Postmortem Biochemistry and Immunohistochemistry in Anaphylactic Death Due to Hymenoptera Sting: A Forensic Case Report.

BACKGROUND: Postmortem assessment of anaphylactic death is a challenge for forensic pathologists. One of the most frequent elicitors of anaphylaxis is insect venom. Here, a case of anaphylactic death due to Hymenoptera stings is reported to highlight the contribution of postmortem biochemistry and immunohistochemistry in assessing the cause of death. CASE REPORT: A 59-year-old Caucasian man working on his farm was presumably stung by a bee and died. He had a history of previous sensitization to insect venom. The autopsy revealed no signs of insect puncture, mild edema of the larynx, and foamy…

Sensitivity of a Hymenoptera serological immunoglobulin (Ig)E assay for the diagnosis of venom hypersensitivity in dogs.

BACKGROUND: Hymenoptera envenomation with honey bee (Apis mellifera) and paper wasp (Polistes spp.) may cause life-threatening anaphylaxis in dogs. In human patients, clinical history, intradermal testing (IDT) and measurement of allergen-specific serological immunoglobulin (Ig)E (sIgE) are used to support a diagnosis of Hymenoptera venom hypersensitivity. The utility of venom allergen-specific sIgE has not yet been evaluated for this purpose in dogs. OBJECTIVES: The objective of the study was to investigate the sensitivity (sn), specificity (sp) and positive predictive value (PPV) of honey b…

Lethal and Demographic Impact of Chlorpyrifos and Spinosad on the Ectoparasitoid Habrobracon hebetor (Say) (Hymenoptera: Braconidae).

The appropriate use of biological agents and chemical compounds is necessary to establish successful integrated pest management (IPM) programs. Thus, the off-target effects of pesticides on biological control agents are essential considerations of IPM. In this study, the effects of lethal and sublethal concentrations of chlorpyrifos and spinosad on the demographic parameters of Habrobracon hebetor (Say) (Hymenoptera: Braconidae) were assessed. Bioassays were carried out on immature and adult stages by using dipping and contact exposure of dry pesticide residue on an inert material, respectivel…

Clinical Features and the Diagnostic Value of Component Allergen-Specific IgE in Hymenoptera Venom Allergy.

PURPOSE: Although patient history is vital for the diagnosis of hymenoptera venom allergy, specific IgE detection is also important to identify the culprit insect and monitor the effect of immunotherapy. We evaluated the diagnostic value of serum-specific IgE detection of hymenoptera venom component allergens and documented changes in allergen-specific IgE after immunotherapy. METHODS: Fifty-six hymenoptera venom allergy patients receiving venom immunotherapy were recruited from Ajou University Hospital, Korea. The clinical manifestations of the patients were noted, and serum-specific IgE det…

Investigations on Oecophylla longinoda (Latreille) (Hymenoptera: Formicidae) as a biocontrol agent in the protection of cashew plantations.

BACKGROUND: Cashew (Anacardium occidentale L.) has become a very important non-traditional tree crop in Ghana. The crop is, however, attacked by sap-sucking insects, particularly the mosquito bug, Helopeltis schoutedeni Reuter, the leaf-footed bug, Pseudotheraptus devastans (Dist.), and the coreid bug, Anoplocnemis curvipes (F.), which feed on shoots, panicles and fruits. Their damage is characterised by withering of the latter. In Ghana, Oecophylla longinoda Latr. occurs in large numbers on cashew and other native plants, but little is known about its relationship with insect pests. The relat

Distribution, prevalence and host associations of Hymenoptera parasitic on Calliphoridae occurring in flystrike in New Zealand.

Between 1920 and 1930, four species of parasitoid Hymenoptera (Tachinaephagus zealandicus, Alysia manducator, Nasonia vitripennis and Brachymeria ucalegon) were imported to New Zealand as an aid in the control of the blowflies causing myiasis in sheep (flystrike). Their long-term effects have never been investigated and the extent to which they were successful in establishing and enlarging their ranges has been given scant regard over the last 60 years. A long-term epidemiological study of flystrike in New Zealand between 1984 and 1996 obtained 4061 samples of blowfly larvae from flystruck she…

IgE antibodies to Hymenoptera venoms in the serum are common in the general population and are related to indications of atopy.

Determination of Hymenoptera venom (HV)-specific serum IgE antibodies is a useful diagnostic method in patients with systemic anaphylactic reaction (SAR) to Hymenoptera stings. In a general population cohort, we determined the prevalence of SAR and HV-specific IgE antibodies and assessed parameters associated with the latter. A total of 277 voluntarily participating inhabitants of rural Bavaria (Germany) (232 adults, mean age 38.0 years; 45 children, mean age 8.4 years) were investigated for a history of atopic disease or SAR to insect stings; in 258 of these, total IgE and specific IgE antibo…

Evolution of sensitivity to Hymenoptera venom in 200 allergic patients followed for up to 3 years.

Duration of venom immunotherapy still remains questionnable, since some patients outgrow their sensitivity. Two hundred venom-allergic patients (4 to 82 years; mean +/- SD, 37.3 +/- 14.2 years) who were first observed with systemic reactions (SRs) were investigated yearly for up to 3 years. Among these patients, 72 were allergic to honeybee venom, 83 to yellow jacket venom, and 45 to both yellow jacket and Polistes venom. At each visit patients had skin tests and RAST to venom. All patients had the same semirush protocol and a monthly maintenance dose of 200 micrograms. After 1 year of treatme

The first two complete mitochondrial genomes for the genus Neotrichoporoides (Hymenoptera, Eulophidae) and their phylogenetic analysis.

