Rabies virus (RABV) is an infectious and neurotropic pathogen that causes rabies and infects humans and almost all warm-blooded animals, posing a great threat to people and public safety. It is well known that innate immunity is the critical first line of host defense against viral infection. It monitors the invading pathogens by recognizing the pathogen-associated molecular patterns and danger-associated molecular patterns through pattern-recognition receptors, leading to the production of type I interferons (IFNα/β), inflammatory cytokines, and chemokines, or the activation of autophagy or a

INTRODUCTION: The ocular surface is a frontline mucosal immune barrier that protects the eye from pathogens while preserving immune tolerance essential for vision. Breakdown of this innate immune homeostasis can trigger chronic ocular surface inflammation, exemplified by Stevens-Johnson syndrome/toxic epidermal necrolysis with severe ocular complications (SJS/TEN with SOC) and atopic keratoconjunctivitis (AKC).
AREAS COVERED: The review summarizes current knowledge of ocular surface innate immunity, focusing on pattern-recognition receptors (TLR3, RIG-I, MDA5) and downstream interferon-stimul

When microorganisms invade a host, the innate immune system first recognizes the pathogen-associated molecular patterns of these microorganisms through pattern recognition receptors (PRRs). Toll-like receptors (TLRs) are known transmembrane PRRs existing in both invertebrates and vertebrates. Upon ligand recognition, TLRs initiate a cascade of signaling events; promote the pro-inflammatory cytokine, type I interferon, and chemokine expression; and play an essential role in the modulation of the host's innate and adaptive immunity. Therefore, it is of great significance to improve our understan

Human cytomegalovirus (CMV) and BK polyomavirus are archetypal latent DNA viruses that establish lifelong persistence and cause severe disease upon reactivation in immunocompromised hosts. Although they differ markedly in virion architecture, genome size, and coding capacity, both viruses converge on a shared biological principle: durable persistence is achieved through sustained immune invisibility rather than maximal replication. CMV is a large, enveloped double-stranded DNA (dsDNA) virus whose multilayered structure supports the expression of numerous immunomodulatory proteins that disrupt

Pneumonia is caused by both viral and bacterial pathogens and is responsible for a significant health burden in the Unites States. The innate immune system is the human body's first line of defense against these pathogens. The recognition of invading pathogens via pattern recognition receptors leads to proinflammatory cytokine and chemokine production, followed by recruitment and activation of effector immune cells. The nonspecific inflammatory nature of the innate immune response can result in immunopathology that is detrimental to the host. In this review, we focus on one class of pattern re

Defences against the bacteria that usually infect the endometrium of postpartum cattle are impaired when there is metabolic energy stress, leading to endometritis and infertility. The endometrial response to bacteria depends on innate immunity, with recognition of pathogen-associated molecular patterns stimulating inflammation, characterised by secretion of interleukin (IL)-1β, IL-6 and IL-8. How metabolic stress impacts tissue responses to pathogens is unclear, but integration of energy metabolism and innate immunity means that stressing one system might affect the other. Here we tested the h

This chapter underscores the pivotal role of immune receptors, particularly Toll-like receptors (TLRs) and NOD-like receptors (NLRs), in the intricate interplay between metabolic diseases and their associated cardiovascular comorbidities. Their involvement extends beyond mere initiation, profoundly influencing the progression of these complex conditions. The recognition of TLRs and NLRs as promising therapeutic targets is a significant advancement, offering potential avenues for intervention in both metabolic disorders and their cardiovascular sequels. The significance of these pattern recognit

In cattle, innate immunity plays a key role in the crosstalk between sperm, egg, and uterus, which is critical for the success of assisted reproductive technologies (ARTs). This review highlights Toll-like receptor 2 (TLR2) as a physiological sensor that modulates these interactions. Following artificial insemination, TLR2-mediated sperm-uterine interactions initiate a controlled immune response. This response is essential for clearing excess sperm and preparing the endometrium for embryo implantation. Similar to sperm, the egg yolk-based semen extender could also activate endometrial TLR2, le

BACKGROUND: Eimeria tenella, one of the most virulent causative agents of coccidiosis, specifically colonizes the cecum and imposes substantial economic losses on the poultry industry. Toll-like receptors (TLRs), key mediators of the innate immune response, initiate rapid defense against invading parasites. However, their roles during E. tenella infection remain poorly defined.
METHODS: In this study, coccidiosis infection parameters (cecal lesions, oocyst output, and clinical signs) were monitored, and the expression profiles of 10 chicken TLRs (chTLRs) and related immune genes in the cecum

Atlantic cod possess a unique immune system due to the loss of the MHC II antigen-presenting system and an extreme expansion of the MHC class I repertoire. Additional modifications are demonstrated within the innate immune system with losses and expansions of Toll-like receptor (TLR) genes. While Atlantic cod has lost its mammalian orthologs for plasma membrane localized TLRs, such as TLR1/6 and TLR2, it has expanded its intracellular TLRs including TLR7, -8, and -9. It has been hypothesized that teleost-specific TLR1-family related members TLR14 and TLR25 localize at the plasma membrane with

