A study of the major Australian snake venoms in the monkey (Macaca fascicularis). II. Myolytic and haematological effects of venoms.

Rhesus monkeys were kept in a restraining apparatus and while conscious injected with the venoms of a range of Australian snakes. Although the action of restraining itself caused an elevation of the animals' plasma creatine kinase (C.K.), the rise was up to 5 times greater in envenomed monkeys. The venoms of the Tiger snake, Mulga, Beaked Sea snake, Rough-Scaled, Copperhead and Red-Bellied snakes were all powerfully myolytic, and those of the Taipan and Small-Eyed snake less so. No myolytic activity was found in the venoms of the Common Brown snake and Death Adder, and the results with Dugite,

The Potency of Extracellular Vesicles Derived From Mesenchymal Stem Cells Macaca fascicularis as a Candidate Cancer Therapeutic Agents.

BACKGROUND: Extracellular vesicles (EVs) are membrane vesicles secreted by cells that carry several bioactive molecules, which are currently used in biomedical research as a source of exosomes and to investigate their effect in response to cancer studies. METHODS: EVs isolated from the supernatant of Macaca fascicularis Mesenchymal Stem Cells (MSCs), using the commercial ExoQUICK kit, and markers validation evaluated using quantitative reverse transcription Polymerase Chain Reaction (qRT-PCR). EVs were further applied on breast cancer cells (MCF-7) and colorectal cancer cells (WiDr). RESULTS…

Characterisation of erythrocyte binding activity of Plasmodium cynomolgi duffy binding protein haplotypes from Macaca fascicularis in Peninsular Malaysia.

Human infections with the zoonotic Plasmodium cynomolgi are rising across Southeast Asia. This parasite employs two Duffy binding proteins (DBPs), PcDBP1(II) and PcDBP2(II), which interact with the Duffy antigen receptor chemokine (DARC) for erythrocyte invasion. Previous studies on P. cynomolgi DBPs have reported two distinct haplotype groups (groups 1 and 2) in Peninsular Malaysia. In this study, the differential erythrocyte binding activity of PcDBP1(II) and PcDBP2(II) haplotypes was investigated. Four isolates were selected for each DBP: group 1 consisted of a recent field isolate and the

Long-Tailed Macaques (Macaca fascicularis) in Urban Landscapes: Gastrointestinal Parasitism and Barriers for Healthy Coexistence in Northeast Thailand.

Gastrointestinal parasites have diverse life cycles that can involve people, animals, and the environment (e.g., water and soil), demonstrating the utility of One Health frameworks in characterizing infection risk. Kosumpee Forest Park (Thailand) is home to a dense population of long-tailed macaques (Macaca fascicularis) that frequently interact with tourists and local residents. Our study investigated the presence of zoonotic parasites, and barriers to healthy coexistence by conducting stool analysis on macaques (N = 102) and people (N = 115), and by examining risk factors for infection with …

Comparison of Continuous Glucose Monitoring between Dexcom G4 Platinum and HD-XG Systems in Nonhuman Primates (Macaca Fascicularis).

Timely knowing glucose level helps diabetic patients to manage the disease, including decisions about food, physical activity and medication. This study compared two continuous glucose monitoring systems in conscious and moving-free nonhuman primates (NHPs, Macaca fascicularis). Each normoglycemic or diabetic monkey was implanted with one Dexcom G4 Platinum subcutaneously or one HD-XG glucose sensor arterially for glucose monitoring. The glucose levels measured by both telemetry devices significantly correlated with the glucometer readings. The data of oral glucose tolerance test (oGTT) showed…

Pathology of experimental Machupo virus infection, Chicava strain, in cynomolgus macaques (Macaca fascicularis) by intramuscular and aerosol exposure.

