Melanoma's ability to progress and develop resistance to therapies are the result of both genotypic changes and substantial levels of adaptability at the phenotypic and metabolic level. One of the most clinically relevant adaptive states is the microphthalmia associated transcription factor MITF low, which has been correlated with de-differentiation, invasion potential and drug tolerance, as well as altered redox status. As important regulators of this adaptation, lipid droplets have become involved in coordinating fat stores and mobilization, organelle signaling, membrane lipid composition an

Adipose tissue plays a central role in systemic metabolic regulation, and impairments in lipid turnover are a hallmark of obesity associated … by increased stearoyl-CoA desaturase-1 (SCD1). These contribute to adipocyte hypertrophy, impaired lipid mobilization, and dysfunctional extracellular matrix. This study investigated whether a brief strength training protocol following fourteen weeks of induction on a high-fat diet containing 35% fat could restore the lipolytic machinery and rebalance lipid metabol …

High yielding dairy cows undergo a period of energy deficit in early lactation and mobilise body reserves for milk production. In this period a high proportion of cows develop moderate or severe fatty liver. Fat is deposited in other tissues at the same time, particularly skeletal muscle. The relationship between measurements of fat mobilisation and fat deposition in liver and muscle has been investigated in a group of 19 cows in the post calving period. Cows with higher levels of indices of mobilisation, namely plasma free fatty acid and D(--) -3- hydroxybutyrate concentration and subcutaneou

The effects of noradrenaline (NA) and isopropyl-noradrenaline (ISNA) on glycerol release and cAMP levels in sc adipose tissue were studied in vitro in 27 patients with hyperthyroidism. In 11 patients, the studies were repeated after 6--12 months of treatment for hyperthyroidism. A third group comprised 21 euthyroid patients otherwise healthy except for morbid obesity. The lipolytic response to ISNA, observed in untreated thyrotoxic patients, was found to be reduced by 30% when the patients were reexamined after treatment for thyrotoxicosis. This reduction was attributable to a decrease in the

Herein we report an improved method to separate cardiolipin (Ptd(2)Gro) from tissue total lipid extracts using a biphasic solvent system combined with high performance liquid chromatography. This method uses a normal phase silica column and two mobile phases: mobile phase A that was n-hexane:2-propanol (3:2 by vol) and mobile phase B that was n-hexane:2-propanol:water (56.7:37.8:5.5 by vol). The initial solvent conditions were 95% A and 5% B, with a flow rate of 1.5 mL/min. The samples were from non-derivatized aliquots of liver, heart, or brain lipid extracts. The peak corresponding to Ptd(2)

Mycobacterium leprae (M. leprae), the causative agent of leprosy, parasitizes within the foamy or enlarged phagosome of macrophages where rich lipids accumulate. Although the mechanisms for lipid accumulation in the phagosome have been clarified, it is still unclear how such large amounts of lipids escape degradation. To further explore underlying mechanisms involved in lipid catabolism in M. leprae-infected host cells, we examined the expression of hormone-sensitive lipase (HSL), a key enzyme in fatty acid mobilization and lipolysis, in human macrophage THP-1 cells. We found that infection by

Hyperglycemia is common after acute stroke and is associated with a worse outcome of stroke. Thus, a better understanding of stress hyperglycemia is helpful to the prevention and therapeutic treatment of stroke. Chromium is an essential nutrient required for optimal insulin activity and normal carbohydrate and lipid metabolism. Beyond its nutritional effects, dietary supplement of chromium causes beneficial outcomes against several diseases, in particular diabetes-associated complications. In this study, we investigated whether post-stroke hyperglycemia involved chromium dynamic mobilization i

1. Lipid mobilization during a hypocaloric diet may be enhanced by a pharmacological approach using beta 2-adrenoceptor agonists or alpha 2-adrenoceptor antagonists. Studies were undertaken in the dog, an animal model presenting fat cell antilipolytic alpha 2- and lipolytic beta-adrenoceptors, in order, firstly, to demonstrate the presence of beta 2 subtype adrenoceptors on adipocytes and, secondly, to compare the effects of procaterol (beta 2-adrenoceptor agonist) and of yohimbine (alpha 2-adrenoceptor antagonist) on metabolic, endocrinological and cardiovascular parameters. 2. BackgroundCancer metabolism is a field focused on the unique alterations in metabolic pathways that occur in cancer cells, distinguishing them from the metabolic processes in normal cells.MethodsAn extensive review of the current literature on the metabolic adaptation of cancer cells was carried out in the current study.ResultsThe rapidly proliferating cells require high levels of molecules, such as glucose, amino acids, lipids, and nucleotides, along with increased energy demand (ATP). These requirements are met through alterations in the processes involving glucose, amino acid, lipid, and nu

