PURPOSE: Pegylation of therapeutic protein usually results in a mixture of monopegylated proteins with differing sites of modification. With rh-interferon-alpha2A pegylation, we have found that this heterogeneity includes two classes of pegylation site chemistry, the relative proportions of which can be adjusted by reaction pH.
METHODS: The effect of pegylation reaction pH on the relative proportion of three peaks produced was investigated. Products were purified and characterized by peptide mapping, chemical stability to neutral hydroxylamine, and biologic activity.

[reaction: see text] A new capture-ROMP-release method for chromatography-free purification of N-hydroxysuccinimde Mitsunobu reactions is described. The Mitsunobu reaction captures a variety of alcohols onto a norbornenyl N-hydroxysuccinimide monomer. Subjection of the resulting crude reaction mixture to ROM-polymerization generates a polymer that can be precipitated with methanol and filtered from the Mitsunobu byproducts. Treatment of the polymer with hydrazine releases the substrate from the water-soluble polymer, producing a variety of O-alkylhydroxylamines with good purity.

A direct injection method has been developed for the determination of salicylic acid and its metabolites in urine. Urine samples are treated with hydroxylamine to convert salicyl acyl glucuronide to salicylhydroxamic acid, which can be accurately quantitated by direct injection into a high-performance liquid chromatographic system along with salicylic acid, gentisic acid and salicyluric acid. Some plastic syringes introduce a nitroxide free radical into the fluids they contain. The EPR spectrum shows a triplet with aN = 17.0 +/- 0.1 G in plasma. We report here that this nitroxide/hydroxylamine couple serves as an antioxidant system, whose redox potential appears to be between that of vitamin C and vitamin E. This adventitious antioxidant can alter the course of free radical processes.

A series of beta-hydroxylamine-phenylpropyl hydroxamic acids were prepared. The ability of these compounds to act as hypotensive agents was studied pharmacodynamically. Key structural features for the hypotensive action were identified and correlated to the degree of responses observed. It was ascertained that these compounds do not act through the adrenergic, cholinergic and histaminergic receptor system, but can be classified as direct acting vasodilators.

We adapted a simple hydroxylamine-based indole assay to detect indole from stored vaginal swab specimens from women with and without bacterial vaginosis (BV). A green iron-chlorin protein was purified 160-fold from a lyophilized extract of Aspergillus niger by ion-exchange chromatography and gel filtration with a yield of 25%. A method is described for isolation of orotic acid from rat liver tissue for study of the kinetics of its radioisotopic labelling. Within 24 hrs after injection of 400 mg hydroxylamine per kg of body mass, the concentration of hepatic orotate decreased, while its transport into liver tissue and the kinetics of labelling of its endogenous pool remained unaffected.
