On the role of ascending dopamine systems in the control of voluntary ethanol intake and ethanol intoxication.

Selective lesions of the ascending dopamine pathways were made by bilateral injection of the neurotoxin 6-OHDA (8 micrograms/4 microliters). Catecholamine fluorescence histochemistry revealed a marked degeneration of the ascending mesostriatal and mesolimbic dopamine systems, while the hypothalamic dopamine and noradrenaline nerve terminals were unaffected. After recovery of feeding and drinking behaviors the voluntary ethanol intake was not different from that of the controls. The time of ethanol-induced narcosis and the extent of ethanol-induced hypothermia were not affected. In contrast, in

The role of lymphocytic CD5 in the effects of ethanol on mesolimbic circuitry and ethanol consumption behavior.

Cluster of differentiation 5 (CD5) is a negative regulatory molecule that alters lymphocyte activation pathways and is expressed on the surface of T cells, a subset of B cells (B1 cells), and dendritic cells. CD5 expression is sensitive to ethanol (EtOH) exposure, as the number of CD5+ B cells is reduced during chronic EtOH use. Given the apparent sensitivity of these cells to chronic EtOH consumption and the role of some lymphocytes in regulating the development of inhibitory GABA synapses, we assessed the impact of a murine CD5 genetic knockout (CD5KO) on measures related to EtOH reward, sed

Pathophysiology of schizophrenia and the role of newer antipsychotics.

Schizophrenia is an illness with numerous neurobiologic features. It is hypothesized that patients may have a relative deficit of dopamine neurotransmission in the nigrostriatal and mesocortical tracts of the brain, as contrasted with an excess of dopamine neurotransmission in the mesolimbic area. The dopamine deficit may be related to the negative symptoms (blunted affect, anhedonia, asociality, inability to initiate and carry out complex tasks to completion) of schizophrenia, whereas the dopamine excess may be responsible for the positive symptoms (hallucinations, delusions, and thought diso

Roles for the gut microbiota in regulating neuronal feeding circuits.

The gut microbiota has the capacity to affect host appetite via intestinal satiety pathways, as well as complex feeding behaviors. In this Review, we highlight recent evidence that the gut microbiota can modulate food preference across model organisms. We discuss effects of the gut microbiota on the vagus nerve and brain regions including the hypothalamus, mesolimbic system, and prefrontal cortex, which play key roles in regulating feeding behavior. Crosstalk between commensal bacteria and the central and peripheral nervous systems is associated with alterations in signaling of neurotransmitte…

Estradiol, dopamine and motivation.

The gonadal hormone estradiol modulates mesolimbic dopamine systems in the female rat. This modulatory effect is thought to be responsible for the observed effects of estradiol on motivated behaviors. Dopamine acting in the nucleus accumbens is thought to be important for the attribution of incentive motivational properties to cues that predict reward delivery, while dopamine in the striatum is associated with the expression of repetitive or stereotyped behaviors. Elevated concentrations of estradiol are associated with increased motivation for sex or cues associated with access to a mate, whi

Association of Neural Responses to Drug Cues With Subsequent Relapse to Stimulant Use.

IMPORTANCE: Although chronic relapse is a characteristic of addiction to stimulants, conventional measures (eg, clinical, demographic, and self-report) do not robustly identify which individuals are most vulnerable to relapse. OBJECTIVES: To test whether drug cues are associated with increased mesolimbic neural activity in patients undergoing treatment for stimulant use disorder and whether this activity is associated with risk for subsequent relapse. DESIGN, SETTING, AND PARTICIPANTS: This prospective cohort study of 76 participants included a control group for baseline group comparisons. V

Are dopaminergic pathways involved in theory of mind? A study in Parkinson's disease.

The "orbitofrontal" and "cingulate" frontostriatal loops and the mesolimbic dopaminergic system that modulates their function have been implicated in theory of mind (ToM). Parkinson's disease (PD) provides a model for assessing their role in humans. Results of the handful of previous studies of ToM in PD providing preliminary evidence of impairment remain controversial, mainly because the patients included in these studies were not accurately described, making it difficult to determine whether their ToM deficits were due to general cognitive deterioration or to a more specific dopaminergic def

The role of mesolimbic circuitry in aversive signaling and opioid dependence.