Perilipin 5 (PLIN5) is a lipid droplet (LD) protein highly expressed in oxidative tissues, including the fasted liver. However, its expression also increases in nonalcoholic fatty liver. To determine whether PLIN5 regulates metabolic phenotypes of hepatosteatosis under nutritional excess, liver targeted overexpression of PLIN5 was achieved using adenoviral vector (Ad-PLIN5) in male C57BL/6J mice fed high-fat diet. Mice treated with adenovirus expressing green fluorescent protein (GFP) (Ad-GFP) served as control. Ad-PLIN5 livers increased LD in the liver section, and liquid chromatography with

Adiponectin (ADPN) is related to fatty acid synthesis and oxidation in mammals. In chickens, the lipid metabolism, structure and sequence of ADPN are different from that in mammals. The aim of this study was to determine the role of ADPN in broilers lipid metabolism by investigating the temporal and spatial expression profiles of ADPN and its receptors, as well as their response to feed restriction. The results showed that the abdominal fat has the highest expression level, followed by the duodenum, glandular stomach, heart, hypothalamus, liver, and skeletal muscle. Broilers have high energy m

Undernutrition accelerates body fat mobilization to alleviate negative energy balance, which disrupts homeostasis of lipid metabolism in maternal liver. However, little is known about its effect on fetal metabolism and development. Here, a sheep model was used to explore whether maternal undernutrition induces fetal lipid metabolism disorder and further inhibits fetal hepatic development. Twenty pregnant ewes were either fed normally or restricted to 30% level for 15 d, after which fetal hepatic samples were collected to conduct transcriptome, metabolome, histomorphology, and biochemical analy

The plant phloem is a long-distance conduit for the transport of assimilates but also of mobile developmental and stress signals. These signals can be sugars, metabolites, amino acids, peptides, proteins, microRNA, or mRNA. Yet small lipophilic molecules such as oxylipins and, more recently, phospholipids have emerged as possible long-distance signals as well. Analysis of phloem (phospho)lipids, however, requires enrichment, purification, and sensitive analysis. This chapter describes the EDTA-facilitated approach of phloem exudate collection, phase partitioning against chloroform-methanol for

Meibomian lipid layers were studied in the anterior mirror area by reflecting microscopy and interference microscopy. Using these techniques, it was not possible to correlate the biophysical and morphological data. Brewster angle microscopy provides direct observation of the spread Meibomian lipid layer with simultaneous registration of the surface pressure. It is based on the fact that He-Ne laser light, which is incident at a water surface under the Brewster angle, does not reflect visible light. After spreading of a lipid film, the angle of the incident light beam varies, causing reflection

Although biological membranes exhibit complex and dynamic organization, the mechanistic role of molecular crowding in governing lateral membrane heterogeneity remains poorly characterized experimentally. Here, we provide direct experimental visualization showing that crowding-induced condensation of membrane-anchored macromolecules above the bilayer interface reorganizes membrane dynamics and gives rise to spatially heterogeneous lipid mobility. Both PEGylated lipids and membrane-anchored proteins undergo surface-density-driven condensation on supported lipid bilayers (SLBs), forming immobile

Untargeted lipidomics enables comprehensive characterization of lipid profiles; however, analytical variability in sample preparation, chromatographic conditions, and data processing often limits reproducibility. Thus, this study aimed to optimize untargeted lipidomics analysis by evaluating chromatographic columns, mobile-phase additives, and lipid extraction methods to maximize feature detection, reproducibility, and overall data quality. Chromatographic performance was compared between ethylene-bridged hybrid (BEH) and charged-surface hybrid columns. For mobile-phase additives, 10 mM ammoni
