Lung pathology and immediate hypersensitivity in a mouse model after vaccination with pertussis vaccines and challenge with Bordetella pertussis.

While evaluating vaccine efficacy against clinical Bordetella pertussis isolates in mice, after challenge vaccinated mice showed increased lung pathology with eosinophilia, compared to challenged, non-vaccinated animals. This led us to study bacterial clearance, lung pathology, lung TNF-alpha expression, and parameters of immediate hypersensitivity (IH), being serum IgE levels, eosinophil numbers in the bronchoalveolar lavage fluid, and ex vivo IL-4, IL-5, IL-10, IL-13, and IFN-gamma production by the bronchial lymph node cells. BALB/c mice received a combined Diphtheria (D), Tetanus (T), Poli

Readministration of high-dose methotrexate in a patient with suspected immediate hypersensitivity and T-cell acute lymphoblastic lymphoma.

There is a paucity of literature addressing the management of methotrexate (MTX) hypersensitivity. We developed a high-dose MTX readministration protocol based on a modified, prolonged carboplatin desensitization protocol. Over 1.5 hours, 1/1000 of the total intravenous dose was administered followed by 1/100 over 1.5 hours, 1/10 over 6 hours, and the rest of the full dose over 24 hours. MTX readministration was successfully tolerated on three occasions in a 17-year-old male patient with T-cell acute lymphoblastic lymphoma and a history of urticarial reactions to MTX. This high-dose MTX readmi

Transfer of an antigen-specific immediate hypersensitivity-like reaction with an antigen-binding factor produced by T cells.

The fact that T cell-dependent activation of mast cells occurs in delayed-type hypersensitivity led us to investigate whether a T cell product could mimic some of the functions of IgE. We report that 24- or 48-hr cultures of T cells from mice immunized optimally for delayed-type hypersensitivity resulted in release of an antigen-binding factor that transferred the ability to elicit an antigen-specific immediate hypersensitivity-like skin reaction in normal recipients. The responsible factor was concentrated and purified by affinity chromatography on antigen columns and was distinguished from i

NON-SPECIFIC INHIBITION OF THE IMMEDIATE AND DELAYED TYPES OF HYPERSENSITIVITY DURING IMMUNE PARALYSIS OF ADULT GUINEA-PIGS.

Immunological paralysis induced in adult guinea-pigs with various protein overloading treatments is specific only at the beginning and soon after the end of the paralysis-inducing treatment. Reported experiments show that during a period which starts between the 8th and 10th days of the paralysing treatment and finishes when this treatment is discontinued, there is established an unresponsiveness towards antigens unrelated to the paralysing one. This non-specific unresponsiveness is manifested by the inhibition of reactions of both immediate and delayed types of hypersensitivity, at least to c

Immediate hypersensitivity to Metastrongylus spp. infection in the pig. I. The passive transfer of skin sensitivity to uninfected recipients.

Homocytotropic antibody has been shown to occur in pigs experimentally infected with lungworms, Metastrongylus spp. This antibody is heat labile, inactivated by 2-mercaptoethanol and persists at passively sensitized skin sites for up to 14 days. On electrophoresis in a polyvinyl copolymer at pH 8.6 homocytotropic antibody was recovered in the fast γ-slow β area. Gel filtration of antiserum indicated that the molecular size of homocytotropic antibody was intermediate between 19S and 7S, and on DEAE cellulose the antibody was eluted with the bulk of IgA. Attempts to separate homocytotropic antib

Postoperative morbidity after suspected grade III perioperative immediate hypersensitivity reactions with completed surgery: a matched case-control study.

BACKGROUND: Severe immediate perioperative hypersensitivity reactions (POH) are uncommon but potentially life-threatening events. Postoperative morbidity after suspected grade III POH when surgery is completed remains uncertain. This study compared postoperative morbidity between patients with suspected grade III POH whose surgery was completed and matched controls. METHODS: We conducted a retrospective, single-centre, matched case-control study at Lille University Hospital between 2012 and 2023. Cases were adults with suspected grade III POH, defined by a Hypersensitivity Clinical Scoring Sc

Anaphylactic reaction to carboplatin diagnosed by skin testing-a reliable tool in platinum-based immediate-type hypersensitivity reactions.

