Sphingosine-1-phosphate Maintains Normal Vascular Permeability by Preserving Endothelial Surface Glycocalyx in Intact Microvessels.

OBJECTIVE: S1P was found to protect the ESG by inhibiting MMP activity-dependent shedding of ESG in cultured endothelial cell studies. We aimed to further test that S1P contributes to the maintenance of normal vascular permeability by protecting the ESG in intact microvessels. METHODS: We quantified the ESG in post-capillary venules of rat mesentery and measured the vascular permeability to albumin in the presence and absence of 1 μM S1P. We also measured permeability to albumin in the presence of MMP inhibitors and compared the measured permeability with those predicted by a transport model

A metalloproteinase secreted by Streptococcus pneumoniae removes membrane mucin MUC16 from the epithelial glycocalyx barrier.

The majority of bacterial infections occur across wet-surfaced mucosal epithelia, including those that cover the eye, respiratory tract, gastrointestinal tract and genitourinary tract. The apical surface of all these mucosal epithelia is covered by a heavily glycosylated glycocalyx, a major component of which are membrane-associated mucins (MAMs). MAMs form a barrier that serves as one of the first lines of defense against invading bacteria. While opportunistic bacteria rely on pre-existing defects or wounds to gain entry to epithelia, non opportunistic bacteria, especially the epidemic diseas…

Lung heparan sulfates modulate K(fc) during increased vascular pressure: evidence for glycocalyx-mediated mechanotransduction.

Lung endothelial cells respond to changes in vascular pressure through mechanotransduction pathways that alter barrier function via non-Starling mechanism(s). Components of the endothelial glycocalyx have been shown to participate in mechanotransduction in vitro and in systemic vessels, but the glycocalyx's role in mechanosensing and pulmonary barrier function has not been characterized. Mechanotransduction pathways may represent novel targets for therapeutic intervention during states of elevated pulmonary pressure such as acute heart failure, fluid overload, and mechanical ventilation. Our o…

Diagnostic Value of Glycocalyx Shedding in Blood for Differentiating between Parkinson's Disease and Multiple System Atrophy.

BACKGROUND: Blood-brain barrier disruption is increasingly recognized in synucleinopathies, but the role of the endothelial glycocalyx (GLX) in Parkinson's disease (PD) and multiple system atrophy (MSA) remains unclear. OBJECTIVES: The aim was to determine whether plasma GLX markers differ between PD, MSA, and healthy controls (HC), relate to clinical measures, and support differential diagnosis. METHODS: Nine GLX analytes were quantified in plasma from 38 PD, 24 MSA, and 46 HC. Group differences were tested with multilinear regression including age and sex; associations with disease duratio…

Shedding of endothelial glycocalyx and increased vascular permeability in patients with obstructive jaundice: A prospective observational study.

During the perioperative period, increased vascular permeability is associated with elevated morbidity and mortality. Obstructive jaundice (OJ) has been demonstrated to increase vascular permeability. The endothelial glycocalyx (EG), comprising hyaluronic acid (HA), chondroitin sulfate (CS), and syndecan-1 (SDC1), is essential for vascular integrity. However, its role in OJ-related vascular dysfunction remains unclear. This study aimed to investigate whether EG degradation contributes to vascular hyperpermeability in OJ patients. Plasma was preoperatively collected from 35 OJ patients and 33 n…

Time-of-day variation in complement activation and endothelial glycocalyx nanomechanics in ST-elevation myocardial infarction.

Time-of-day variation in myocardial infarction onset and ischemia-reperfusion injury is well established, but whether complement activation and the endothelial glycocalyx in ST-elevation myocardial infarction (STEMI) show temporal organization remains unclear. This study evaluated whether complement activation and endothelial glycocalyx nanomechanics in STEMI exhibit time-of-day structure and relate to short-term clinical outcomes. Endothelial glycocalyx height and stiffness and cortical stiffness were quantified in vitro using nanoindentation of human umbilical vein endothelial cells incubate…

Effects of Induced Endotoxaemia on Global Cardiovascular, Oxygenation and Haematologic Variables and the Integrity of the Endothelial Glycocalyx in the Horse.

