Evident hypopigmentation without other ocular deficits in Dutch patients with oculocutaneous albinism type 4.

To describe the phenotype of Dutch patients with oculocutaneous albinism type 4 (OCA4), we collected data on pigmentation (skin, hair, and e … C > T; (p.Pro104Ser), and c.1368 + 3_1368 + 9del; (p.?). OCA4 patients of this Dutch cohort all had hypopigmentation of skin, hair, and iris translucency. However, patients were either severely affected with regard to visual acuity, foveal hypoplasia, and misrouting, or visually not affected at all. We describe for the first time OCA4 patients with an evident lack …

Linear hypopigmentation and cutaneous atrophy following intra-articular steroid injections for de Quervain's tendonitis.

Intralesional and intra-articular corticosteroids are commonly used therapies in the fields of dermatology, orthopedic surgery and rheumatology. The authors present the case of a 32-year-old female who received intra-articular corticosteroids for de Quervain's tendonitis and developed bilateral hypopigmented patches near the areas of injection. Unilateral linear extension of the hypopigmentation was subsequently noted, presumably secondary to venous or lymphatic uptake of corticosteroid crystals. The patient complained of temperature sensitivity and increased skin fragility affecting all of th…

Association between the skin microbiome and lichen striatus hypopigmentation: Cutibacterium acnes as a potential cause.

BACKGROUND: Lichen striatus (LS) is an acquired skin disorder with a linear pattern along Blaschko's lines. It commonly occurs in childhood, and the lesions spontaneously regress within several months. OBJECTIVES: Although up to 50% of LS cases exhibit hypopigmentation that can persist for several months to years, it is unknown why LS is associated with such a high incidence of hypopigmentation compared to other inflammatory skin diseases. Therefore, this study aimed to analyse the differences in the skin microbiome between LS patients with and without hypopigmentation. METHODS: Differences …

A novel pathogenic RHOA variant in a patient with patterned cutaneous hypopigmentation associated with extracutaneous findings.

RHOA-related neuroectodermal syndrome is characterised by linear skin hypopigmentation along Blaschko's lines associated with alopecia, leukoencephalopathy, facial and limb hypoplasia, and ocular, dental, and acral anomalies. Herein, we report a patient with patterned cutaneous hypopigmentation with a similar phenotype due to a novel postzygotic RHOA variant (c.210G>T; p.Arg70Ser). This illustrates that the complexity of the orchestration of morphogenesis and organogenesis can be affected by different variants in the same gene.

[Clinical phenotype and genetic analysis of a Chinese pedigree affected with familial progressive hyperpigmentation and hypopigmentation].

OBJECTIVE: To explore the clinical phenotype and genetic basis for a Chinese pedigree affected with familial progressive hyperpigmentation and hypopigmentation (FPHH). METHODS: Clinical data and family history for a child with FPHH were collected. Peripheral blood samples were collected from the child, his parents and two sisters. Following the extraction of DNA, high-throughput sequencing was carried out to screen for genetic variant associated with the disease. Candidate variant was verified by Sanger sequencing of his family members. RESULTS: The main clinical features of the proband have…

Loss-of-Function SOX10 Mutation in a Patient with Kallmann Syndrome, Hearing Loss, and Iris Hypopigmentation.

BACKGROUND: Kallmann syndrome (KS) is a clinically and genetically heterogeneous disorder consisting of hypogonadotropic hypogonadism and anosmia. KS is occasionally associated with deafness. Recently, mutations in SOX10, a well-known causative gene of Waardenburg syndrome (WS) characterized by deafness, skin/hair/iris hypopigmentation, Hirschsprung disease, and neurological defects, have been identified in a few patients with KS and deafness. However, the current understanding of the clinical consequences of SOX10 mutations remains fragmentary. CASE REPORT: A Japanese male patient presented …

Oral zinc sulphate causes murine hair hypopigmentation and is a potent inhibitor of eumelanogenesis in vivo.

