We investigated the mechanisms of the direct positive inotropic effect (PIE) of helenalin, a sesquiterpene lactone isolated from the plant genus Helenium of the family Compositae in guinea pig ventricular myocardium. Ultraviolet difference spectroscopy has been used to study the binding of the transition state analog saccharo-1,4-lactone to purified rat preputial gland beta-glucuronidase. At pH 4.5 (the pH optimum), the inhibitor induces a difference spectrum indicative of a change in the environment of tryptophyl residues. To investigate the frequency of Compositae sensitivity, the recently-developed sesquiterpene lactone mix (SL mix) was included in the standard patch test series. Patients with positive reactions to this or patients suspected of having a Compositae allergy were supplementarily tested with a Compositae mix, consisting of ether extracts of 5 European Compositae plants.
