Application of spinosad increases the susceptibility of insecticide-resistant Alphitobius diaperinus (Coleoptera: Tenebrionidae) to pyrethroids.

The effect of spinosad exposure on the susceptibility of pyrethroid- and organophosphate-resistant populations of lesser mealworm, Alphitobius diaperinus (Panzer) (Coleoptera: Tenebrionidae), to insecticides was investigated in broiler house farm and laboratory studies. A field pyrethroid- and organophosphate-resistant population showed a 3.6-fold increase in susceptibility to gamma-cyhalothrin following spinosad treatment. Overall, cyfluthrin- and fenitrothion-resistant field populations were more susceptible to these insecticides following spinosad treatments, but populations that were not r

Differential expression of salivary proteins between susceptible and insecticide-resistant mosquitoes of Culex quinquefasciatus.

BACKGROUND: The Culex quinquefasciatus mosquito, a major pest and vector of filariasis and arboviruses in the tropics, has developed multiple resistance mechanisms to the main insecticide classes currently available in public health. Among them, the insensitive acetylcholinesterase (ace-1(R) allele) is widespread worldwide and confers cross-resistance to organophosphates and carbamates. Fortunately, in an insecticide-free environment, this mutation is associated with a severe genetic cost that can affect various life history traits. Salivary proteins are directly involved in human-vector conta

Distribution of Anopheles gambiae s.l siblings, insecticide resistance and prevalence of markers of resistance (Kdr & Ace-1) in Edea and Buea: forest region of Cameroon.

BACKGROUND: The complexity and heterogeneity of malaria transmission in Cameroon is enhanced by the different eco-systems and topology which enhance the proliferation of Anopheline mosquitoes. Though long-lasting insecticide-treated nets have been massively distributed across the country, malaria still remains a significant public health concern, with innate and adaptive resistance mechanisms exploited by malaria vectors against different insecticides; the micro-ecological variations in Cameroon could be playing a viral role in the expression of essential insecticide resistance genes in malari…

Selection for resistance to pirimiphos-methyl, permethrin and spinosad in a field strain of Sitophilus oryzae: resistance risk assessment, cross-resistance potential and synergism of insecticides.

Indiscriminate use of insecticides in food storage facilities for controlling insect pests has deleterious effects on the environment and non-targeted organisms in the premises. Continuous use of insecticides may result in resistance development in insect pests, which compel the stakeholders to increase the dosage of insecticides to manage resistant insect pests. The increased dosage of insecticides ultimately may result in contamination of stored food stuff that affects human health. The present study was planned to generate data that will be helpful to delay resistance development and to red

Insecticide resistance monitoring of Aphis spiraecola (Hemiptera: Aphididae) to neonicotinoids and abamectin in a major apple-growing region.

Aphis spiraecola is a significant pest of fruit trees worldwide, causing substantial yield and economic losses in apple production. The control of A. spiraecola primarily depends on chemical pesticides; however, long-term field monitoring of insecticide resistance is lacking. This study assessed the susceptibility of a laboratory-susceptible strain (SS) of A. spiraecola to abamectin, imidacloprid, thiamethoxam, and acetamiprid. The LC50 values of A. spiraecola to abamectin, imidacloprid, thiamethoxam, and acetamiprid were 0.13, 1.44, 1.76, and 1.19 mg L-1, respectively. The resistance levels o…

Malaria vector control in sub-Saharan Africa: complex trade-offs to combat the growing threat of insecticide resistance.

Mass distribution of insecticide-treated nets (ITNs) has been a key factor in reducing malaria cases and deaths in sub-Saharan Africa. A shortcoming has been the over-reliance on pyrethroid insecticides, with more than 2·13 billion pyrethroid ITNs (PY ITNs) distributed in the past two decades, leading to widespread pyrethroid resistance. Progressive changes are occurring, with increased deployment of more effective pyrethroid-chlorfenapyr (PY-CFP) or pyrethroid-piperonyl butoxide (PY-PBO) ITNs in areas of pyrethroid resistance. In 2023, PY-PBO ITNs accounted for 58% of all ITNs shipped to sub-

Insecticide Resistance of Xenopsylla cheopis in Madagascar: Revision of Diagnostic Doses for Bioassay and Exploration of Biochemical Mechanisms.