Immediate-type hypersensitivity reactions (IHRs) to carboplatin (CA) are most commonly reported in ovarian cancer patients. A 54-year-old woman with stage IV melanoma suffering from metastasis in the entire right lower extremity was presented to our allergy outpatient clinic for diagnostic work-up due to an anaphylactic reaction with palmoplantar erythema, conjunctivitis along with facial erythema, and an incipient decrease in blood pressure during a chemotherapy regimen with dacarbazine and carboplatin upon re-administration. A subsequently carried out allergological work-up with skin testing

Immediate hypersensitivity to human recombinant granulocyte-macrophage colony-stimulating factor associated with a positive prick skin test reaction.

BACKGROUND: Recombinant human granulocyte-macrophage colony-stimulating factor (rhu GM-CSF), also known as sargramostim, is used to accelerate myeloid recovery following bone marrow transplantation or cytotoxic chemotherapy. "Anaphylactic" reactions to sargramostim have been reported on a limited basis and are poorly characterized. OBJECTIVE: It is the purpose of this report to describe an adverse reaction to sargramostim treatment involving palmar itching, urticaria, angioedema, and throat tightness and to demonstrate the utility of prick skin testing to determine type I sensitization. Immediate hypersensitivity skin reactions to human dander in atopic dermatitis. Partial purification and characterization of human dander allergens.

Using high-speed gel filtration chromatography, an aqueous extract of human dander was divided into four fractions (fractions I to IV). Scratch tests with the crude human dander extract and each fraction were performed in 147 patients with atopic dermatitis and 100 control subjects. The crude extract provoked positive reactions in 61.9% (91/147) of patients with atopic dermatitis and 6% (6/100) of control subjects. The most potent allergenic component was present in fraction II, which induced positive reactions in 61.2% (90/147) of patients with atopic dermatitis. Sodium dodecyl sulfate polyac

Patients with previous immediate hypersensitivity reactions to polyethylene glycol can safely receive the BNT162b2 mRNA COVID-19 vaccine.

Previous anaphylaxis or immediate allergic reaction to polyethylene glycol (PEG; also known as macrogol) is considered a contraindication to the BNT162b2 mRNA COVID-19 vaccine, which contains 50 ug of PEG at a molecular weight of 2000, and this component is thought to account for the higher rate of anaphylaxis seen with this vaccine (4.7 per million doses) than with other non-mRNA vaccines. However, there is evidence that both anaphylaxis to PEG and anaphylaxis to the Pfizer COVID-19 reaction may not be IgE-mediated, with patients with anaphylaxis to first dose of the Pfizer COVID-19 vaccine r

Antibiotic-induced immediate type hypersensitivity is a risk factor for positive allergy skin tests for neuromuscular blocking agents.

BACKGROUND: Skin tests for neuromuscular blocking agents (NMBAs) are not currently recommended for the general population undergoing general anaesthesia. In a previous study we have reported a high incidence of positive allergy tests for NMBAs in patients with a positive history of non-anaesthetic drug allergy, a larger prospective study being needed to confirm those preliminary results. The objective of this study was to compare the skin tests results for patients with a positive history of antibiotic-induced immediate type hypersensitivity reactions to those of controls without drug allergie

Penicillin allergy: anti-penicillin IgE antibodies and immediate hypersensitivity skin reactions employing major and minor determinants of penicillin.

300 children considered to have had adverse reactions to penicillin were examined. Informed consent was obtained from the parents. Skin tests were conducted by the scratch/prick and intradermal techniques, using benzylpenicilloyl polylysine conjugate and a mixture of minor determinants of penicillin. Specific anti-penicillin IgE antibodies were estimated by the radioallergosorbent test. There was a good correlation between the two methods. The overall frequency of positive tests was 19%. 11 children showed cutaneous reactivity only to the minor determinants mixture. Positive results were found

Detection of drug-specific immunoglobulin E (IgE) and acute mediator release for the diagnosis of immediate drug hypersensitivity reactions.

