The impact of insecticide-resistance on control of vectors and vector-borne diseases.

A questionnaire inquiring into the nature of schemes for the insecticidal control of disease vectors, the development of resistance in these vectors, and the effect of any such resistance on their control and on the extent of disease was sent to more than 100 health authorities throughout the world. The replies to the questionnaire are summarized in this paper.Until recently, the use of insecticides in public health has been largely based on three organochlorine compounds-DDT, HCH and dieldrin. However, in some countries resistance to these has now severely affected control both of many insect

Ruvbl1 silencing affects reproduction of the corn planthopper, Peregrinus maidis.

Ruvbl1 (also known as TIP49, Pontin) encodes an ATPase of the AAA+ protein superfamily involved in several cellular functions, including chromatin remodeling, control of transcription, and cellular development (motility, growth, and proliferation). While its role has been well established in model organisms including vertebrates and invertebrates (e.g. mice, Xenopus and Drosophila), putative functions of Ruvbl1 in non-model insect pests have not been addressed. To exploit Ruvbl1 as a potential target gene for applications in insect control, we used an in-vivo RNA interference (RNAi) approach t…

RNA Interference Is Enhanced by Knockdown of double-stranded RNases in the Yellow Fever Mosquito Aedes Aegypti.

RNA interference (RNAi) techniques are being developed for a range of pest insect control technologies, including the sterile insect technique (SIT) and double-stranded RNA (dsRNA)-based insecticides. In SIT applications, where >99% of the released males should be sterile to meet industry standards, the efficiency of RNAi will need to be improved for many insect species if this technology is to be adopted. Endogenous dsRNases can impede dsRNA delivery in some insects, and, here, we investigated whether dsRNases in the midgut could limit RNAi efficacy in the mosquito Aedes aegypti. Ten putative…

Field performance of sterile male mosquitoes released from an uncrewed aerial vehicle.

Genetic control methods of mosquito vectors of malaria, dengue, yellow fever, and Zika are becoming increasingly popular due to the limitations of other techniques such as the use of insecticides. The sterile insect technique is an effective genetic control method to manage insect populations. However, it is crucial to release sterile mosquitoes by air to ensure homogeneous coverage, especially in large areas. Here, we report a fully automated adult mosquito release system operated from an uncrewed aerial vehicle or drone. Our system, developed and tested in Brazil, enabled a homogeneous dispe

Effect of volatile constituents from Securidaca longepedunculata on insect pests of stored grain.

Securidaca longepedunculata Fers (Polygalaceae) is commonly used as a traditional medicine in many parts of Africa as well as against a number of invertebrate pests, including insects infesting stored grain. The present study showed that S. longepedunculata root powder, its methanol extract, and the main volatile component, methyl salicylate, exhibit repellent and toxic properties to Sitophilus zeamais adults. Adult S. zeamais that were given a choice between untreated maize and maize treated with root powder, extract, or synthetic methyl salicylate in a four-way choice olfactometer significan

Insect-resistant tobacco plants expressing insect-specific neurotoxin AaIT.

The recombinant plant expression vector pNGY-2 with designed and synthesized AaIT gene had been constructed. The AaIT gene was fused behind the sequence of TMV and inserted into expression vector under the control of two linked 35s promoters. The recombinant plasmid pNGY-2 was transferred into tobacco NC89 by agrobacterium-mediated transfer system. The GUS activity analysis and Southern blotting of regenerated plants indicated that AaIT gene had been integrated into tobacco genome. Insect bioassays showed that some transgenic plants had notable insect-resistant activity.

Toxicity of methyl bromide and other gaseous insecticides to Triatoma infestans.

Cuticular penetration of lipid soluble substances in Triatoma infestans is very poor (less than 1% in 24 hr), while the respiratory route is particularly efficient (28% in 5 min). Accordingly, several noxious gases killed the insect at short periods of time. CH3Br and SO2 were particularly effective and they also evidenced ovicide action. With the former compound this effect is also observed at low concentrations. The CH3Br hydrolysis products or its metabolites were not similarly effective, while other methylating agent, diazomethane, was very active. The irreversible binding of 14CH3Br to pr

Captures of Boll Weevils (Coleoptera: Curculionidae) in Relation to Trap Orientation and Distance From Brush Lines.

Eradication programs for the boll weevil (Anthonomus grandis grandis Boheman) rely on pheromone-baited traps to trigger insecticide treatments and monitor program progress. A key objective of monitoring in these programs is the timely detection of incipient weevil populations to limit or prevent re-infestation. Therefore, improvements in the effectiveness of trapping would enhance efforts to achieve and maintain eradication. Association of pheromone traps with woodlots and other prominent vegetation are reported to increase captures of weevils, but the spatial scale over which this effect occu

New measures of insecticidal efficacy and safety obtained with the 39K promoter of a recombinant baculovirus.

Baculoviruses are orally infectious to insects and considered to be natural insecticides. To enhance their speed-of-kill these viruses were engineered to express arthropod neurotoxins under the control of various strong promoters. Although this strategy proved to be efficient, it raised recently concerns about safety. We analyzed the speed-of-kill and safety of Autographa californica multiple nucleopolyhedrovirus expressing the insecticidal scorpion neurotoxin AaIT and found that the mortality of Helicoverpa armigera larvae was enhanced significantly when the expression was controlled by the b

Paederus Dermatitis Outbreak in Addis Ababa, Ethiopia: A Case-Control Study.

