Post-traumatic stress disorder (PTSD) is a severe psychiatric disorder in which traumatic memories result in flashbacks and nightmares. With one-third of patients not responding to standard exposure-based psychotherapy, new treatment strategies are needed. Sleep offers a unique time window to enhance therapeutic efficacy. Traumatic memories that are neutralized in therapy need to be stored back into memory (consolidated) during sleep to solidify the treatment effect. New basic research shows that memory consolidation can be enhanced by presenting sounds or scents that were linked to the memory

The notion that young healthy adults can substantially improve in activities that are part of their daily routine is often overlooked because it is assumed that such activities have come to be fully mastered. We followed, in young healthy adults, the effects of repeated executions of the Timed-Up-and-Go (TUG) task, a clinical test that assesses the ability to execute motor activities relevant to daily function-rising from a seated position, walking, turning and returning to a seated position. The participants (N = 15) performed 18 consecutive trials of the TUG in one session, and were retested

Reconsolidation of a contextual fear memory is a protein synthesis-dependent process in which a previously destabilized memory returns to a stable state. This process has become the subject of many studies due to its importance in memory processing, maintenance and updating, and its potential role as a therapeutical target in fear memory disorders such as phobias and post-traumatic stress disorder. In this sense, understanding the underlying mechanisms of memory reconsolidation is paramount in developing potential treatments for such memory dysfunctions. In the present work, we studied the int

Multiple trace theory (Nadel & Moscovitch, Current Opinion in Neurobiology, 1997, 7, 217-227) has proven to be one of the most novel and influential recent memory theories, and played an essential role in shifting perspective on systems-level memory consolidation. Here, we briefly review its impact and testable predictions and focus our discussion primarily on nonhuman animal experiments. Perhaps, the most often supported claim is that episodic memory tasks should exhibit comparable severity of retrograde amnesia (RA) for recent and remote memories after extensive damage to the hippocampus (HP

This paper explores memory from a cognitive neuroscience perspective and examines associated neural mechanisms. It examines the different types of memory: working, declarative, and non-declarative, and the brain regions involved in each type. The paper highlights the role of different brain regions, such as the prefrontal cortex in working memory and the hippocampus in declarative memory. The paper also examines the mechanisms that underlie the formation and consolidation of memory, including the importance of sleep in the consolidation of memory and the role of the hippocampus in linking new

Acute moderate-intensity exercise (AME) after learning has been reported to exogenously boost consolidation of hippocampus-dependent memory, resulting in improved long-term persistence. However, the neuronal mechanism remains poorly understood. Short-term, hippocampus-dependent memory produced by weak encoding can be transformed into long-term memory through an immediate, strong behavioral event, which causes overlapping activation of the hippocampus. Hippocampal de novo protein synthesis is essential for achieving memory consolidation in this way. As AME activates the hippocampus, enhanced me

This review provides an integrative overview of the functional roles of adult neurogenesis in the hippocampal dentate gyrus (DG), focusing specifically on its impact on memory processes across the lifespan. A distinguishing feature of this review is its systematic approach, organizing the contributions of adult-born neurons (ABNs) chronologically through the stages of memory-from initial encoding, through sleep-dependent consolidation, retrieval, and finally forgetting. Although the existence and extent of adult neurogenesis in the human DG remain debated, accumulating evidence suggests that A

Accelerated long-term forgetting (ALF) is the rapid loss of newly-learnt information, typically noticeable after ∼30 min, and observed across diverse neuropsychiatric disorders including epilepsy, Alzheimer's disease (AD), Parkinson's disease (PD), traumatic brain injury, stroke, and genetic conditions such as 22q11.2 deletion syndrome. Traditionally, ALF has been attributed to impaired memory consolidation or acquisition, with emphasis on hippocampal dysfunction or epileptic activity. However, these explanations are insufficient, as ALF also occurs in individuals without hippocampal damage or

INTRODUCTION: Memory is initially stored as a transitory change that can become consolidated and converted into a long term memory trace. Consolidation largely depends on the emotional state. It is known that the hippocampus plays a role in the consolidation process of certain types of memory and that the amygdala might modulate the consolidation of the memory traces in other parts of the brain. The interaction between these two structures is crucial in many forms of learning and memory.
METHOD: The hippocampus, as well as the amygdala, display a type of synaptic plasticity known as long term

Sleep serves a fundamental role in memory consolidation, but this can be influenced by physiological context. For instance, satiated Drosophila require sleep for appetitive memory consolidation, but starved flies that must forage for food switch to sleep-independent memory. As ethanol intoxication alters behaviors such as sleep and memory, we asked how it affects the role of sleep in memory consolidation. Here, we demonstrate that acute ethanol inverts the canonical link between sleep and memory in Drosophila. Ethanol selectively impairs memory consolidation in satiated flies by causing a swit

