Retinal TRP channels: Cell-type-specific regulators of retinal homeostasis and multimodal integration.

Transient receptor potential (TRP) channels are a widely expressed family of 28 evolutionarily conserved cationic ion channels that operate as primary detectors of chemical and physical stimuli and secondary effectors of metabotropic and ionotropic receptors. In vertebrates, the channels are grouped into six related families: TRPC, TRPV, TRPM, TRPA, TRPML, and TRPP. As sensory transducers, TRP channels are ubiquitously expressed across the body and the CNS, mediating critical functions in mechanosensation, nociception, chemosensing, thermosensing, and phototransduction. This article surveys cu

The Cardiac Genome Clinic: implementing genome sequencing in pediatric heart disease.

PURPOSE: This study investigated the diagnostic utility of nontargeted genomic testing in patients with pediatric heart disease. METHODS: We analyzed genome sequencing data of 111 families with cardiac lesions for rare, disease-associated variation. RESULTS: In 14 families (12.6%), we identified causative variants: seven were de novo (ANKRD11, KMT2D, NR2F2, POGZ, PTPN11, PURA, SALL1) and six were inherited from parents with no or subclinical heart phenotypes (FLT4, DNAH9, MYH11, NEXMIF, NIPBL, PTPN11). Outcome of the testing was associated with the presence of extracardiac features (p = 0.02

Cell-Based Mechanosensation, Epigenetics, and Non-Coding RNAs in Progression of Cardiac Fibrosis.

The heart is par excellence the 'in-motion' organ in the human body. Compelling evidence shows that, besides generating forces to ensure continuous blood supply (e.g., myocardial contractility) or withstanding passive forces generated by flow (e.g., shear stress on endocardium, myocardial wall strain, and compression strain at the level of cardiac valves), cells resident in the heart respond to mechanical cues with the activation of mechanically dependent molecular pathways. Cardiac stromal cells, most commonly named cardiac fibroblasts, are central in the pathologic evolution of the cardiovas

Importance of mechanical signals in promoting exercise-induced improvements in vasomotor function of aged skeletal muscle resistance arteries.

Current research indicates that vasomotor responses are altered with aging in skeletal muscle resistance arteries. The changes in vasomotor function are characterized by impaired vasodilator and vasoconstrictor responses. The detrimental effects of aging on vasomotor function are attenuated in some vascular beds after a program of endurance exercise training. The signals associated with exercise responsible for inducing improvements in vasomotor function have been proposed to involve short-duration increases in intraluminal shear stress and/or pressure during individual bouts of exercise. Here

Mechanotransduction mechanisms for coordinating uterine contractions in human labor.

This review presents evolving concepts of how the human uterus contracts in pregnancy, with emphasis on the mechanisms of long-distance signaling. Action potential propagation has historically been assumed to be the sole mechanism for signaling and tissue recruitment over both short and long distances. However, data in animals and humans indicate that a single action potential does not travel distances greater than a few centimeters. To address this enigma, a long-distance signaling mechanism based on hydraulic signaling and mechanotransduction is developed. By combining this mechanism for lon

Role of the extracellular matrix in epithelial morphogenesis: a view from C. elegans.

Between March and May of 2011, a cluster of three fatal cases of meningococcal sepsis occurred in Andalusia, Spain, in a municipality with a population of around 20,000 inhabitants. The cases were in their mid-teens to early thirties and were notified to the epidemiological surveillance system of Andalusia (Sistema de Vigilancia Epidemiológica de Andalucía, SVEA) during a 68-day period from March through May 2011. All three were infected with the same strain of Neisseria meningitidis serogroup C genosubtype VR1:5-1;VR2:10-8. None of the cases had been previously vaccinated against N. meningiti

[A new mechanosensitive channel SAKCA and a new MS channel blocker GsTMx-4].