Machupo virus, the causative agent of Bolivian hemorrhagic fever (BHF), is a highly lethal viral hemorrhagic fever of which little is known and for which no Food and Drug Administration-approved vaccines or therapeutics are available. This study evaluated the cynomolgus macaque as an animal model using the Machupo virus, Chicava strain, via intramuscular and aerosol challenge. The incubation period was 6 to 10 days with initial signs of depression, anorexia, diarrhea, mild fever, and a petechial skin rash. These were often followed by neurologic signs and death within an average of 18 days. Co

[Effect of TSA and VPA treatment on long-tailed macaque (Macaca fascicularis)-pig interspecies somatic cell nuclear transfer].

Long-tailed macaque-pig interspecies somatic cell nuclear transfer (iSCNT) is beneficial to yield embryonic stem cells from iSCNT embryos with similar genetic background as human, which can be used as materials for medical and basic research. The primary objective of this study was to investigate the effects of concentrations and treatment duration of two histone deacetylase inhibitors-Trichostatin A (TSA) and Valproic acid (VPA) and two different embryo culture media (PZM-3 and HECM-10) on the in vitro development of iSCNT embryos. The results suggested that when PZM-3 was used as the embryo

Induction of acquired factor IX inhibitors in cynomolgus monkey (Macaca fascicularis): a new primate model of hemophilia B.

Inherited hemophilia dog and other transient hemophilic animal models have been used for evaluation of hemostatic agents for use in treatment of hemophilia. We established the first nonhuman primate hemophilic model by immunizing cynomolgus monkeys with human FIX (hFIX) in adjuvants. FIX activities of all three hFIX-immunized monkeys decreased transiently to less than 10% in accordance with prolongation of activated partial thromboplastin time (APTT). Forty micrograms of human factor VIIa (hFVIIa) per kilogram body weight (that was reported to be clinically effective) was administered to the m

Chromosome abnormalities in peripheral blood lymphocytes from Macaca fascicularis and Erythrocebus patas (Cercopithecidae, Catarrhini) after X-ray irradiation.

In this paper, we describe the results of a qualitative and quantitative study of chromosomal reorganizations in X-irradiated (1 Gy and 2 Gy) lymphocytes from Macaca fascicularis (MFA) and Erythrocebus patas (EPA). A total of 515 breakpoints in M. fascicularis and 271 breakpoints in E. patas have been detected, identified and localized in the ideogram of the species. The Chi square test indicates that the distribution of breakpoints along the chromosomes is not random in M. fascicularis, and is not random for the p and q arms and bands in both species. Chromosome 5 of M. fascicularis (MFA5), c…

Regulation of bilirubin glucuronide synthesis in primate (Macaca fascicularis) liver. Kinetic analysis of microsomal bilirubin uridine diphosphate glucuronyltransferase.

Hepatic bilirubin uridine diphosphate glucuronyl-transferase (UDP-glucuronyltransferase) catalyzes the formation of bilirubin monoglucuronides (BMG, C-8 and C-12 isomers) and bilirubin diglucuronide (BDG) from bilirubin and the cosubstrate, UDP-glucuronic acid. Distinctive patterns of bile pigment excretion occur in different species and in pathologic disorders (e.g., Gilbert's syndrome). In normal human and monkey (Macaca fascicularis) bile, the proportion of BDG exceeds that of BMG and the C-8/C-12 BMG isomer ratio approaches unity. To investigate the mechanisms responsible for the patterns …

Prevalence and characterization of gastrointestinal and ectoparasites in long-tailed macaques (Macaca fascicularis) from ecotourism regions of Aceh, Indonesia.

BACKGROUND AND AIM: Long-tailed macaques (Macaca fascicularis) serve as critical sentinels for zoonotic disease surveillance due to their ecological proximity to human populations. Understanding their parasitic burden is vital for conservation and public health, particularly in ecotourism areas where human-primate interactions are frequent. This study aimed to investigate the prevalence, diversity, and morphological characteristics of gastrointestinal (GI) and ectoparasites in M. fascicularis across four natural habitats in Aceh Province, Indonesia. MATERIALS AND METHODS: A total of 100 fecal…

Development and validation of a multiplex PCR assay with melt curve analysis for detecting simian Plasmodium in wild Macaca fascicularis.