BACKGROUND: Endotoxaemia is a significant cause of morbidity and mortality in equids due to perfusion impairment and possible destruction of the glycocalyx. OBJECTIVES: To evaluate our hypothesis that endotoxaemia induces changes in global cardiovascular and haematologic parameters and compromises glycocalyx integrity, evidenced by an early rise in plasma shedding products. STUDY DESIGN: In vivo experiments METHODS: In a prospective, randomised, controlled experimental trial, endotoxaemia was induced with E. coli B55:O5 LPS 30 ng kg-1 over 30 min IV in six healthy adult horses ventilated wit…

Endothelial Glycocalyx: The Missing Link Between Angiogenic Imbalance in Preeclampsia and Systemic Inflammation in HELLP Syndrome.

The pathophysiology of preeclampsia and HELLP syndrome relies on systemic vascular endothelial dysfunction, resulting from angiogenic imbala … the knowledge of maternal systemic endothelial and renal diseases. We hypothesize that endothelial glycocalyx may be the missing link between angiogenic factors, inflammatory regulation, and endothelial maternal lesions. Targeting the glycocalyx-endothelium axis may enable novel therapeutic strategies that delay delivery and reduce maternal-neonatal morbidi …

Atrial natriuretic peptide does not degrade the endothelial glycocalyx: A secondary analysis of a randomized porcine model.

BACKGROUND: The atrial natriuretic peptide (ANP) released from the heart regulates intravascular volume and is suspected to increase capillary permeability. Contradictory results regarding ANP and glycocalyx degradation have been reported. The aim of this study was to investigate if an infusion of ANP causes degradation of the endothelial glycocalyx. METHODS: Twenty pigs, pretreated with 250 mg methylprednisolone, were randomized to receive an infusion of either ANP (50 ng/kg/min) (n = 10) or 0.9% NaCl (n = 10) during 60 min. Increased Plasma Heparanase Activity and Endothelial Glycocalyx Degradation in Dengue Patients Is Associated With Plasma Leakage.

BACKGROUND: Endothelial hyper-permeability with plasma leakage and thrombocytopenia are predominant features of severe dengue virus infection. It is well established that heparanase, the endothelial glycocalyx degrading enzyme, plays a major role in various diseases with vascular leakage. It is yet to be elucidated whether heparanase activity plays a major role in dengue-associated plasma leakage. Moreover, the major source of heparanase secretion and activation in dengue remains elusive. Since a relatively high amount of heparanase is stored in platelets, we postulate that heparanase released…

An Acquired and Endogenous Glycocalyx Forms a Bidirectional "Don't Eat" and "Don't Eat Me" Barrier to Phagocytosis.

Macrophages continuously survey their environment in search of pathogens or apoptotic corpses or debris. Targets intended for clearance expose ligands that initiate their phagocytosis ("eat me" signals), while others avoid phagocytosis by displaying inhibitory ligands ("don't eat me" signals). We report that such ligands can be obscured by the glycosaminoglycans and glycoproteins that coat pathogenic as well as malignant phagocytic targets. In addition, a reciprocal barrier of self-synthesized or acquired glycocalyx components on the macrophage surface shrouds phagocytic receptors, curtailing …

Nanoarchitecture and dynamics of the mouse enteric glycocalyx examined by freeze-etching electron tomography and intravital microscopy.

The glycocalyx is a highly hydrated, glycoprotein-rich coat shrouding many eukaryotic and prokaryotic cells. The intestinal epithelial glycocalyx, comprising glycosylated transmembrane mucins, is part of the primary host-microbe interface and is essential for nutrient absorption. Its disruption has been implicated in numerous gastrointestinal diseases. Yet, due to challenges in preserving and visualizing its native organization, glycocalyx structure-function relationships remain unclear. Here, we characterize the nanoarchitecture of the murine enteric glycocalyx using freeze-etching and electr…

Changes in surface glycosylation and glycocalyx shedding in Trichobilharzia regenti (Schistosomatidae) during the transformation of cercaria to schistosomulum.