Cells can respond to a variety of mechanical stimuli such as tension, pressure, and shear stress. However, the mechanisms of mechanotransduction are largely unknown. The major reason for this lies in the ambiguity of the molecular entity of cell mechanosensors. Currently only MS (mechanosensitive) channels conform to an established class of mechanosensors due to the firm and detailed analyses by electrophysiolgy. Although molecular structures of MS channels are known for limited members, higher order structures of bacterial MS channels have been resolved and their detailed structure-function s

Stress-dependent growth in breast cancer arises from a mechano-osmotic coupling and cell-sizing checkpoint.

Mechanoresponsive cell proliferation is a feature of growing tumors, despite the suppression of many other regulatory checkpoints in cancer, but the underlying cell-scale mechanisms driving this behavior have not yet been established. In this study, we propose a biophysical model for cell growth as governed by actively controlled osmolarity, which we integrate with a discrete particle framework to simulate growth and remodeling of breast cancer spheroids. Confinement and biomechanical feedback from the extracellular environment are analyzed through a neural-network-accelerated finite element s

Multi-scale and multi-physics model of the uterine smooth muscle with mechanotransduction.

Preterm labor is an important public health problem. However, the efficiency of the uterine muscle during labor is complex and still poorly understood. This work is a first step towards a model of the uterine muscle, including its electrical and mechanical components, to reach a better understanding of the uterus synchronization. This model is proposed to investigate, by simulation, the possible role of mechanotransduction for the global synchronization of the uterus. The electrical diffusion indeed explains the local propagation of contractile activity, while the tissue stretching may play a

Precision acoustofluidics for high-throughput mechanobiology in suspension cells.

Mechanomodulation, the process of altering cellular behavior through applied mechanical forces, plays a critical role in physiological processes and has substantial implications for cancer therapy, immunology, and drug development. However, precise and efficient stimulation of nonadherent cells remains a major challenge, limiting the investigation of mechanotransduction pathways and the development of targeted therapeutics. Here, we developed an acoustofluidic platform named Suspension-cell Targeted Response to Excitation via Acoustofluidic Mechanomodulation (STREAM) to enable precise, high-th

Targeting Fibrotic Scarring by Mechanoregulation of Il11ra1+/Itga11+ Fibroblast Patterning Promotes Axon Growth after Spinal Cord Injury.

Fibrotic scarring remains a critic obstacle to axonal regeneration after spinal cord injury (SCI). Current strategies primarily concentrating on eliminating extracellular matrix (ECM) components neglect their dispensable roles in maintaining tissue integrity. Here, it is reported that the mechanical strength of an integrated hydrogel composed of hyaluronic acid-graft-dopamine and HRR peptide directs fibroblast migration, determining ECM deposition. The mechanical strength matching that of spinal cord induces fibroblast alignment, reshaping fibrotic scars into a parallel matrix, while the mecha

Breaking the Mechanical Compass: Disrupting Durotaxis to Halt Fibrosis and Metastasis.

Durotaxis is the directed migration of cells along tissue stiffness gradients and is emerging as a fundamental mechanobiological process orchestrating progression in diseases such as fibrosis and cancer. Despite compelling in vitro evidence demonstrating durotactic behavior across multiple cell types, translating these findings to in vivo contexts has remained challenging because of the inherent complexity of isolating biomechanical cues from the myriad biochemical and architectural features that collectively define tissue properties. In their recent Nature Cell Biology study, Al-Hilal and col

Cnidarian mechanosensory cells as ancestors of vertebrate hair cells: Molecular targeting of an ancient mechanotransduction complex.

Conversion of mechanical stimuli into electrical signals underlies many physiological processes, including auditory and vestibular functions. In vertebrate hair cells, the mechanoelectrical transduction complex (METC)-a multiprotein and Ca2+-dependent sound and motion receptor-is located at the stereociliary tips within the hair bundle. Transmembrane channel-like 1 and 2 (TMC1 and TMC2) proteins function as the mechanosensitive channels in this complex. To investigate the evolutionary origins of vertebrate hair cells we have evaluated whether cnidarian nematocytes are related to the vertebrate

Pulsatility as a Potential Regulator of Cardiovascular Biology: Molecular, Cellular, and Hemodynamic Remodeling During Continuous-Flow Left Ventricular Assist Device Support and Following Heart Transplantation.