Accurate and sensitive detection of simian malaria parasites is essential for surveillance and risk assessment of zoonotic malaria. We developed and validated a SYBR Green-based real-time PCR assay targeting the msp1 gene to detect and differentiate P. knowlesi, P. cynomolgi, and P. inui. Species-specific amplification was confirmed through distinct melting temperature (Tm) profiles. The assay demonstrated high analytical sensitivity, with a limit of detection of 10 copies/µL, excellent specificity with no cross-reactivity, and strong reproducibility, with low coefficients of variation for bot

Changes in the number of astrocytes and microglia in the thalamus of the monkey Macaca fascicularis following long-term subclinical methylmercury exposure.

The effects of long-term subclinical exposure to methylmercury on the number of neurons, oligodendrocytes, astrocytes, microglia, endothelial cells and pericytes within the thalamus from the left side of the brain of the monkey Macaca fascicularis has been determined by use of the Optical Volume Fractionator stereological method. The accumulated burden of inorganic mercury (IHg) within these same cell types has been determined by autometallographic methods. Four groups of monkeys were exposed to methylmercury (MeHg; 50 micrograms Hg/kg body weight/day) by mouth for 6 months, 12 months, 18 mont…

Effects of sensitization to dinitrochlorobenzene (DNCB) on clinical pathology parameters and mitogen-mediated blastogenesis in cynomolgus monkeys (Macaca fascicularis).

Immunotoxicologic testing of drug candidates and environmental contaminants is of growing importance. Cutaneous delayed type hypersensitivity (DTH) is a convenient way of testing immune function in vivo. However, DTH testing must not interfere with interpretation of other relevant parameters. We have evaluated the effects of sensitization and challenge with dinitrochlorobenzene (DNCB) on clinical parameters routinely evaluated in toxicity testing and on lectin-mediated blastogenesis. Female cynomolgus monkeys were sensitized to DNCB with 4 daily applications of DNCB in acetone to the skin of t

Increased atherosclerosis and glomerulonephritis in cynomolgus monkeys (Macaca fascicularis) given injections of BSA over an extended period of time.

A study was conducted to compare the effects of experimental immune complex disease on the development of glomerulonephritis and aortic and coronary artery atherosclerosis. Fourteen adult male macaques (Macaca fascicularis) were fed a mildly atherogenic diet. Ten of these animals were given repeated intravenous injections of bovine serum albumin (BSA), and the remaining 4 were given similar injections of saline. Three of the monkeys given BSA responded with a high antibody titer, 4 with a moderate titer, and 3 with a low level titer to BSA. In all 4 monkeys with the moderate antibody response …

Differential effects of western versus mediterranean diets and psychosocial stress on ovarian function in female monkeys (Macaca fascicularis).

Ovarian dysfunction increases risk for chronic diseases of aging including cardiovascular disease, depression, cognitive impairment, as well as bone and muscle loss which promote frailty. Psychosocial stress can disrupt ovarian function, and recent observations suggest that consumption of a Western Diet may also. Determination of causal relationships among diet, psychosocial stress, and ovarian physiology is difficult in humans. A Cervical Spinal Cord Hemi-Contusion Injury Model Based on Displacement Control in Non-Human Primates (Macaca fascicularis).

Non-human primate (NHP) spinal cord injury (SCI) models can be informative in the evaluation of treatments that show promise in rodent models prior to translation to humans. In the present study, we aimed to establish a cervical spinal hemi-contusion model with controlled displacement and evaluate the abnormalities in behavior, electrophysiology, histology, and magnetic resonance imaging. Twelve adult NHPs were divided into an SCI group (n = 8, 24 and 48 weeks) and a control group (n = 4). An impactor (Φ = 4 mm) was driven to compress the left C5 cord at 800 mm/sec.