Neotrichoporoides belongs to the family Eulophidae (Hymenoptera: Chalcidoidea). As a group of parasitic wasps, it plays an indispensable role in the biological control of agricultural and forest pests and in maintaining ecosystem balance. To date, only nine complete mitochondrial genomes of Eulophidae have been sequenced worldwide, including the two newly sequenced species in this study. To enrich our understanding of the mitochondrial genomic diversity of Eulophidae and to provide preliminary insights into its phylogenetic relationships, we sequenced and comparatively analyzed the mitochondri…

Identification and validation of reference genes for RT-qPCR analysis in Sclerodermus guani (Hymenoptera: Bethylidae).

Gene expression studies in organisms are often conducted using reverse transcriptase-quantitative polymerase chain reaction (RT-qPCR), and the accuracy of RT-qPCR results relies on the stability of reference genes. We examined ten candidate reference genes in Sclerodermus guani, a parasitoid wasp that is a natural enemy of long-horned beetle pests in forestry, including ACT, EF1α, Hsc70, Hsp70, SRSF7, α-tubulin, RPL7A, 18S, 28S, and SOD1, regarding variable biotic and abiotic factors such as body part, life stage, hormone, diet, and temperature. A Pyr-loaded polymer microparticle for effectively controlling Solenopsis invicta (Hymenoptera: Formicidae) in the nest.

Human interference and incorrect use of pesticides are easy to induce red imported fire ant (RIFA) escape and migrate from a nest, resulting in ineffective control of RIFA. In order to avoid RIFA alert, we designed an amphiphilic PSI-mPEG-Boc-DAH loaded Pyr to make the microparticles with effective controlled release. The investigation showed that the quantity of Pyr released by Pyr@PSI-mPEG-Boc-DAH under acidic environment was only 36.40 ± 1.90% at 48 h, whereas the release rate of original Pyr was 75.23 ± 5.71%. And the RIFA mortality rate of 1 ppm Pyr in Pyr@PSI-mPEG-Boc-DAH microparticles

The antennal transcriptome analysis and characterizations of odorant-binding proteins in Megachile saussurei (Hymenoptera, Megachilidae).

BACKGROUND: Odorant-binding proteins (OBPs) are essential in insect's daily behaviors mediated by olfactory perception. Megachile saussurei Radoszkowski (Hymenoptera, Megachilidae) is a principal insect pollinating alfalfa (Medicago sativa) in Northwestern China. The olfactory function have been less conducted, which provides a lot of possibilities for our research. RESULTS: Our results showed that 20 OBPs were identified in total. Multiple sequence alignment analysis indicated MsauOBPs were highly conserved with a 6-cysteine motif pattern and all belonged to the classic subfamily, coding 113…

Precision Medicine in Hymenoptera Venom Allergy: Diagnostics, Biomarkers, and Therapy of Different Endotypes and Phenotypes.

Allergic reactions to stings of Hymenoptera species may be severe and are potentially fatal deviations of the immunological response observed in healthy individuals. However, venom-specific immunotherapy (VIT) is an immunomodulatory approach able to cure venom allergy in the majority of affected patients. An appropriate therapeutic intervention and the efficacy of VIT not only depend on a conclusive diagnosis, but might also be influenced by the patient-specific manifestation of the disease. As with other diseases, it should be borne in mind that there are different endotypes and phenotypes of…

Extraction and preliminary chemical characterization of the venom of the spider wasp Pepsis decorata (Hymenoptera: Pompilidae).

Arthropod venoms may be considered important sources of bioactive molecules; however, technical difficulties, such as venom extraction and homogeneity may impair the biochemical identification of new molecules. In this context, we have developed a method to maintain wasps in captivity that allows the collection of the venom, without use of chemical, mechanical or electrical stimuli. The crude venom was analyzed by RP-HPLC-ESIQ-ToF and 20 peptides were identified by de novo peptide sequencing, among them Eumenine-Mastoparan and a Ponericin-G2-simile peptide.

Development of Liquid Bait With Unique Bait Station for Control of Dolichoderus thoracicus (Hymenoptera: Formicidae).

The Dolichoderus thoracicus (Smith) has seriously infested urban, village, and agricultural environments in Taiwan. To develop optimal bait for the effective control of D. thoracicus, we investigated the feeding preferences of this ant at different sugar and amino acid concentrations in a laboratory setting. The number of feeding workers was significantly higher for 20% sucrose compared with that for other sugars at 10% and 20% concentrations. Furthermore, among all tested concentrations of monosodium glutamate (MSG; 1%, 3%, 5%, and 10%), the highest number of ants were attracted by 1% MSG wit

Morphology and ultrastructure of Dufour's and venom glands in the ant Camponotus japonicus Mayr (Hymenoptera: Formicidae).

The Dufour's and venom glands are two important exocrine glands in ants. In this study, the morphology and fine structure of these two glands are described in the ant Camponotus japonicus Mayr. The Dufour glands have a characteristic bilobed shape and show a difference in size and color between the female castes (minor and major workers, alate and dealate queens). The external surface of Dufour's gland shows different features among the female castes. It appears more hypertrophied in major workers than in the other castes, indicating for a more pronounced function. The cells of the glandular e
