In anaesthetized rats studied in a body plethysmograph, 1-(4'6-diallylamino-2'-triazinyl)-4(bis parafluorobenzydryl) piperazine (almitrine) caused large increases in ventilation (VE) which were abolished by cutting the IXth and Xth cranial nerves. Maximal effects followed infusions of 0.5-1 mg X kg-1; the solvent had no action. When the rats breathed 10% O2 or 10% O2 + 5% CO2, gas mixtures which increased VE, almitrine caused further increases in VE and improved blood gas tensions. Chronically hypoxic (4 weeks in 10% O2) and chronically hypoxic and hypercapnic rats (4 weeks in 10% O2 + 4% CO2)

Halothane is known to inhibit the ventilatory responses to hypoxia and hypercapnia. In order to determine whether this inhibition was mediated by peripheral chemoreceptors, the authors measured the effect of halothane on the response of carotid body chemoreceptors to these stimuli. Cats were decerebrated under brief halothane anesthesia, paralyzed, and ventilated. Chemoreceptor activity was recorded from single- or few-fiber preparations of carotid sinus nerve, and the inspiratory drive was recorded from the whole phrenic nerve. Steady-state responses were measured at three levels of CO2 tensi…

With the aid of potassium-sensitive microelectrodes reinforced by bitumen (tip diameter, 1.5 micrometer), extracellular potassium activity ([K+]e) and DC potential were measured in the cat's carotid body. Under normoxic and normocapnic conditions, potassium values of 1--16 mM (mean value 7.2 mM, standard deviation 3.8 mM) and DC potential values of -11 mV to + 13 mV were recorded. With hypoxia, [K+5e increased by between 1 mM and 9 mM; DC potential was reduced by between 0.5 and 3 mV. With hypercapnia, [K+]e decreased by between 1 mM and 5 mM changes in DC potential were variable. The results …

Changes in the ability to discriminate speech from a eucapnic state to hyperventilation and hypercapnia were investigated. Standard speech audiometric techniques were employed to determine the speech reception threshold and the speech discrimination values, while respiratory conditions were varied and measured utilizing a mixed-gas breathing method. Respiratory parameters were similar to those encountered in aircraft personnel who experience oxygen/pressure system malfunction. The results of the study suggest no significant change in the speech reception threshold while in a hyperventilated or…

The effects of ventilatory hypoxia and hypercapnia and perfusion hypoxia and hypercapnia on pulmonary vascular resistance were studied in the intact lamb using right heart techniques to isolate and perfuse the left lower lobe. Ventilatory hypoxia increased vascular resistance in the left lower lobe by constricting predominantly vessels upstream from small lobar veins, presumably small arteries. The response to hypoxia was not blocked by phentolamine and diphenhydramine in doses that markedly decreased pressor responses to norepinephrine and histamine in the lung. Perfusion hypoxia did not alte…

STUDY OBJECTIVES: To elucidate the neural mechanisms underlying hypercapnia triggered micro-arousal events during sleep apnea, focusing on neuronal activation and synaptic functions in medial parabrachial nucleus (MPB).
METHODS: Mice underwent hypercapnic challenges to induce micro-arousal events, which were recognized by cortical EEG and hippocampal local field potential (LFP) during NREM sleep. Neuronal activation was quantified via Fos immunohistochemistry in MPB. Neuronal excitability and spontaneous excitatory postsynaptic current (sEPSC) under hypercapnia in MPB slices were assessed by …

BACKGROUND: Right ventricular (RV) failure is a serious complication after cardiac surgery. Postoperative hypercapnic acidosis can increase … f the RV. Measurements were made in normocapnia (target PaCO2 5 kPa) and in hypoventilation induced hypercapnia (target PaCO2 7.5 kPa). A paired t-test was used to compare the measured differences in the values under normo- and hypercapnia.
RESULTS: Altogether, 20 patients were included in the study. We achieved a significant (p < 0.05) difference both in Pa …