Autobiographical memory refers to events and information about personal life and the self. Within autobiographical memory, many authors make a difference between episodic and semantic components. Study of retrograde amnesia gives information about memory consolidation. According to the "standard model" of consolidation, the medial temporal lobe plays a time-limited role in retrieval memory. Functional neuroanatomy studies of autobiographical memory are very few and many are recent. These studies concern which brain regions are involved in the autobiographical retrieval, episodic or semantic au

Background: Repeat sports-related concussive/subconcussive injury (RC/SCI) is related to memory impairment. Objective & Methods: We sought to determine memory differences between persons with RC/SCI, moderate-to-severe single-impact traumatic brain injury (SI-TBI), and healthy controls. MRI scans from a subsample of participants with SI-TBI were used to identify the neuroanatomical correlates of observed memory process differences between the brain injury groups. Results: Both brain injury groups evidenced worse learning and recall in contrast to controls, although SI-TBI group had poorer memo

BACKGROUND: Accumulating evidence suggests that rapid eye movement sleep (REM) supports the consolidation of extinction memory. REM is disrupted in posttraumatic stress disorder (PTSD), and REM abnormalities after traumatic events increase the risk of developing PTSD. Therefore, it was hypothesized that abnormal REM in trauma-exposed individuals may pave the way for PTSD by interfering with the processing of extinction memory. In addition, PTSD patients display reduced vagal activity. Vagal activity contributes to the strengthening of memories, including fear extinction memory, and recent stud

Memory storage in the brain is one of the most extensively studied subjects in neuroscience. However, due to the highly complex structure of the memory-related systems in the brain, the mystery remains unsolved. Consolidation is one of the most important parts of the memory process, and one that can be affected by numerous neurodegenerative diseases. Hypothalamic melanin-concentrating hormone (MCH) neuronal activity has been of particular interest to researchers in terms of the association between sleep, neurodegenerative diseases, and memory consolidation. We used Pmch-Cre animals to investig

BackgroundMemory consolidation problems are often prototypical in Alzheimer's disease (AD). However, it remains undetermined whether episodic memory presents similarly in early-onset Alzheimer's disease (EOAD) relative to the more commonly occurring late-onset Alzheimer's disease (LOAD).ObjectiveThis study examined episodic memory and its neuroanatomical correlates in amnestic early-onset (aEOAD) relative to amnestic late-onset AD (aLOAD).MethodsUsing our single center data set obtained from a memory clinic setting (N = 180), we examined group differences in multiple markers of episodic memory

Post-traumatic stress disorder (PTSD) is a psychiatric disorder caused by traumatic past experiences, rooted in the neurocircuits of fear memory formation. Memory processes include encoding, storing, and recalling to forgetting, suggesting the potential to erase fear memories through timely interventions. Conventional strategies such as medications or electroconvulsive therapy often fail to provide permanent relief and come with significant side-effects. This review explores how fear memory may be erased, particularly focusing on the mnemonic phases of reconsolidation and extinction. Reconsoli

Exercise-induced molecular signaling is transient, resolving within hours, whereas the structural and functional adaptations to repeated training develop over weeks to months. The processes that convert signaling into durable adaptation, the conditions that best support this conversion, and the sources of interindividual variability in training outcomes have largely been studied separately. This review proposes a conceptual framework for integrating these approaches. Adaptive Consolidation is introduced as an organizing concept for this intermediate transition, synthesized as a multi-filter ca

There is an ongoing need to identify novel pharmacological agents for the effective treatment of depression. One emerging candidate, which has demonstrated rapid-acting antidepressant effects in treatment-resistant groups, is nitrous oxide (N2O)-a gas commonly used for sedation and pain management in clinical settings and with a range of pharmacological effects, including antagonism of NMDA glutamate receptors. A growing body of evidence suggests that subanaesthetic doses of N2O (50%) can interfere with the reconsolidation of maladaptive memories in healthy participants and across a range of d

One can be aware of the effort needed to memorize a new fact or to recall the name of a new acquaintance. Because of experiences like this, learning can seem to have only two components, encoding information and, after some delay, retrieving information. To the contrary, learning entails additional, intervening steps that sometimes are hidden from the learner. For firmly acquiring fact and event knowledge in particular, learners are generally not cognizant of the necessity of offline consolidation. The memories that persist to be available reliably at a later time, according to the present con
