Regional homogeneity and interhemispheric connectivity alterations in major depressive disorder.

BACKGROUND: Frontotemporal neural synchronization and hemispheric lateralization imbalances are core features of major depressive disorder (MDD), but objective Functional near-infrared spectroscopy (fNIRS) - derived biomarkers capturing these abnormalities remain underdeveloped. METHODS: We recruited 236 MDD patients and 124 healthy controls (HCs) who completed a verbal fluency task (VFT) during 52-channel fNIRS scanning. Regional Homogeneity (ReHo) and hemispheric Laterality Index (LI) were computed to assess local neural synchronization and interhemispheric balance across four VFT phases (p

Deep brain stimulation and neuroimmune mechanisms in major depressive disorder.

Major depressive disorder (MDD) is increasingly associated with neuroinflammation, hypothalamic-pituitary-adrenal (HPA) axis dysregulation, and disrupted neuroimmune interactions mediated by cytokine signaling aberrations, glial dysfunction, inflammasome activation, and impaired neuroplasticity. Within this pathophysiological context, deep brain stimulation (DBS) has emerged as a promising neuromodulatory intervention for treatment-resistant depression (TRD), though its underlying biological mechanisms remain poorly elucidated. This review systematically examines the role of neuroinflammation

Alterations in aperiodic neural activity associated with major depressive disorder.

Historically, aperiodic neural activity (i.e., the 1/f exponent and offset of electroencephalogram (EEG) power spectra) has been treated as neural noise. However, recent work has highlighted associations among aperiodic activity, aging, cognition, and psychopathology. While depression has been associated with cognitive deficits and dysregulation of excitatory/inhibitory neural mechanisms linked to aperiodic activity, to date, few studies have compared aperiodic activity in adults with and without depression, and none have evaluated aperiodic activity as a function of depressive chronicity. Lower ANXA3 Levels May Be Related to Major Depressive Disorder.

Annexin A3 (ANXA3) is a calcium-binding protein that plays a role in membrane phospholipid metabolism and inflammation, and significant alterations have been shown in some psychotic disorders. Because its association with major depressive disorder (MDD) is unclear, we aimed to compare serum ANXA3 levels in patients with MDD and healthy controls and to investigate their relationship with depression severity. Serum ANXA3 concentrations in 90 patients diagnosed with MDD and 90 healthy controls were measured by ELISA. Depression severity was assessed using the Beck Depression Inventory (BDI). Seru

Neural circuits mediating chronic stress: Implications for major depressive disorder.

Major depressive disorder (MDD), also known as depression, is a prevalent mental disorder that leads to severe disease burden worldwide. Over the past two decades, significant progress has been made in understanding the pathogenesis and developing novel treatments for MDD. Among the complicated etiologies of MDD, chronic stress is a major risk factor. Exploring the underlying brain circuit mechanisms of chronic stress regulation has been an area of active research for recent years. A growing body of preclinical and clinical research has revealed that abnormalities in the brain circuits are clo

Emotion regulation repertoires predict the risk of major depressive disorder.

BACKGROUND: Few studies explored emotion regulation (ER) repertoires (the tendency to utilize multiple ER strategies in different contexts) in patients with major depressive disorder (MDD). It remains unclear to what extent ER repertoires can assess the risk of MDD and other psychiatric disorders. METHOD: Three subsamples including 1397 non-clinical individuals, 135 MDD patients, and 127 healthy controls (HCs) were combined to explore ER repertoires. We further built classifiers of ER repertoires to distinguish MDD patients from HCs, and additionally examined its performance based on publishe

Heart rate variability in subthreshold depression and major depressive disorder.

BACKGROUND: The relationship between heart rate variability (HRV) and major depressive disorder (MDD) has been well explored. However, current researches lack an observation of HRV in subthreshold depression (SubD), which increases the risk of MDD and presents significant societal challenges. METHODS: This study compared resting state HRV among 128 MDD patients, 131 SubD individuals and 222 healthy controls (HC) recruited from the hospital, physical examination center, and colleges. Depression, anxiety and sleep quality were assessed by the Patient Health Questionnaire-9 (PHQ-9), Generalized

The RNA Demethyltransferase FTO Regulates Ferroptosis in Major Depressive Disorder.

Major depressive disorder (MDD) is a widespread and severe mental health condition characterized by persistent low mood and loss of interest. Emerging evidence suggests that ferroptosis, an iron-dependent form of cell death, and epigenetic dysregulation contribute to the pathogenesis of MDD. This study investigates the role of RNA demethylase FTO and autophagy regulator BECN1 in ferroptosis and their regulation by the active compound ginsenoside Rb1 (GRb1) as a potential antidepressant strategy. Hippocampal tissues from postmortem MDD patient brains and mice with chronic restraint stress (CRS)

Peripheral Endothelial Dysfunction Is Associated With Incident Major Depressive Disorder.

