The majority of polydiacetylenes (PDAs) described to date display thermochromic transitions above room temperature. By following a strategy that employs headgroups that do not participate in strong interactions, we have designed and prepared a liquid diacetylene (DA) monomer that solidifies at a temperature near 0 °C. A series of N'-phenyl-N-hydroxyureas has been prepared by reacting hydroxylamine with aromatic isocyanates. These compounds were investigated as inhibitors of human carbonic anhydrases (hCAs, EC 4.2.1.1), considering four physiologically relevant isoforms, the cytosolic isoforms hCA I and II, and tumor associated, transmembrane isoforms hCA IX and XII. After the UV irradiation of suspensions of barbiturate sodium salts in paraffin oil the IR spectra revealed two IR absorption bands at ca. 2170 and 2250 cm-1. The effects of different conditions of the photochemical process and the addition of nucleophilic reagents after irradiation on the intensity of these bands were studied. The origin of 2170 cm-1 band was interpreted in terms of the isocyanate intermediate which is formed after the pyrimidine ring opening.

A novel series of twelve aromatic bis-ureido-substituted benzenesulfonamides was synthesised by conjugation of aromatic aminobenzenesulfonamides with aromatic bis-isocyanates. The obtained bis-ureido-substituted derivatives were tested against four selected human carbonic anhydrase isoforms (hCA I, hCA II, hCA IX and hCA XII). A sensitive personal passive dosimeter, based on an enzymatic reaction, was developed to monitor exposure to airborne concentrations of hexamethylene diisocyanate (HDI), used in the production of polyurethanes. Dosimeters were prepared by adsorption of butyryl- or acetylcholinesterase on sheets of polystyrene. The automation of the azapeptide submonomer synthesis procedure is reported. All steps for on-resin aza-amino acid synthesis, including semicarbazone incorporation, alkylation, deprotection, and semicarbazide acylation were adapted for use on a commercially available peptide synthesizer. Key requirements for success included the use of a stable isocyanate precursor for the semicarbazone installation, the use of stable reagents in DMF, and elimination of cooling steps. The synthesis of > 30 azapeptides was accomplished using our optimized automated submonomer azapeptide synthesis procedure.

Using dichloromethane (DCM) as the C1 feedstock, the first Pd(0)-catalyzed synthesis of aryl-methylene ether motifs from hydrobenzoxazoles (hydrothiazoles) has been reported. This protocol has exhibited an ample substrate scope (38 examples) and wide functional group tolerance. Aryl-methylene ether adducts have been successfully engaged in coupling with isocyanate to provide bis-iminoisoindolinones. In this reaction, DCM has served as a twofold electrophile for reaction with hydrobenzoxazoles via double C-Cl bond cleavage.
