Lipopeptide-loaded mesoporous silica nanoparticles suppress root rot disease of soybean (Glycine max L.): modulating metabolite profile and endophytic bacterial community.

BACKGROUND: Soybean production is severely affected by the soil-borne phytopathogenic, Fusarium oxysporum, which causes root rot disease. Artificial nanoplatforms have attracted attention as a sustainable strategy to promote crop growth and protection. RESULTS: In this study, mesoporous silica nanoparticles (MSNs) loaded with lipopeptide (Lip), referred to as Lip@MSNs, were synthesized with a diameter of approximately 50 nm and a Lip loading capacity of 22.40 wt%. Lip@MSNs demonstrated potent antifungal activity against F. oxysporum, inducing cell membrane disruption, permeability alterations

Functional Characterization of a Bacillus-Derived Novel Broad-Spectrum Antifungal Lipopeptide Variant against Candida tropicalis and Candida auris and Unravelling Its Mode of Action.

Limited treatment options, recalcitrance, and resistance to existing therapeutics encourage the discovery of novel antifungal leads for alternative therapeutics. Antifungal lipopeptides have emerged as potential candidates for developing new and alternative antifungal therapies. In our previous studies, we isolated and identified the lipopeptide variant AF4 and purified it to homogeneity via chromatography from the cell-free supernatant of Bacillus subtilis. AF4 was found to have broad-spectrum antifungal activity against more than 110 fungal isolates. In this study, we found that clinical iso…

Microcolin A, a lipopeptide from marine cyanobacteria, suppresses the non-small cell lung carcinoma by regulating the C-terminal of Hsp90α and ROS/ERK signaling pathways.

Lung cancer remains to be the leading cause of cancer-related mortality worldwide, with non-small cell lung carcinoma (NSCLC) accounting for approximately 85% and the five-year survival rate below 20%. Microcolin A (Mic A), a lipopeptide derived from marine cyanobacteria, exhibits significant antitumor activity, however, its underlying mechanism is unclear. Hsp90 is highly expressed in various cancer tissues and positively correlated with tumor progression and aggressiveness. Existing studies have shown that most Hsp90 inhibitors target the N-terminal ATP-binding pockets, however, the clinical

A Family of Cyclic Lipopeptides Found in Human Isolates of Bacillus Ameliorates Acute Colitis via Direct Agonism of Toll-Like Receptor 2 in a Murine Model of Inflammatory Bowel Disease.

BACKGROUND: The Bacillus-derived cyclic lipopeptides (surfactin, iturin, and fengycin) form potent Heterogeneous Lipopeptide Micelle (HeLM) complexes. HeLM is a small molecule that has been shown to have immunomodulatory effects. However, how HeLM regulates inflammation is not clear, moreover its application to Inflammatory Bowel Disease (IBD), specifically Ulcerative Colitis (UC), has not been tested before. AIMS: To use a murine model of IBD and determine the effects of HeLM and related molecular mechanisms of action. METHODS: Colitis was induced in mice by administration of 4% Dextran Sod…

Effective inhibition of lung metastatic melanoma by pH-responsive lipopeptide/PLGA hybrid nanoparticles silencing PD-L1 with augmented suppression of the AKT/β-catenin pathway.

Boosting immune response has been extensively used for improving the inhibition of metastatic melanoma with immune checkpoint inhibitors (ICIs). However, its efficacy has been largely counteracted by complicated immune escape mechanisms and immune-related adverse events. To address these challenges, we presented a new approach for effective inhibition of lung metastatic melanoma (LMM) by augmented suppression of the AKT/β-catenin pathway. The pH-responsive C18-pArg8-pHis10 lipopeptide/PLGA hybrid nanoparticles (SLNPs) were developed to complex with PD-L1 siRNA (siP) and encapsulate hydrophobic

Liposomal co-encapsulation of a novel gemini lipopeptide and a CpG-ODN induces a strong Th1 response with the co-activation of a Th2/Th17 profile and high antibody levels.