BACKGROUND: Paederus dermatitis is an irritant contact dermatitis caused by crushing insects of the genus Paederus, which releases a vesicant toxin called pederin. On July 28, 2018, the district health office received a report of cases with erythema, itching, and burning after contact with the Paederus insect. In response, we investigated the outbreak intending to describe, confirm, and identify the risk factors associated with the country's first reported outbreak. METHODS: A community-based unmatched case-control study was conducted from August 10 to 22, 2018. Two hundred twenty-five (75 ca

Knowledge, attitudes, and behaviors regarding tick-borne disease prevention in endemic areas.

As part of a TickNET collaboration we evaluated the knowledge, attitudes, and behaviors related to tick-borne disease (TBD) prevention among persons living in endemic areas of Connecticut (CT) and Maryland (MD). Up-to-date information on the use of various prevention methods, as well as attitudes toward available and potential prevention options, is critical for effective promotion of recommended behaviors. During 2016-2017, printed invitations were mailed via the post office to 27,029 households requesting participation in an online survey regarding knowledge of TBD, risk perceptions, and pre

Temperature-dependence of life history in an edible cricket: Implications for optimising mass-rearing.

Optimisation of life history and organismal performance underlies success in insect mass-rearing. Rearing schedules, resource use and production yield depend on many aspects of insect fitness and performance within and across generations, such as growth, development, longevity, and fecundity, which are all temperature dependent. Despite this general understanding, we often lack species-specific information needed to make informed decisions about manipulating rearing temperatures to optimise insect growth and development. Here, we characterise the effects of rearing temperature on nymph to adul

Transcriptomic Insights into the Insect Immune Response to Nematode Infection.

Insects in nature interact with a wide variety of microbial enemies including nematodes. These include entomopathogenic nematodes that contain mutualistic bacteria and together are able to infect a broad range of insects in order to complete their life cycle and multiply, filarial nematodes which are vectored by mosquitoes, and other parasitic nematodes. Entomopathogenic nematodes are commonly used in biological control practices and they form excellent research tools for understanding the genetic and functional bases of nematode pathogenicity and insect anti-nematode immunity. In addition, cl

Body condition constrains immune function in field populations of female Australian plague locust Chortoicetes terminifera.

The insect innate immune system comprises both humoral and cellular defence responses. In the laboratory, the insect immune system is well characterized. In the field, however, little is known about the role of constitutive insect immune function and how it varies within and between populations. Laboratory studies suggest that host nutrition has significant impact upon insect immune function. Thus, the rationale for this study was to sample natural populations of the Australian Plague Locust Chortoicetes terminifera to establish whether locust body condition (as determined by protein and lipid

Dispersal and survival of gamma-irradiated Culex quinquefasciatus: Implications for sterile insect technique applications.

BACKGROUND: Culex quinquefasciatus is a major vector of filariasis and other mosquito-borne diseases. The sterile insect technique (SIT) has been widely used to suppress mosquito populations, but its effectiveness depends on the dispersal, survival, and competitiveness of sterile males. AIM: This study evaluated the dispersal range, survival rate, and recapture success of gamma-irradiated Cx. quinquefasciatus under field conditions. METHODS: A mark-release-recapture (MRR) experiment was conducted using sterile male and female Cx. quinquefasciatus. Two release events were conducted, and recap

Evaluation of the house fly Musca domestica as a mechanical vector for an anthrax.

Anthrax is a disease of human beings and animals caused by the encapsulated, spore-forming, Bacillus anthracis. The potential role of insects in the spread of B. anthracis to humans and domestic animals during an anthrax outbreak has been confirmed by many studies. Among insect vectors, the house fly Musca domestica is considered a potential agent for disease transmission. In this study, laboratory-bred specimens of Musca domestica were infected by feeding on anthrax-infected rabbit carcass or anthrax contaminated blood, and the presence of anthrax spores in their spots (faeces and vomitus) wa

Investigating impact of slash and clear vector control strategy on blackfly population and onchocerciasis transmission in a hotspot in Nigeria.

BACKGROUND: The Edo-Ondo border remains a hotspot for onchocerciasis transmission and has been designated as a special intervention zone by Nigeria's National Onchocerciasis Elimination Committee because of its persistent transmission and high blackfly biting rates. This study evaluated the effectiveness of the "slash-and-clear" vector control strategy and annual ivermectin distribution to interrupt transmission of Onchocerca volvulus in the area. METHODS: This study was implemented in two phases. Phase 1 (July-August 2023) involved slash-and-clear in four communities along the Ose River and

Innovative applications for insect viruses: towards insecticide sensitization.

The effective management of emerging insect-borne disease is dependent on the use of safe and efficacious chemical insecticides. Given the inherent ability of insects to develop resistance, it is essential to propose innovative strategies because insecticides remain the most important element of integrated approaches to vector control. Recently, intracellular phosphorylation and dephosphorylation of membrane receptors and ion channels targeted by insecticides have been described as new processes for increasing the sensitivity of insecticides. An efficient method might be to infect host insects
