Cuproptosis shows good application prospects in tumor therapy. However, the copper efflux mechanism and highly expressed intracellular reducing substances can inhibit the cuproptosis effects. In this study, a platelet vesicle (PV) coated cuprous oxide nanoparticle (Cu2O)/TBP-2 cuproptosis sensitization system (PTC) was constructed for multiple induction of tumor cuproptosis. PTC was prepared by physical extrusion of AIE photosensitizer (TBP-2), Cu2O, and PV. After the biomimetic modification, PTC can enhance its long-term blood circulation and tumor targeting ability. Subsequently, PTC was rap

Previously we have reported the capacity of the fusion protein PD3, composed of the P64k protein and the envelope (E) fragment from amino acids (aa) 286-426 of dengue-2 virus (DEN-2), to induce a functional immune response in mice against the homologous virus. In that case, the E fragment was inserted within the lipoyl-binding domain of the meningococcal P64k protein. In the present study, to test the functionality of the same E region from dengue-1 (DEN-1), a similar construct was made. Furthermore, another alternative of fusion protein was also constructed where the same E fragment from DEN-

BACKGROUND: Autoimmune diseases (AIDs) are characterized by disrupted immune tolerance, chronic inflammation, and organ-specific tissue injury. Their pathogenesis involves the interplay between genetic susceptibility and environmental factors. Although current biological agents and glucocorticoid therapies have improved clinical outcomes, long-term remission rates remain limited, with persistent risks including infection, organ toxicity, and immunogenicity. Emerging evidence indicates that copper homeostasis imbalance, mitochondrial dysfunction, redox stress, and immunometabolic remodeling coe

The advent of immune checkpoint inhibitors has fundamentally changed the face of oncology, but the prevalence of primary and acquired resistance has led to the need for the development of novel approaches to remodel the immunosuppressive tumor microenvironment. Central to this challenge is the dichotomy between hot inflamed and cold (immune-excluded) tumors, which is increasingly associated with the immunogenicity of cell death modalities used by therapeutic agents. While apoptosis is largely immunologically silent, regulated cell death pathways driven by the catastrophic failure of mitochondr

BACKGROUND: Cuproptosis is a novel copper-dependent form of cell death closely associated with mitochondrial metabolism and protein lipoylation. We propose a "Copper Buffering-Execution Imbalance" hypothesis where metallothionein 2A (MT2A) exhaustion leads to free copper accumulation, activating the FDX1-PDH axis and triggering mitochondrial cuproptosis. Metallothioneins (MTs) are key regulators of copper homeostasis, but their mechanistic roles in severe infections remain unclear.
METHODS: We employed a systems biology approach integrating in silico modeling and experimental validation. We a

Diabetic cardiomyopathy (DCM) is a myocardial injury induced by chronic hyperglycemia. Recent literature has reported that hyperglycemia causes cardiac copper overload, triggering copper-induced cell death (cuproptosis). Copper leads to the dysregulation of ferredoxin-1 (FDX-1) and lipoic acid synthase (LIAS), two enzymes essential for the lipoylation process in the Krebs cycle, and are considered key markers of cuproptosis. We aim to explore the protective effects of carvacrol (CAR) against DCM by targeting cuproptosis. Diabetes was induced by a high-fat diet/streptozotocin (35 mg/kg) model.

The recent identification of cuproptosis, a regulated cell death pathway dependent on copper, has reshaped our understanding of post-stroke neurodegeneration. This review comprehensively analyzes the dual role of copper dyshomeostasis in stroke pathophysiology, emphasizing its contributions to mitochondrial impairment, disrupted protein lipoylation, and dysregulated copper-iron crosstalk. We delineate three principal mechanisms through which copper aggravates neuronal damage: (1) copper overload disrupts Fe-S cluster assembly, leading to subsequent collapse of the TCA cycle; (2) FDX1-driven th

Friedreich's ataxia (FRDA) is a neurodegenerative disorder caused by frataxin deficiency, characterized by progressive cerebellar dysfunction and neuronal loss. Although synaptic abnormalities are an early feature of FRDA, the molecular mechanisms linking frataxin deficiency to synaptic dysfunction remain incompletely defined. In this study, we investigated post-translational regulation of the AMPA receptor subunit GluR2 in the cerebellum of the frataxin knockdown (FRDAkd) mouse model. GluR2 protein levels are reduced early following frataxin knockdown, despite unchanged mRNA expression and pr

Failure of appropriate synaptic regulation in the epileptogeniclesion leads to sudden onset ictogenesis in severe epileptic seizure. Using surgically excised patient brain tissue, our recent biochemical study revealed chronic reduction of synaptic protein expressions in the epileptogenic lesions of patients with hippocampal sclerosis. α-amino-3-hydroxy-5-methyl-4-isoxazole propionate (AMPA) receptors are the major excitatory neurotransmitter receptors in human brain. Decreased AMPA receptors expression and their phosphorylations in the epileptogenic lesion induce an overall reduction of excita

BACKGROUND: Protein palmitoylation, a critical posttranslational modification, plays an indispensable role in various cellular processes, including the regulation of protein stability, mediation of membrane fusion, facilitation of intracellular protein trafficking, and participation in cellular signaling pathways. It is also implicated in the pathogenesis of diseases, such as cancer, neurological disorders, inflammation, metabolic disorders, infections, and neurodegenerative diseases. However, its regulatory effects on sperm physiology, particularly motility, remain unclear. This study aimed t

