by enabling precise, programmable, and scalable control of gen

Modeling taxa-abundance distributions in microbial communities using environmental sequence data.

We show that inferring the taxa-abundance distribution of a microbial community from small environmental samples alone is difficult. The difficulty stems from the disparity in scale between the number of genetic sequences that can be characterized and the number of individuals in communities that microbial ecologists aspire to describe. One solution is to calibrate and validate a mathematical model of microbial community assembly using the small samples and use the model to extrapolate to the taxa-abundance distribution for the population that is deemed to constitute a community. We demonstrat

Gene sharing has stabilised the genetic code.

The genetic code is nearly universal across life. Yet, The National Center for Biotechnology Information (NCBI) genetic code table recognises 27 distinct variants, most of which are confined to eukaryotic nuclei and organelles. Comparative genomics and synthetic recoding studies reveal that the code is far more flexible than once believed, but why has the standard code remained so remarkably conserved among prokaryotes? Here, we propose that horizontal gene transfer (HGT) acts as a stabilising evolutionary force by enforcing translational compatibility among gene-sharing organisms. In prokaryo

Quantifying Shared and Unique Gene Content across 17 Microbial Ecosystems.

Measuring microbial diversity is traditionally based on microbe taxonomy. Here, in contrast, we aimed to quantify heterogeneity in microbial gene content across 14,183 metagenomic samples spanning 17 ecologies, including 6 human associated, 7 nonhuman host associated, and 4 in other nonhuman host environments. In total, we identified 117,629,181 nonredundant genes. The vast majority of genes (66%) occurred in only one sample (i.e., "singletons"). In contrast, we found 1,864 sequences present in every metagenome, but not necessarily every bacterial genome. Additionally, we report data sets of o

Optimizing Therapeutic Potential of Fecal Transplant in Inflammatory Bowel Disease.

Inflammatory bowel disease (IBD) is a chronic inflammatory condition of the gastrointestinal tract influenced by genetic, environmental, immune, and microbial factors. Reduced gut microbial diversity and elevated proinflammatory bacteria levels in IBD disrupt mucosal immunity, barrier function, and inflammatory pathways. Fecal microbiota transplantation (FMT) is a potential therapy to restore microbial balance. Studies suggest that FMT may induce remission in mild-to-moderate ulcerative colitis but show limited efficacy in Crohn's disease and pouchitis. Donor microbiota colonization correlates

Bacillus alcalophilus RecJ (BaRecJ) can drive the adaptive evolution of S. cerevisiae.

With the continuous advancement of technologies such as microbial cultivation, DNA sequencing, bioinformatics, and genetic engineering, in vivo mutagenesis methods based on perturbation factors are now widely utilized. We identified a RecJ enzyme (BaRecJ) with endonuclease and exonuclease activities from Bacillus alcalophilus, and established a broad-spectrum mutagenic method based on the endonuclease and exonuclease activities of BaRecJ. The BaRecJ mutagenesis method was applied to S cerevisiae to enhance its ethanol or acetic acid tolerance, resulting in mutant strains with improved fermenta
