Quantifying the interaction of the C-terminal regions of polycystin-2 and polycystin-1 attached to a lipid bilayer by means of QCM.

The pkd1 and pkd2 genes encode for the proteins polycystin-1 (PC1) and polycystin-2 (PC2). These genes are mutated in patients diagnosed with autosomal dominant polycystic kidney disease. PC1 and PC2 interact via their C-terminal, cytosolic regions, which is an essential step in the regulation of cell proliferation and differentiation. Here, we developed an assay that allowed us to quantitatively monitor the interaction of the C-terminal region of PC1 (cPC1) with that of PC2 (cPC2) to be able to answer the question of how Ca(2+) influences the PC1/PC2 complex formation. By means of the quartz

Geometry of hydrogen bonds formed by lipid bilayer nitroxide probes: a high-frequency pulsed ENDOR/EPR study.

Solvent effects on magnetic parameters of nitroxide spin labels in combination with side-directed spin labeling EPR methods provide very useful means for elucidating polarity profiles in lipid bilayers and mapping local electrostatic effects in complex biomolecular systems. One major contributor to these solvent effects is the hydrogen bonds that could be formed between the nitroxide moiety and water and/or the available hydroxyl groups. Here, formation of hydrogen bonds between a lipid bilayer spin probe -5-doxyl stearic acid, 5DSA – and hydrogen bond donors has been studied using high-freque…

Electrochemical, spectroelectrochemical and molecular dynamics investigation of doxorubicin interactions with nucleolipid-containing floating lipid bilayer on gold electrodes.

Floating lipid bilayers (fBLMs) of 1,2-dipalmitoyl-sn-glycero-3-cytidine diphosphate: 1,2-dipalmitoyl-sn-glycero-3-phosphocholine (DG-CDP: DPPC) (7:3) assembled on β-D-thioglucose-modified Au electrodes were used as biomimetic systems to investigate, at the molecular level, the interaction of doxorubicin (DOX) with nucleolipid-containing membranes under electrochemical control. Pseudocapacitance and electrochemical impedance spectroscopy (EIS) reveal that the presence of DOX decreases membrane capacitance and increases defect resistance, indicating reduced ionic permeability. ATR-SEIRAS measur…

Interaction Profiles and Stability of Rigid and Polymer-Tethered Lipid Bilayer Models at Highly Charged and Highly Adhesive Contacts.

Understanding interaction force versus distance profiles of supported lipid bilayers (SLBs) is relevant to a number of areas, which rely on these model systems, including, e.g., characterization of ligand/receptor interactions or bacterial adhesion. Here, the stability of 4 different SLB architectures was compared using the surface forces apparatus (SFA) and atomic force microscopy (AFM). Specifically, the outer envelope of the bilayer systems remained constant as 1,2-dipalmitoyl-sn-glycero-3-phosphocholine (DPPC). The inner layer was varied between DPPC and 1,2-dipalmitoyl-3-trimethylammonium…

Adaptation Independent Modulation of Auditory Hair Cell Mechanotransduction Channel Open Probability Implicates a Role for the Lipid Bilayer.

The auditory system is able to detect movement down to atomic dimensions. This sensitivity comes in part from mechanisms associated with gating of hair cell mechanoelectric transduction (MET) channels. MET channels, located at the tops of stereocilia, are poised to detect tension induced by hair bundle deflection. Hair bundle deflection generates a force by pulling on tip-link proteins connecting adjacent stereocilia. The resting open probability (P(open)) of MET channels determines the linearity and sensitivity to mechanical stimulation. Classically, P(open) is regulated by a calcium-sensitiv

Theoretical design of the cyclic lipopeptide nanotube as a molecular channel in the lipid bilayer, molecular dynamics and quantum mechanics approach.

In this article, cyclic peptides (CP) with lipid substituents were theoretically designed. The dynamical behavior of the CP dimers and the cyclic peptide nanotube (CPNT) without lipid substituents in the solution (water and chloroform) during the 50 ns molecular dynamic (MD) simulations has been investigated. As a result, the CP dimers and CPNT in a non-polar solvent are more stable than in a polar solvent and CPNT is a good container for non-polar small molecules such as chloroform. The effect of the lipid substituents on the CP dimers and CPNT has been investigated in the next stage of our s

Manipulation of a spider peptide toxin alters its affinity for lipid bilayers and potency and selectivity for voltage-gated sodium channel subtype 1.7.