PURPOSE: To evaluate the relationship between both dynamic and steady-state measures of cerebrovascular reactivity (CVR) and the progression of age-related white matter disease.
METHODS: Blood oxygen level-dependent (BOLD) MRI CVR scans were acquired from forty-five subjects (age range: 50-90 years, 25 males) with moderate to severe white matter disease, at baseline and one-year follow-up. To calculate the dynamic (τ) and steady-state (ssCVR) components of the BOLD signal response, the PETCO2 signal waveform was convolved with an exponential decay function. The τ corresponding to the best fit

Hypercapnia, an elevation of the level of carbon dioxide (CO2) in blood and tissues, is a marker of poor prognosis in chronic obstructive pulmonary disease and other pulmonary disorders. We previously reported that hypercapnia inhibits the expression of TNF and IL-6 and phagocytosis in macrophages in vitro. In the present study, we determined the effects of normoxic hypercapnia (10% CO2, 21% O2, and 69% N2) on outcomes of Pseudomonas aeruginosa pneumonia in BALB/c mice and on pulmonary neutrophil function. We found that the mortality of P. aeruginosa pneumonia was increased in 10% CO2-exposed …

OBJECTIVE: The online rating of perceived breathlessness in a rebreathing test (RT) is a new and powerful technique to analyze the psychological and physiological process related to the mechanisms of breathlessness. The aim of the present study was to assess the influence of rating type on respiratory sensation and behavior during repeated hypercapnic exposures.
METHODS: Sixty-one healthy women performed three rebreathing trials. Half of them were auditorily cued to rate breathlessness intensity every 12 s (CR, n=30), the other half was instructed to rate breathlessness each time a change in

The objective of the present study was to examine the relative roles of peripheral versus central mechanisms in producing ventilatory adjustments in pekin ducks during prolonged (5 h) hypercapnia (5% inspired CO2), and to determine whether these adjustments differed between male and female ducks. After 20 min of CO2 exposure, intact ducks increased total ventilation (VE) 2.5-3-fold above control values, due to large increases (approximately 200%) in tidal volume (VT) and slightly smaller increases (approximately 140%) in breathing frequency (fR). This response was accompanied by respiratory ac…

To show relationship between degree of carotid arterial stenosis and cerebral blood flow reactivity (RES%) to induced hypercapnia, fluorine-18-fluoromethane and positron emission tomography (PET) was used to study 18 patients with carotid distribution transient ischaemic attacks (TIA), all free of stroke, who had angiographic-proven unilateral arterial disease. Non-involved carotid arteries were either normal or had non-stenotic plaque. Either normal arteries or nonstenotic ulcerations in the symptomatic carotid arteries were present in five of 18 (28%), ipsilateral carotid stenosis from 50-99…

BACKGROUND: Respiratory failure in idiopathic interstitial pneumonia (IIP) is mainly non-hypercapnic hypoxia, although hypercapnia also occurs in advanced stages. Although sleep-disordered breathing in IIPs is drawing attention, the role of nocturnal hypercapnia remains largely unknown. In this study, we aimed to clarify whether nocturnal hypercapnia in patients with advanced IIPs is associated with the clinical characteristics.
METHODS: We retrospectively analyzed 41 patients with IIPs who underwent nocturnal transcutaneous carbon dioxide partial pressure (PtcCO2) monitoring during the evalu…

Recovery after an ischemic stroke depends not only on neuronal survival but also on inflammatory mechanisms that determine secondary injury and reparative plasticity. This review summarizes the evidence on hypoxic conditioning, permissive hypercapnia, and their combined application as modulators of neuroinflammation and neurorehabilitation. This review does not aim to describe the fundamental mechanisms of neuroinflammation, but rather to examine how hypoxia, hypercapnia, and their interaction provide potential targets for its modulation. Prolonged or severe hypoxia exacerbates neuroinflammati…

Hypoventilation is a complication that is not uncommon in chronic obstructive pulmonary disease and calls for both medical treatment of the underlying disease and, frequently, noninvasive ventilation either during exacerbations requiring hospitalization or in a chronic state in the patient at home. Obesity hypoventilation syndrome by definition is associated with ventilatory failure and hypercapnia. It may or may not be accompanied by obstructive sleep apnea, which when detected becomes an additional target for positive airway pressure treatment. Intensive research has not completely resolved …