UNLABELLED: Hepatitis B virus (HBV) infects hepatocytes specifically and causes immune-mediated liver damage. How HBV interacts with the innate immunity at the early phase of infection, either with hepatocytes or other cells in the liver, remains controversial. To address this question, we utilized various human cell-culture models and humanized Alb-uPA/SCID mice. All these models were unable to mount an interferon (IFN) response despite robust HBV replication. To elucidate the mechanisms involved in the lack of IFN response, we examined whether HBV actively inhibits innate immune functions of

Platelets are chief cells in hemostasis. Apart from their hemostatic roles, platelets are major inflammatory effector cells that can influence both innate and adaptive immune responses. Activated platelets have thromboinflammatory functions linking hemostatic and immune responses in several physiological and pathological conditions. Among many ways in which platelets exert these functions, platelet expression of pattern recognition receptors (PRRs), including TLR, Nod-like receptor, and C-type lectin receptor families, plays major roles in sensing and responding to pathogen-associated or damag

Innate immunity is considered the first line of defense against microbial invasion, and its dysregulation can increase the susceptibility of hosts to infections by invading pathogens. Host cells rely on pattern recognition receptors (PRRs) to recognize invading pathogens and initiate protective innate immune responses. Toll-like receptor 2 (TLR2) is believed to be among the most important Toll-like receptors for defense against mycobacterial infection. TLR2 has been reported to have very broad functions in infectious diseases and also in other diseases, such as chronic and acute inflammatory d

Aspergillus moulds exist ubiquitously as spores that are inhaled in large numbers daily. Whilst most are removed by anatomical barriers, disease may occur in certain circumstances. Depending on the underlying state of the human immune system, clinical consequences can ensue ranging from an excessive immune response during allergic bronchopulmonary aspergillosis to the formation of an aspergilloma in the immunocompetent state. The severest infections occur in those who are immunocompromised where invasive pulmonary aspergillosis results in high mortality rates. The diagnosis of Aspergillus-asso

Activation of cytosolic RNA and DNA sensing pathways that normally recognize viral nucleic acids triggers the expression and release of type I interferons (IFN-I). This release drives the expression of interferon-stimulated genes (ISGs) that activate acute inflammation and counteract viral replication at various stages. Recent studies have highlighted how interferon-stimulated gene 15 (ISG15), a small, ubiquitin-like modifier, regulates cytosolic RNA and DNA sensing pathway proteins to control the antiviral response and inflammation. ISG15 is a multifunctional protein that acts as a secreted c

BACKGROUND: Airway epithelial cells not only constitute a physical barrier, but also the first line of defence against airborne pathogens. At the same time, they are constantly exposed to reactive oxygen species. Therefore, airway epithelia cells have to possess a sophisticated innate immune system and a molecular armamentarium to detoxify reactive oxygen species. It has become apparent that deregulation of epithelial innate immunity is a major reason for the development of chronic inflammatory lung diseases. To elucidate the molecular architecture of the innate immune system of airway epithel

The Gram-positive facultative intracellular bacterium Listeria monocytogenes is a model pathogen for elucidating important mechanisms of the immune response. Infection of mice with a sub-lethal dose of bacteria generates highly reproducible innate and adaptive immune responses, resulting in clearance of the bacteria and resistance to subsequent L. monocytogenes infection. Both the innate and adaptive immune systems are crucial to the recognition and elimination of this pathogen from the host.

BACKGROUND & AIMS: Most viruses are detected at early stages of cell infection and induce an innate immune response mediated by production of interferons (IFNs). IFNs induce expression of hundreds of IFN-stimulated genes (ISGs). Infection of chimpanzees with hepatitis C virus, but not hepatitis B virus (HBV), induces ISG expression in the liver. HBV might not induce an innate immune response because it is not detected by pattern recognition receptors (the stealth properties of HBV) or because HBV suppresses IFN production or signaling despite detection by pattern recognition receptors. We stud

INTRODUCTION: Spontaneous control of HIV-1 replication in the absence of anti-retroviral therapy (ART) naturally occurs in a small proportion of HIV-1-infected individuals known as elite controllers (EC), likely as a result of improved innate and adaptive immune mechanisms. Previous studies suggest that enhanced cytosolic immune recognition of HIV-1 reverse transcripts in conventional dendritic cells (mDC) from EC enables effective induction of antiviral effector T cell responses. However, the specific molecular circuits responsible for such improved innate recognition of HIV-1 in mDC from the

Myelodysplastic neoplasms (MDS) are clonal hematologic disorders characterized by ineffective hematopoiesis and a variable risk of progression to acute myeloid leukemia (AML). Despite growing recognition of the role of innate immunity in MDS pathogenesis, the precise mechanisms remain unclear. In this study, we analyzed bone marrow CD34+ expression data from 183 MDS patients to investigate the impact of the Toll-like receptor (TLR) pathway on disease progression. Six key innate immunity genes (IRAK1, IRAK2, IRAK4, MYD88, TRAF6, and NFKB1) were used to define two distinct immune clusters: a hyp
