In vivo antibacterial activity of RWJ-54428, a new cephalosporin with activity against gram-positive bacteria.

RWJ-54428 (MC-02,479) is a new cephalosporin with activity against resistant gram-positive organisms, including methicillin-resistant Staphylococcus aureus, vancomycin-resistant enterococci, and penicillin-resistant Streptococcus pneumoniae. The in vivo efficacy of RWJ-54428 was evaluated against gram-positive bacteria in four mouse models of infection. RWJ-54428 was effective in vivo against methicillin-susceptible and -resistant S. aureus in a mouse model of sepsis, with 50% effective doses being similar to those of vancomycin. In a single-dose neutropenic mouse thigh model of infection, RWJ…

Activity of linezolid against multi-resistant gram-positive bacteria from diverse hospitals in the United Kingdom.

The in vitro activity of linezolid, an oxazolidinone, was assessed against 374 gram-positive cocci, with an emphasis on testing multi-resistant, epidemiologically unrelated isolates. MICs of linezolid for staphylococci, pneumococci and streptococci had a narrow range, from 0.5 to 2 mg/L, whereas MICs for enterococci were uniformly 4 mg/L. For all the species tested, the MICs of linezolid were unrelated to those of other antimicrobials. Linezolid appears to be a potentially useful drug for infections caused by gram-positive cocci.

Expanding the antimicrobial spectrum of lugdunin: Discovery of multi-cationic derivatives of lugdunin with antimicrobial activity against gram-positive and gram-negative bacteria.

Lugdunin, a newly discovered antibiotic with a unique structure, emerged during a decade-long antibiotic discovery void and is considered a promising lead for combating drug-resistant bacteria. However, its narrow spectrum targeting only Gram-positive bacteria and its structural limitations have hindered its development and clinical application. Herein, inspired by our previous combinatorial modification strategies for lugdunin, we designed and synthesized a series of multi-cationic lugdunin derivatives using a biphenylmethyl modification on the tryptophan indole structure combined with multi-…

In Vitro Evaluation of the Antimicrobial Activity of Eighteen Essential Oils Against Gram-Positive and Gram-Negative Bacteria in Two Different Growth Media.

The rise in antimicrobial resistance and tolerance over time represents a significant threat to human and animal health. This has led to a n … dium used. However, the antimicrobial effects of the EOs were more pronounced in BHI than in MH for Gram-positive bacteria. While variations in antimicrobial activity were recorded among different species and strains, oregano EO proved to be the most effective against both Gram-positive and Gram-negative bacteria, followed by thyme and tea tree. Radiological findings in septic pulmonary embolism with detection of gram-negative and gram-positive bacteria in microbiological culture: A rare pediatric case report.

Septic pulmonary embolism rarely occurs in children, displaying fairly distinctive radiological features that can, however, lead to misdiagnosis. We present a case of an Indonesian pediatric patient with an infection originating from cellulitis in his right calf, who underwent a chest X-ray followed by a CT scan. The findings from these examinations revealed characteristics consistent with septic pulmonary embolism, correlating with the growth of both gram-positive and gram-negative bacteria in microbiological cultures obtained from wound specimens, as indicated in the literature. Awareness of

The Combined Use of Gentamicin and Silver Nitrate in Bone Cement for a Synergistic and Extended Antibiotic Action against Gram-Positive and Gram-Negative Bacteria.

Using bone cement as a carrier, gentamicin was for years the default drug to locally treat orthopedic infections but has lost favor due to increasing bacterial resistance to this drug. The objective of this study was to investigate the effect of combining gentamicin with silver nitrate in bone cement against S. aureus and P. aeruginosa. Antibacterial effects (CFU counts) of gentamicin and silver were initially studied followed by studies using subtherapeutic concentrations of each in combination. The release rates from cement were measured over 10 days and day 7 release samples were saved and

Biofilm-Related Infections in Gram-Positive Bacteria and the Potential Role of the Long-Acting Agent Dalbavancin.

Infections caused by Gram-positive bacteria are a major public health problem due to their increasing resistance to antibiotics. Staphylococcus and Enterococcus species' resistance and pathogenicity are enhanced by their ability to form biofilm. The biofilm lifestyle represents a significant obstacle to treatment because bacterial cells become highly tolerant to a wide range of antimicrobial compounds normally effective against their planktonic forms. Thus, novel therapeutic strategies targeting biofilms are urgently needed. The lipoglycopeptide dalbavancin is a long-acting agent for treating …

Promysalin is a salicylate-containing antimicrobial with a cell-membrane-disrupting mechanism of action on Gram-positive bacteria.

Promysalin was previously described as a narrow spectrum molecule with a unique species-specific activity against Pseudomonas aeruginosa. Here we demonstrate that promysalin is active against Gram-positive and Gram-negative bacteria using a microdilution assay. Promysalin acts on Gram-positive bacteria with a mechanism of action involving cell membrane damage with leakage of intracellular components. The evaluation of MICs and MBCs on 11 promysalin analogs, synthesized utilizing diverted total synthesis, allowed the identification of the structural moieties potentially involved in cell membran…

The effect of Indomethacin and Betamethasone on the cytokine response of human neonatal mononuclear cells to gram-positive bacteria.

Intrauterine infections with gram-positive bacteria pose a serious threat to neonates since they can result in neonatal sepsis, induce a fetal inflammatory response and also cause preterm birth. Despite intensive care, prematurity remains a leading cause of neonatal death, and is often accompanied by a number of morbidities. In order to prevent premature birth, tocolytic agents like Indomethacin are administered. Betamethasone is used to promote lung maturation and prevent respiratory distress syndrome. A combination of both drugs is assumed to prevent premature delivery while simultaneously f…

Structural optimization of natural product honokiol to discover novel membrane-targeting antibacterial agents against Gram-positive bacteria.

The global increase in infections caused by multidrug-resistant bacteria, coupled with a dwindling antibiotic pipeline, underscores an urgent need for new therapeutic agents. Inspired by the design principles of cationic antimicrobial peptides, we developed a new class of honokiol-based amphiphiles. From this series, compound 20C emerged as a promising lead, demonstrating potent in vitro activity against Gram-positive bacteria (MICs = 0.5-2 μg/mL). It exhibited a favorable safety profile characterized by low hemolytic (HC50 = 771.8 μg/mL) and cytotoxic activity (CC50 = 28.03 μg/mL), along with…