BACKGROUND: C57BL/6 a/a mice have been widely used to study melanogenesis, including in electron paramagnetic resonance (EPR) studies. Zinc cations modulate melanogenesis, but the net effect of Zn2+ in vivo is unclear, as the reported effects of Zn2+ on melanogenesis are ambiguous: zinc inhibits tyrosinase and glutathione reductase in vitro, but also enhances the activity of dopachrome tautomerase (tyrosinase-related protein-2) and has agonistic effects on melanocortin receptor signalling. OBJECTIVES: To determine in a C57BL/6 a/a murine pilot study whether excess zinc ions inhibit, enhance o

A new form of complicated hereditary spastic paraplegia with cataracts, atretic ear canals and hypopigmentation.

A 16-year-old Hispanic boy born of consanguineous parents is described as having a history of cataracts, progressive lower-extremity spasticity and atrophy starting at 4 years of age, atretic ear canals with hearing dysfunction and diffuse patchy cutaneous hypopigmented areas. Clinical examination showed the typical signs of spastic paraplegia with increased tone, hyperreflexia, muscle atrophy and contractures. Sensation, autonomic and cerebellar functions were not disturbed. Neuroimaging studies were normal. Laboratory findings did not support a diagnosis of metabolic disturbance or infectiou

Clinical and Genetic Characterization of 8 Patients with Syndromic Patterned Cutaneous Hypopigmentation: A Descriptive Study.

BACKGROUND: Patterned cutaneous hypopigmentation (PCH) can be associated with an array of extracutaneous findings. OBJECTIVE: Determine clinical and genetic characteristics of patients with PCH with extracutaneous involvement. METHOD: Thirty patients were identified, and eight patients with neurological involvement agreed to participate in this study. They were reassessed to collect clinical anomalies associated with their PCH. Exome sequencing was performed on patient blood and lesional skin biopsies as well as blood from both parents when available. Array comparative genomic hybridization …

Fundus hypopigmentation and choroidal thinning associated with tebentafusp therapy: report of a case and literature review.

PURPOSE: To describe a novel case of progressive fundus hypopigmentation and choroidal thinning associated with tebentafusp monotherapy for metastatic uveal melanoma. METHODS: Observational case report and review of the literature. CASE PRESENTATION: A 69-year-old male was diagnosed with a choroidal melanoma, measuring 13.3 mm in diameter and 5.8 mm in thickness, in the left eye. Seven years after iodine-125 plaque brachytherapy, systemic imaging identified a solitary liver metastasis, which was laparoscopically resected. About one year later, two new liver metastases were detected. The pati…

Steroid-Induced Hypopigmentation of the Dorsal Hand: Clinicopathologic Correlation With Distinct Melan-A/SOX10 Findings.

Steroid-induced hypopigmentation (SIH) is an uncommon and often underappreciated complication of local corticosteroid therapy, especially in visible sites such as the dorsal hand. We report a case of a 51-year-old woman who developed an elongated hypopigmented patch following intralesional triamcinolone injection for hand pain. The lesion appeared several weeks after injection, was sharply defined, and tracked the extensor tendons without associated surface changes or significant atrophy. Histologic examination revealed a largely unremarkable epidermis with subtle atrophic changes and preserve…

The toxicity of 4-tert-butylphenol in early development of zebrafish: morphological abnormality, cardiotoxicity, and hypopigmentation.

Endocrine disrupting effects of 4-tert-butylphenol (4-t-BP) are well described in literature. However, the evidence regarding developmental … sion of genes related to cardiac development and melanin synthesis, resulting in cardiotoxicity and hypopigmentation. 4-t-BP also caused oxidative stress, and initiated apoptosis through p53-bcl-2/bax-capase3 pathway. Integrative biomarker response analysis showed time- and dose-dependent effects of 4-t-BP on oxidative damage and developmental toxicity in zebrafis …