The diagnosis of a drug hypersensitivity reaction (DHR) is complex. The first step after taking the clinical history is to look for a sensitization to confirm or exclude the diagnosis and to identify the culprit drug. Skin tests are the primary means of detecting sensitization in DHR, but are associated with a risk for a severe reaction and may be contraindicated. In vitro tests offer the potential to support or confirm a diagnosis of DHR and influence medical decision making. For immediate-type DHR, a few validated assays for measurement of specific IgE (sIgE) are commercially available to a

Compound Kushen Injection Induces Immediate Hypersensitivity Reaction Through Promoting the Production of Platelet-Activating Factor via de Novo Pathway.

Compound Kushen Injection (CKI) is a bis-herbal formulation extracted from Kushen (Radix Sophorae Flavescentis) and Baituling (Rhizoma Heterosmilacis Yunnanensis). Clinically, it is used as the adjuvant treatment of cancer. However, with the increased application, the cases of immediate hypersensitivity reactions (IHRs) also gradually rise. In this study, we investigated the underlying mechanism(s) and active constituent(s) for CKI-induced IHRs in experimental models. The obtained results showed that CKI did not elevate serum total IgE (tIgE) and mouse mast cell protease 1 (MMCP1) after consec

Food immediate-contact hypersensitivity (FICH) and elimination diet in young children with atopic dermatitis. Preliminary results in 107 children.

In atopic dermatitis [AD], not only food consumption, but direct skin-contact too can provoke hypersensitivity reactions. We imitated food immediate-contact hypersensitivity [FICH] to cow's milk, egg, peanut or soy by a skin provocation test. This skin application food test [SAFT] was applied in 91 patients aged up to 5 years and suffering from AD, and in 16 healthy controls (all SAFT-negative). In the SAFT-positive patients (n = 61), FICH to egg was observed in 72%, to cow's milk in 47%, to peanut in 34% and soy in only 1 patient. SAFT and RAST scores correlated weakly. Nevertheless, many dis

In Vitro Research Tools in the Field of Human Immediate Drug Hypersensitivity and Their Present Use in Small Animal Veterinary Medicine.

Drug hypersensitivity reactions (DHR) are immune-mediated idiosyncratic adverse drug events. Type I DHR are often referred to as "immediate" and involve B lymphocyte-secreted IgE that bind to the membrane of basophils and mast cells, inducing their degranulation. This review presents various in vitro tests that were developed in the field of human type I HS and implemented as clinical diagnostic tools in human cases of immediate DHR. The respective strengths and weaknesses of each test will be discussed in parallel of validation data such as specificity and sensitivity whenever available. Some

Immediate-type hypersensitivity to a bean lectin source with commentary on occupational allergy in allergy-immunology laboratory and research workers.

A case of acute, severe allergic conjunctivitis is reported in a biochemist who had been grinding Bandeiraea simplicifolia, the source of an anti-B lectin. A crude extract of these beans produced immediate-type skin test reactivity, and reaginic antibodies were demonstrated in her serum. Several other examples of occupationally induced allergy in laboratory and research workers in the field of allergy-immunology are briefly cited. Exposure to potential allergens in a dry, powdered form seems particularly hazardous.

Immediate Reactions to More Than 1 NSAID Must Not Be Considered Cross-Hypersensitivity Unless Tolerance to ASA Is Verified.

BACKGROUND AND OBJECTIVES: Individuals who develop drug hypersensitivity reactions (DHRs) to chemically unrelated nonsteroidal anti-inflammatory drugs (NSAIDs) are considered cross-hypersensitive. The hallmark for this classification is that the patient presents a reaction after intake of or challenge with acetylsalicylic acid (ASA). Whether patients react to 2 or more NSAIDs while tolerating ASA remains to be studied (selective reactions, SRs). Objective: To identify patients with SRs to 2 or more NSAIDs including strong COX-1 inhibitors. METHODS: Patients who attended the Allergy Service of