The invasive larvae (cercariae) of schistosomes penetrate the skin of their definitive hosts. During the invasion, they undergo dramatic ultrastructural and physiological transitions. These changes result in the development of the subsequent stage, schistosomulum, which migrates through host tissues in close contact with host's immune system. One of the striking changes in the transforming cercariae is the shedding of their thick tegumental glycocalyx, which represents an immunoattractive structure; therefore its removal helps cercariae to avoid immune attack. A set of commercial fluorescently…

The Endothelial Glycocalyx Controls Interactions of Quantum Dots with the Endothelium and Their Translocation across the Blood-Tissue Border.

Advances in the engineering of nanoparticles (NPs), which represent particles of less than 100 nm in one external dimension, led to an increasing utilization of nanomaterials for biomedical purposes. A prerequisite for their use in diagnostic and therapeutic applications, however, is the targeted delivery to the site of injury. Interactions between blood-borne NPs and the vascular endothelium represent a critical step for nanoparticle delivery into diseased tissue. Here, we show that the endothelial glycocalyx, which constitutes a glycoprotein-polysaccharide meshwork coating the luminal surfac…

Drug-induced shape change in erythrocytes correlates with membrane potential change and is independent of glycocalyx charge.

The influence of the anionic drugs indomethacin, barbitone, salicylate and the cationic drugs chlorpromazine and tetracaine on the morpholog … manner which is consistent with the bilayer couple hypothesis for drug-membrane interactions. Cell glycocalyx charge depletion did not affect the shape induced by either the cationic or anionic drugs. The result suggests that any sialic acid dependent electric potential at the bilayer surface is small compared to the cell's zeta potential.

Targeting heparanase-associated epithelial glycocalyx remodeling attenuates Akt/NF-κB-related barrier dysfunction in experimental COPD.

BACKGROUND: Chronic obstructive pulmonary disease (COPD) is characterized by persistent airway inflammation, epithelial barrier dysfunction, and progressive structural remodeling. Although glycocalyx impairment has been implicated in COPD pathogenesis, the mechanisms linking glycocalyx degradation to epithelial barrier injury remain incompletely understood. Heparanase (HPSE), the only mammalian endoglycosidase capable of degrading heparan sulfate, may contribute to COPD progression by regulating epithelial glycocalyx integrity. METHODS: HPSE expression was evaluated in serum samples from pati…

Preoperative Glycocalyx Status and Postoperative Organ Failure After Cardiac Surgery in the GLYPOCS Observational Cohort Study.

OBJECTIVES: Cardiac surgery with cardiopulmonary bypass induces systemic inflammation and glycocalyx degradation, both associated with postoperative complications. Cardiac surgery patients, who have multiple conditions affecting systemic microvascularization, may present with pre-existing alterations before surgery, and it is not known whether this increases the risk of postoperative complications. This study aimed to assess whether preoperative glycocalyx and microcirculatory impairments are associated with an increased risk of postoperative complications in cardiac surgery. DESIGN, SETTING,…

Changes in immune cell signatures during early infection reflect decoupling of capillary perfusion and glycocalyx dimensions.

INTRODUCTION: Microvascular injury is central to the pathophysiology of sepsis, but its interaction with the immune system in early infection is unclear. This study aimed to phenotype peripheral blood mononuclear cells (PBMC) from emergency department (ED) patients with suspected bacterial infection and correlate the results with microvascular changes. METHODS: This prospective observational study included 49 adult ED patients with suspected infection and 17 healthy controls. Capillary density and glycocalyx dimensions were measured by sublingual microscopy, while peripheral blood immune cell…