A successful vaccine depends on its capacity to elicit a protective immune response against the target pathogen. The adjuvant used plays an important role in enhancing and directing the immune response. Liposomes are vaccine adjuvants that allow the co-encapsulation of antigens and immunostimulants. Our aim was to evaluate the adjuvanticity of a cationic liposome (Lip) formulated with a novel gemini lipopeptide (AG2-C16) alone or in combination with CpG-ODN as immunostimulants. To achieve this, we used the recombinant clumping factor of Staphylococcus aureus (rClfA) as a model antigen, in a mu

A dePEGylated Lipopeptide-Based Pan-Coronavirus Fusion Inhibitor Exhibits Potent and Broad-Spectrum Anti-HIV-1 Activity without Eliciting Anti-PEG Antibodies.

We previously identified a lipopeptide, EK1C4, by linking cholesterol to EK1, a pan-CoV fusion inhibitory peptide via a polyethylene glycol (PEG) linker, which showed potent pan-CoV fusion inhibitory activity. However, PEG can elicit antibodies to PEG in vivo, which will attenuate its antiviral activity. Therefore, we designed and synthesized a dePEGylated lipopeptide, EKL1C, by replacing the PEG linker in EK1C4 with a short peptide. Similar to EK1C4, EKL1C displayed potent inhibitory activity against severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) and other coronaviruses. In this

Increases in IL-33 production by fimbriae and lipopeptide from Porphyromonas gingivalis in mouse bone marrow-derived dendritic cells via Toll-like receptor 2.

Interleukin-33 (IL-33) is an IL-1 cytokine family member that is involved in the development of chronic inflammatory diseases and the initiation of allergic inflammation in response to pathogens. Porphyromonas gingivalis is a primary pathogen that is involved in chronic periodontitis and its bacterial components induce inflammatory responses. Dendritic cells (DCs) recognize pathogen- associated molecular patterns by expression of pattern-recognition receptors, such as Toll-like receptors (TLRs). DCs play an essential role in resistance to infection and maintenance of mucosal immune system. In

Activation of human monocytic cells by Treponema pallidum and Borrelia burgdorferi lipoproteins and synthetic lipopeptides proceeds via a pathway distinct from that of lipopolysaccharide but involves the transcriptional activator NF-kappa B.

There is increasing evidence that lipoproteins of Treponema pallidum and Borrelia burgdorferi are key inflammatory mediators during syphilis … bserved that expression of human CD14 imparted responsiveness to LPS but not to OspA or spirochetal lipopeptides (assessed by induction of NF-kappa B and expression of surface immunoglobulin M). Finally, the biological relevance of the observation that T. pallidum lipoproteins-lipopeptides induce both NF-kappa B and cytokine production in monocytes was support …

A cystine-containing cationic lipopeptide-based injectable hydrogel with antimicrobial activities against multi-drug resistant strains and anti-biofilm efficacy against methicillin-resistant Staphylococcus aureus.

A ubiquitous global threat of emerging multi-drug resistant (MDR) strains causing outbreaks of biofilm-mediated hospital-acquired infections (HAIs) has resulted in severe nosocomial contagious diseases, chronic wound inflammation, and lethal sepsis. Surface contamination of medical devices, implants and community transmission have further worsened the persistently high rate of morbidity and mortality spawned by epidemic resistant strains of opportunistic pathogens such as methicillin-resistant Staphylococcus aureus (MRSA). Herein, a disulphide bridge containing an amphiphilic cationic peptide

Effect of Fatty Acid Chain Modification on the Self-Assembly Behavior and Antimicrobial Activity of Antimicrobial Peptides.

Background: The overuse of traditional antimicrobial agents has accelerated the global spread of drug-resistant bacteria, posing a severe threat to global public health. Methods: In this work, a series of lipopeptides with varying fatty acid chain lengths were designed using the targeting antimicrobial peptide CL5 as the parental peptide. A variety of technical methods, including spectroscopic techniques, electron microscopy and computer simulation, were adopted to explore the self-assembly properties of the lipopeptides and their antimicrobial properties against Gram-positive and Gram-negativ…

Mutanobactin-D, a Streptococcus mutans Non-Ribosomal Cyclic Lipopeptide, Induces Osteogenic/Odontogenic Differentiation of Human Dental Pulp Stem Cells and Human Bone Marrow Stem Cells.