Huwentoxin-IV (HwTx-IV) is a gating modifier peptide toxin from spiders that has weak affinity for the lipid bilayer. As some gating modifier toxins have affinity for model lipid bilayers, a tripartite relationship among gating modifier toxins, voltage-gated ion channels, and the lipid membrane surrounding the channels has been proposed. We previously designed an HwTx-IV analogue (gHwTx-IV) with reduced negative charge and increased hydrophobic surface profile, which displays increased lipid bilayer affinity and in vitro activity at the voltage-gated sodium channel subtype 1.7 (NaV1.7), a chan…

Fluorescence readouts of KL4-induced changes in lipid order and fluidity in surfactant-like bilayers relevant to ARDS and COVID-19 lung injury.

KL4, a 21-residue mimetic of the C-terminus of surfactant protein B, is effective in treating infant respiratory distress syndrome (RDS). It works by reducing alveolar surface tension and improving oxygen exchange. Here, we use steady-state fluorescence measurements to probe how KL4 modulates bilayer order and organization in pulmonary surfactant-like lipid mixtures with varying acyl chain saturation. We show how fluorescence-based methods can distinguish between a more surface-associated and a more deeply inserted mode of interaction of KL4 in phospholipid membranes with varying acyl chain sa

Formation of Tethered Bilayer Lipid Membranes (tBLMs) Using Plasma Membrane Blebs from Human Lung Cancer Cells.

Plasma membrane blebs, or giant plasma membrane vesicles (GPMVs), serve as excellent biomimetics for studying the compositional complexity of native cell membranes. However, current supported membrane models often lack the stability or submembrane reservoir necessary for extended functional studies. This work presents a proof of concept for the formation of tethered bilayer lipid membranes (tBLMs) using blebs derived from human lung cancer cells (A549). We report that successful fusion of blebs into planar tBLMs on gold substrates requires a significantly higher density of tethering anchors (6

Channel Formation by LktA of Mannheimia (Pasteurella) haemolytica in Lipid Bilayer Membranes and Comparison of Channel Properties with Other RTX-Cytolysins.

Cytolysin LktA is one of the major pathogenicity factors of Mannheimia haemolytica (formerly Pasteurella haemolytica) that is the cause of pasteurellosis, also known as shipping fever pneumonia, causing substantial loss of sheep and cattle during transport. LktA belongs to the family of RTX-toxins (Repeats in ToXins) that are produced as pathogenicity factors by a variety of Gram-negative bacteria. Sublytic concentrations of LktA cause inflammatory responses of ovine leukocytes. Higher concentrations result in formation of transmembrane channels in target cells that may cause cell lysis and ap

Single-Channel Electrophysiology Using a Miniaturized Montal-Mueller Planar Lipid Bilayer System for Recording a Synthetic K2P Channel.

The functional reconstitution of ion channels into planar lipid bilayers provides a strategy to investigate channel activity at the single-molecule level. However, conventional Montal-Mueller bilayer systems typically require bulky experimental setups, manual chamber assembly, and relatively large recording volumes, which increase reagent consumption and may introduce additional sources of electrical noise due to extended wiring and external headstages. Here, we present a miniaturized, ready-to-use bilayer platform (BLMchipMM) integrated with a compact amplifier that enables stable single-chan…

Lateral lipid packing governs bilayer solubilization by styrene-maleic acid copolymers: a case study with cardiolipin-containing membranes.

Styrene-maleic acid (SMA) copolymers are powerful tools for the detergent-free solubilization of biological membranes. Yet the influence of specific lipids on SMA activity remains an open question. Here, we examined the effects of the mitochondriaspecific phospholipid cardiolipin on SMA-mediated membrane solubilization and their ability to form SMA-bound nanodiscs. To this end, we prepared a series of model membranes with cardiolipin and other test lipids with comparable surface charge and lateral packing characteristics. Preparation, Conformational Structure, and Proteolytic Activity of Papain Covalently Conjugated to Poly(ethylene glycol)-Tethered Lipid Bilayer Membranes.