BACKGROUND: A subset of individuals with major depressive disorder (MDD) have a high burden of cardiovascular risk factors and cerebral small-vessel disease, implicating vascular disease in the development of depression. Cross-sectional studies demonstrate a link between endothelial dysfunction and MDD, but the prospective association between peripheral endothelial dysfunction (PED) and an incident diagnosis of MDD is unknown. METHODS AND RESULTS: Patients undergoing a baseline assessment of cardiovascular risk were evaluated for PED using reactive hyperemia-peripheral arterial tonometry (≤1.

Transcriptomic decoding of regional cortical vulnerability to major depressive disorder.

Previous studies in small samples have identified inconsistent cortical abnormalities in major depressive disorder (MDD). Despite genetic influences on MDD and the brain, it is unclear how genetic risk for MDD is translated into spatially patterned cortical vulnerability. Here, we initially examined voxel-wise differences in cortical function and structure using the largest multi-modal MRI data from 1660 MDD patients and 1341 controls. Combined with the Allen Human Brain Atlas, we then adopted transcription-neuroimaging spatial correlation and the newly developed ensemble-based gene category e

The relationship of major depressive disorder with Crohn's disease activity.

INTRODUCTION: Crohn's disease (CD) has been related to an increased prevalence of psychiatric disorders and suicide risk (SR). However, the nature of their relationship still deserves clarification. The aim of this study is to assess the prevalence of major depressive disorder (MDD) in patients with CD, and to investigate the relationship between MDD and CD outcomes. METHODS: A cross-sectional study involving CD patients was performed. CD activity was evaluated by the Harvey-Bradshaw index and CD phenotype by the Montreal classification. The presence of MDD was assessed by the Patient Health

Aberrant degree centrality profiles during rumination in major depressive disorder.

Rumination is closely linked to the onset and maintenance of major depressive disorder (MDD). Prior neuroimaging studies have identified the association between self-reported rumination trait and the functional coupling among a network of brain regions using resting-state functional magnetic resonance imaging (MRI). However, little is known about the underlying neural circuitry mechanism during active rumination in MDD. Degree centrality (DC) is a simple metric to denote network integration, which is critical for higher-order psychological processes such as rumination. During an MRI scan, indi

Major depressive disorder (mdd): emerging immune targets at preclinical level.

BACKGROUND: Major depressive disorder is a mental health disorder that is characterized by a persistently low mood and loss of interest. MDD is affecting over 3.8% of the global population as a major health problem. Its etiology is complex, and involves the interaction between a number of factors, including genetic predisposition and the presence of environmental stresses. AREAS COVERED: The role of the immune and inflammatory systems in depression has been gaining interest, with evidence suggesting the potential involvement of pro-inflammatory molecules like TNF, interleukins, prostaglandins

Aberrant brain dynamics in major depressive disorder with suicidal ideation.

BACKGROUND: Suicidal ideation (SI) is a common symptom of major depressive disorder (MDD). Accumulating studies demonstrated that MDD with SI was associated with static alterations in brain activity and functional connectivity. However, given that brain is a highly dynamic system, the changes of brain dynamic patterns in MDD with SI remain unknown. METHODS: We included 60 MDD patients with SI (MDD-SI), 58 MDD patients without SI (MDD-NSI), and 58 healthy controls (HCs) who underwent resting-state functional magnetic resonance imaging. The sliding-window approach was used to calculate the dyna

Increased Aperiodic Neural Activity During Sleep in Major Depressive Disorder.

BACKGROUND: In major depressive disorder (MDD), patients often express subjective sleep complaints, while polysomnographic studies report only subtle alterations of the electroencephalographic signal. We hypothesize that differentiating the signal into its oscillatory and aperiodic components may bring new insights into our understanding of sleep abnormalities in MDD. Specifically, we investigated aperiodic neural activity during sleep and its relationships with sleep architecture, depression severity, and responsivity to antidepressant treatment. METHODS: Polysomnography was recorded in 38 p

Molecular targets of endothelial phosphatidic acid regulating major depressive disorder.

Major depressive disorder (MDD) is a severe disease of unknown pathogenesis with a lifetime prevalence of ~10%. Therapy requires prolonged treatment that often fails. We have previously demonstrated that ceramide levels in the blood plasma of patients and in mice with experimental MDD are increased. Neutralization of blood plasma ceramide prevented experimental MDD in mice. Mechanistically, we demonstrated that blood plasma ceramide accumulated in endothelial cells of the hippocampus, inhibited phospholipase D (PLD) and thereby decreased phosphatidic acid in the hippocampus. Here, we demonstra