Bacterium-triggered carious lesions implicate dental hard tissue destruction and the simultaneous initiation of regenerative events comprising dental stem cell activation. Streptococcus mutans (S. mutans) is a prominent pathogen of the oral cavity and the principal cause of caries. S. mutans generates complex products involved in interbacterial interactions, including Mutanobactin-D (Mub-D), which belongs to a group of non-ribosomal cyclic lipopeptides. In the present study, we aimed to analyse the potential role of the synthetic Mub-D peptide in cell populations involved in tissue regenerativ…

The bacterial lipopeptide iturins induce Verticillium dahliae cell death by affecting fungal signalling pathways and mediate plant defence responses involved in pathogen-associated molecular pattern-triggered immunity.

Verticillium wilt in cotton caused by Verticillium dahliae is one of the most serious plant diseases worldwide. Because no known fungicides or cotton cultivars provide sufficient protection against this pathogen, V. dahliae causes major crop yield losses. Here, an isolated cotton endophytic bacterium, designated Bacillus amyloliquefaciens 41B-1, exhibited greater than 50% biocontrol efficacy against V. dahliae in cotton plants under greenhouse conditions. Through high-performance liquid chromatography and mass analysis of the filtrate, we found that the antifungal compounds present in the stra

Protein digest extraction strategies improve the identification of lipidated peptides in bottom-up proteomics.

Alteration of proteins by attaching lipid moieties is a type of post-translational modification that affects the properties of proteins. It plays a role in various physiological processes, mostly due to direct interaction with cell membranes. Proteomic identification of lipoproteins typically depends on detecting their unmodified regions. However, lipopeptides, due to their distinct structural and chemical properties, display chromatographic and mass-spectrometric behavior which differs substantially from that of unmodified peptides. The altered chromatographic and mass-spectrometric behavior …

Position Effect of Fatty Acid Modification on the Cytotoxicity and Antimetastasis Potential of the Cytotoxic Peptide Lycosin-I.

Peptide modification with fatty acids is an effective method to improve peptide performance. We previously investigated the fatty acid modification of R-lycosin-I, a cytotoxic peptide derived from lycosin-I from the venom of the spider Lycosa singoriensis. In this study, we further investigated the position effects of fatty acid modification of lycosin-I. Dodecanoic acid was covalently coupled to the α/ε-amino group of one of the seven Lys residues of lycosin-I, generating eight different lipopeptides. Although all the lipopeptides had significantly improved cytotoxicity compared with lycosin-…

The design and proof of concept for a CD8(+) T cell-based vaccine inducing cross-subtype protection against influenza A virus.

In this study, we examined the reactivity of human peripheral blood mononuclear cells to a panel of influenza A virus (IAV) CD8(+) T-cell epitopes that are recognised by the major human leukocyte antigen (HLA) groups represented in the human population. We examined the level of recognition in a sample of the human population and the potential coverage that could be achieved if these were incorporated into a T-cell epitope-based vaccine. We then designed a candidate influenza vaccine that incorporated three of the examined HLA-A2-restricted influenza epitopes into Pam2Cys-based lipopeptides. Th…

Self-assembled Lipid Nanoparticles for Killing Triple Negative Breast Cancer Cells.

Triple negative breast cancers (TNBCs) lacking estrogen receptor (ER), progesterone receptor (PR) and human epidermal growth factor receptor 2 (HER2) on their cell surfaces are highly aggressive, difficult-to-treat and often relapse. Herein, we report on the self-assembled lipid nanoparticles (LNPs) of two new pegylated lipopeptides for killing TNBCs (MDA-MB-231). The pegylated lipopeptides were synthesized by conjugating an n-hexadecyl hydrophobic tail to one end of a (PEG)27 unit the other distal end of which was covalently grafted with two previously reported tumor targeting RGDK- and CGKRK…