Pulsatile blood flow is a fundamental characteristic of cardiovascular physiology that regulates endothelial function, vascular homeostasis, microcirculatory integrity, and organ adaptation through complex mechanobiological pathways. The widespread use of continuous-flow left ventricular assist devices (CF-LVADs) has created a unique clinical model of chronic pulsatility deprivation, whereas heart transplantation restores physiological pulsatile hemodynamics. This review examines the molecular, cellular, and systemic consequences of these contrasting circulatory states. Evidence from experimen

Mechanosensitive Ion Channels and Neuroplasticity in Tuina Therapy: Molecular Mechanisms and Therapeutic Implications.

Tuina is a traditional Chinese manual therapy distinguished from generic massage by its acupoint- and meridian-based manipulations, and it is commonly used in the management of neurological and musculoskeletal disorders, although its underlying biological mechanisms remain incompletely defined. This review summarizes current experimental and clinical evidence on the neurobiological processes associated with Tuina therapy, with a focus on pain modulation and neural repair. Available evidence suggests that mechanical stimulation during Tuina may engage mechanosensitive ion channels, including tr

Mechanobiological engineering strategies for oral and maxillofacial tissue regeneration.

Mechanical forces are central determinants of development, homeostasis, and pathology in oral and maxillofacial tissues, yet most regenerative strategies continue to prioritize biochemical signaling over mechanical control. In anatomically and functionally heterogenous tissues such as dentin-pulp, periodontium, temporomandibular joint, oral mucosa, and salivary glands, mechanics is not a secondary cue but a primary regulator of cell fate, immune responses, and tissue integration. This review reframes oral and maxillofacial regeneration through a mechanobiological engineering lens, summarizing

Beyond the canonical view: steroid receptor network plasticity and mechanosensitive chromatin integration in breast cancer.

Steroid hormone receptors are traditionally described through models derived from simplified experimental conditions that do not fully recapitulate the physiological environment. In this review, we propose an integrated physiological framework for understanding steroid receptor function, in which chromatin organization, mechanotransduction, receptor network plasticity, and dynamic nuclear organization are recognized as fundamental components of hormone signaling rather than secondary regulatory layers. We highlight how steroid receptor signaling in breast cancer emerges from the coordinated in

Structure, Mechanics, and Mechanobiology of Fibrocartilage Pericellular Matrix Mediated by Type V Collagen.

The pericellular matrix (PCM) is the immediate microniche surrounding cells in various tissues, regulating matrix turnover, cell-matrix interactions, and disease. This study elucidates the structure-mechanical properties and mechanobiology of the PCM in fibrocartilage, using the murine meniscus as the model. The fibrocartilage PCM is comprised of thin, randomly oriented collagen fibrils that entrap proteoglycans, contrasting with the densely packed, highly aligned collagen fibers in the bulk extracellular matrix (ECM). Compared to the ECM, the PCM exhibits lower modulus and greater isotropy, b

Propofol inhibits Piezo mechanosensitive channels.

Modulation of ion channels is a key mechanism by which anesthetics exert their effects. Propofol, a widely used anesthetic, has been shown to influence mechanosensitive ion channels, although the details of this interaction remain under investigation. In this study, we show that propofol inhibits Piezo mechanosensitive channels using electrophysiological recordings and calcium imaging in HEK293T cells overexpressing human Piezo1 (hP1) and Piezo2 (hP2) channels. At 50 μM, propofol inhibited hP1 currents across multiple configurations (outside-out, whole cell, and cell attached) with a dissociat