Conjugation of enzymes to lipid membranes is a key approach to reconstitute fascinating features of cell organelles and to deduce the nature of membrane-involved biological events. In this work, papain was covalently conjugated via a cross-linker to phospholipid vesicles (liposomes) tethered with poly(ethylene glycol) (PEG) at 25 °C and pH = 7.0, followed by chromatographic purification. The size of the PEG moiety and the type of cross-linker were optimized to obtain PEG-tethered liposome-conjugated papain (liposome-PEG-papain). Slight conformational changes of the membrane-conjugated papain i

Biodegradable lipid bilayer-assisted indocyanine green J- aggregates for photothermal therapy: Formulation, in vitro toxicity and in vivo clearance.

Indocyanine green (ICG) J-aggregates (IJA) are a unique form of aggregation that exhibits superior properties to monomeric ICG. Despite their higher photoacoustic (PA) signals for imaging and heating stability during photothermal therapy (PTT), they exhibit low stability under a biological milieu. Our group previously proposed a simple procedure for in-situ preparation of IJA into liposomes, accelerating their formation and optical properties. To comprehend their potential applications, we systematically investigated the effect of the lipid bilayer composition on ICG J-aggregation and stabilit

Semi-Rigid Nitroxide Spin Label for Long-Range EPR Distance Measurements of Lipid Bilayer Embedded β-Peptides.

β-Peptides are an interesting new class of transmembrane model peptides based on their conformationally stable and well-defined secondary structures. Herein, we present the synthesis of the paramagnetic β-amino acid β3 -hTOPP (4-(3,3,5,5-tetramethyl-2,6-dioxo-4-oxylpiperazin-1-yl)-d-β3 -homophenylglycine) that enables investigations of β-peptides by EPR spectroscopy. This amino acid adds to the, to date, sparse number of β-peptide spin labels. Its performance was evaluated by investigating the helical turn of a 314 -helical transmembrane model β-peptide. Nanometer distances between two incorpo

Single-molecule force spectroscopy to decipher the early signalling step in membrane-bound penicillin receptors embedded into a lipid bilayer.

Understanding the molecular mechanism by which the signal of the presence of an antibiotic is transduced from outside to inside the bacterial cell is of fundamental interest for the β-lactam antibiotic resistance problem, but remains difficult to accomplish. No approach has ever addressed entire penicillin receptors in a membrane environment. Here we describe a method to investigate the purified Bacillus licheniformis BlaR1 receptor -a membrane-bound penicillin receptor involved in β-lactam resistance- embedded into a lipid bilayer in absence or presence of penicillin. By selecting a mutated r

Substrate-induced conformational changes in the nucleotide-binding domains of lipid bilayer-associated P-glycoprotein during ATP hydrolysis.

P-glycoprotein (Pgp) is an efflux pump important in multidrug resistance of cancer cells and in determining drug pharmacokinetics. Pgp is a prototype ATP-binding cassette transporter with two nucleotide-binding domains (NBDs) that bind and hydrolyze ATP. Conformational changes at the NBDs (the Pgp engines) lead to changes across Pgp transmembrane domains that result in substrate translocation. According to current alternating access models (substrate-binding pocket accessible only to one side of the membrane at a time), binding of ATP promotes NBD dimerization, resulting in external accessibil

Property of large conductance Ca(2+)-activated K+ channels from Fasciola hepatica incorporated into planar lipid bilayer.

Fasciola hepatica causes biliary epithelial hyperplasia and obstructive jaundice in humans and animals. Using a planar lipid bilayer technique, we further characterized the single channel property of large conductance K(+)-permeable channels that were previously identified from F. hepatica. The single channel conductance was 254.7±17.9 pS under a symmetrical 200/200 mM (cis/trans) KCl gradient. Open state probability (P(o)) varied from channel to channel at a given membrane potential and Ca(2+) concentration, but increased with voltage (-60 to +40 mV) and cis Ca(2+) (1-200 μM).