The Oriental rat flea, Xenopsylla cheopis, is known worldwide as an efficient plague vector, including in Madagascar, where the disease remains a public health concern. Chemical control is the primary response method against X. cheopis in Madagascar. Previous bioassays focusing on different flea populations from Madagascar showed phenotypic resistance to various insecticides, including deltamethrin and fenitrothion, which, respectively, represent the previous and current chemicals for flea vector control. Despite apparent insecticide resistance, the associated mechanisms of this resistance rem…

Vectorial competence, insecticide resistance in Anopheles funestus and operational implications for malaria vector control strategies in Benin Republic.

The primary reason for the failure of malaria vector control across endemic regions is the widespread insecticide resistance observed in Anopheles vectors. The most dominant African vectors of malaria parasites are Anopheles gambiae and Anopheles funestus mosquitoes. These species often exhibit divergent behaviours and adaptive changes underscoring the importance of deploying active and effective measures in their control. Unlike An. gambiae, An. funestus mosquitoes are poorly studied in Benin Republic. However, recent reports indicated that An. funestus can adapt and colonize various ecologic…

Toxicity of Ocimum americanum L. extracts against Aedes aegypti larvae (Diptera: Culicidae) and their impact on insecticide resistance.

Mosquitoes are a key threat to millions of people worldwide. They spread the pathogens that cause deadly diseases among humans and animals. Synthetic pesticides are the best agents to control mosquitoes, but they cause several problems for the environment as well as public health. Continuous usage of commonly available insecticides develops multiple resistances among pests. In search of alternatives to synthetic pesticides, botanicals could be one of the best alternatives to control mosquitoes. The present study explores the insecticidal activity of Ocimum americanum against Aedes aegypti larv

Co-variation of insecticide resistance and host-microbe gene expression in geographically distant populations of German cockroaches (Blattodea: Blattellidae).

Insecticides can greatly limit the impacts of cockroaches on indoor environment and human health. Insecticide resistance, however, reduces the efficacy of insecticides and increases insecticide use by both residents and pest control professionals. This study examined the relationship between insecticide cross-resistance and associated gene expression across nine different German cockroach populations from the eastern U.S. Using vial bioassays, we identified significantly correlated resistance between three insecticides from different classes (indoxacarb, lambda-cyhalothrin, fipronil), with pot…

Insecticide susceptibility and knockdown resistance (Kdr) mutations (F1534C and V1016I) in Aedes aegypti populations from Dar es Salaam, Tanzania.

BACKGROUND: Dengue, Zika and chikungunya are Aedes-borne arboviruses that pose a major global public health threat. In the absence of effective treatments or universally available vaccines, disease control in sub-Saharan Africa relies primarily on insecticidal vector control. However, sustained insecticide application imposes selection pressure for insecticide resistance that compromises intervention effectiveness. This study assessed the insecticide resistance profile and knockdown resistance of Aedes aegypti populations in Dar es Salaam, Tanzania. METHODS: Phenotypic resistance of Ae. aegyp…

Biting behaviour, spatio-temporal dynamics, and the insecticide resistance status of malaria vectors in different ecological zones in Ghana.

BACKGROUND: A significant decrease in malaria morbidity and mortality has been attained using long-lasting insecticide-treated nets and indoor residual spraying. Selective pressure from these control methods influences changes in vector bionomics and behavioural pattern. There is a need to understand how insecticide resistance drives behavioural changes within vector species. This study aimed to determine the spatio-temporal dynamics and biting behaviour of malaria vectors in different ecological zones in Ghana in an era of high insecticide use for public health vector control. METHODS: Adult…

Two-step method for rapid isolation of genomic DNA and validation of R81T insecticide resistance mutation in Myzus persicae.

Isolation and amplification of nucleic acid (DNA) is considered a vital and potent instrument in molecular biological research. However, its functioning outside of a laboratory setting is difficult because of complex procedures that demand expert personnel and expensive equipment in addition to the fulfillment of several additional requirements. DNA isolation from minute insects is sometimes difficult, making diagnostic and genotyping procedures problematic. Thus, the current work offers a high-throughput, cost-effective, straightforward, and faster approach for isolating DNA from the aphid My