The downregulation of Cadm4 (Cell adhesion molecular 4) is a prominent feature in demyelination diseases, yet, the underlying molecular mechanism remains elusive. Here, we reveal that Cadm4 undergoes specific palmitoylation at cysteine-347 (C347), which is crucial for its stable localization on the plasma membrane (PM). Mutation of C347 to alanine (C347A), blocking palmitoylation, causes Cadm4 internalization from the PM and subsequent degradation. In vivo experiments introducing the C347A mutation (Cadm4-KI) lead to severe myelin abnormalities in the central nervous system (CNS), characterize

Introduction: S-acylation is the attachment of fatty acids not only to cysteines of cellular, but also of viral proteins. The modification is often crucial for the protein´s function and hence for virus replication. Transfer of fatty acids is mediated by one or several of the 23 members of the ZDHHC family of proteins. Since their genes are linked to various human diseases, they represent drug targets.Areas covered: The authors explore whether targeting acylation of viral proteins might be a strategy to combat viral diseases. Many human pathogens contain S-acylated proteins; the ZDHHCs involve

Eukaryotic parasites possess complex life cycles and utilize an assortment of molecular mechanisms to overcome physical barriers, suppress and/or bypass the host immune response, including invading host cells where they can replicate in a protected intracellular niche. Protein S-palmitoylation is a dynamic post-translational modification in which the fatty acid palmitate is covalently linked to cysteine residues on proteins by the enzyme palmitoyl acyltransferase (PAT) and can be removed by lysosomal palmitoyl-protein thioesterase (PPT) or cytosolic acyl-protein thioesterase (APT). In addition

BACKGROUND: Crohn's disease (CD) is a chronic inflammatory bowel disease marked by immune imbalance and monocyte dysfunction. IFITM3, a palmitoylation-related immune protein, may play a role in this process, but its involvement in CD remains unclear. This study aimed to explore the causal role of IFITM3 and related proteins in CD using Mendelian randomization, multi-omics analysis, and machine learning, to identify potential diagnostic markers and therapeutic targets.
METHODS: We performed Mendelian randomization (MR) analysis using pQTL data to identify causal associations between monocyte p

Pancreatic ductal adenocarcinoma (PDAC) is characterized by frequent SMAD4 inactivation and profound lipid metabolic rewiring, yet how these processes intersect especially in SMAD4+ PDAC remains elusive. Here, we identify palmitoylation as a previously unrecognized post-translational modification of SMAD4. Combining biochemical labeling, mutagenesis, and functional assays, we demonstrate that SMAD4 is palmitoylated at cysteine 363 by the acyltransferase ZDHHC22 and depalmitoylated by APT2. Mechanistically, palmitoylation enhances SMAD4 protein stability, facilitates the interaction of SMAD4 wi

Gasdermin D (GSDMD) executes pyroptosis by forming membrane pores, yet how these structures assemble and are regulated in cells has remained technically inaccessible. We introduce polymer-supported plasma membranes (PSPMs), which preserve native PM properties while providing cytosolic access for nanoscopic imaging. Combining PSPMs with DNA-PAINT super-resolution microscopy, we visualize human and mouse GSDMD nanostructures directly at the PM of pyroptotic cells and uncover species-specific differences in pore size. Quantitative analyses reveal that GSDMD assembles into heterogeneous macromolec

OBJECTIVE: The current work focused on elucidating the regulatory mechanisms of palmitoylation-related genes (PRGs) in influencing the occurrence and development of clear cell renal cell carcinoma (ccRCC), and identifying possible biomarkers and therapeutic targets.
METHODS: Integrative analyses were performed using multiple databases. Two-sample Mendelian randomization (MR) was first applied for identifying PRGs causally associated with ccRCC. Findings were then validated by summary-data-based Mendelian randomization (SMR), bioinformatics analyses, and experimental verification. Functional e

Osteoarthritis (OA) lacks approved disease-modifying therapies. Here, by integrating quantitative proteomics with reanalysis of public single-cell RNA sequencing data from human articular cartilage, we identify thymidine kinase 1 (TK1) as a pathogenic regulator linking inflammatory stress to metabolic rewiring and matrix breakdown. TK1 is markedly upregulated in OA chondrocytes. Mechanistically, IL-1β enhances S-palmitoylation of TK1 at Cys153, which promotes USP9X-dependent deubiquitination and prevents proteasomal degradation, leading to TK1 accumulation. Stabilized TK1 acts as a scaffold to

Tegument proteins of human cytomegalovirus (HCMV) play essential roles in viral assembly, coordinating interactions among capsids, membranes, and host-derived components. pp28 (UL99), a dominant tegument protein expressed during late infection, is essential for cytoplasmic envelopment and proper trafficking to the viral assembly compartment (vAC). Here, we identify a critical role for palmitoylation in pp28 function. Using site-directed mutagenesis and acyl-resin assisted capture (acyl-RAC) assays, we show that palmitoylation occurs at conserved cysteine residues (Cys6, Cys10, Cys11) near the

INTRODUCTION: Neonatal anesthesia-related neurological impairments are of significant concern, closely linked to oligodendrocyte dysfunction. However, there is a notable temporal discrepancy between the sustained development of oligodendrocytes (myelination) and the short-term vulnerability to anesthesia exposures.
OBJECTIVES: Given the significant rise in iron demand by oligodendrocytes during neonatal period, our objective was to clarify the potential roles and underlying mechanisms of iron homeostasis, particularly focusing on transferrin receptor 1 (TfR1), in governing the transient susce