Positive reinforcement via mu-opioid receptor-mediated disinhibition of ventral tegmental area (VTA) dopamine neurons is crucial in opioid use disorder (OUD). However, VTA dopamine neurons are more heterogeneous than initially thought, both at the molecular and computational levels. Besides encoding reward prediction error, subpopulations of dopamine neurons have also been proposed to encode salience and aversion. How opioid use alters these distinct encoding properties remains unclear. Negative reinforcement-learning to avoid adverse outcomes like withdrawal-also drives chronic drug use, impl

Stress Increases Ethanol Self-Administration via a Shift toward Excitatory GABA Signaling in the Ventral Tegmental Area.

Stress is a well-known risk factor for subsequent alcohol abuse, but the neural mechanisms underlying interactions between stress and alcohol remain largely unknown. Addictive drug reinforcement and stress signaling involve common neural circuitry, including the mesolimbic dopamine system. We demonstrate in rodents that pre-exposure to stress attenuates alcohol-induced dopamine responses and increases alcohol self-administration. The blunted dopamine signaling resulted from ethanol-induced excitation of GABA neurons in the ventral tegmental area. Excitation of GABA neurons was mediated by GABA

The influence of personality on nicotine craving: a hierarchical multivariate statistical prediction model.

The present study proposes a hierarchical multivariate statistical prediction model which enables to determine the most prominent variables (physiological, biochemical and personality factors) related to nicotine craving and dopaminergic activation. Based on animal studies reporting a reduction of the rewarding effects of psychotropic drugs after blockade or destruction of the mesolimbic dopamine (DA) system, changes in nicotine craving after pharmacological manipulation by means of a DA agonist (lisuride 0.2 mg) and a DA antagonist (fluphenazine 2 mg) were assessed in 36 healthy male heavy sm

Anxiogenic drugs beta-CCE and FG 7142 increase extracellular dopamine levels in nucleus accumbens.

Two experiments were conducted to study the effects of anxiogenic drugs on dopamine release and metabolism in nucleus accumbens. Microdialysis probes were implanted into the nucleus accumbens, and rats were tested the day after implantation. In the first experiment, groups of rats received injections of saline, 1.25 or 2.5 mg/kg beta-CCE. In the second experiment, groups of rats received injections of saline, 10.0, 20.0 or 30.0 mg/kg FG-7142. Both drugs produced significant increases in dopamine release and metabolism in nucleus accumbens. Neither drug had significant effects on locomotor acti

From Belly to Brain: Targeting the Ghrelin Receptor in Appetite and Food Intake Regulation.

Ghrelin is the only known peripherally-derived orexigenic hormone, increasing appetite and subsequent food intake. The ghrelinergic system has therefore received considerable attention as a therapeutic target to reduce appetite in obesity as well as to stimulate food intake in conditions of anorexia, malnutrition and cachexia. As the therapeutic potential of targeting this hormone becomes clearer, it is apparent that its pleiotropic actions span both the central nervous system and peripheral organs. Despite a wealth of research, a therapeutic compound specifically targeting the ghrelin system

Can Decision Making Research Provide a Better Understanding of Chemical and Behavioral Addictions?

We reviewed the cognitive and neurobiological commonalities between chemical and behavioral addictions. Poor impulse control, limited executive function and abnormalities in reward processing are seen in both group of entities. Brain imaging shows consistent abnormalities in frontoparietal regions and the limbic system. In drug addiction, exaggerated risk taking behavior and temporal discounting may reflect an imbalance between a hyperactive mesolimbic and hypoactive executive systems. Several cognitive distortions are found in pathological gambling that seems to harness the brain reward syste

Roles of "Wanting" and "Liking" in Motivating Behavior: Gambling, Food, and Drug Addictions.

The motivation to seek out and consume rewards has evolutionarily been driven by the urge to fulfill physiological needs. However in a modern society dominated more by plenty than scarcity, we tend to think of motivation as fueled by the search for pleasure. Here, we argue that two separate but interconnected subcortical and unconscious processes direct motivation: "wanting" and "liking." These two psychological and neuronal processes and their related brain structures typically work together, but can become dissociated, particularly in cases of addiction. In drug addiction, for example, repea

Water Drinking Behavior Associated with Aversive Arousal in Rats: An Integrative Approach.

Cholinergic muscarinic stimulation of vast areas of the limbic brain induced a well-documented polydipsia in laboratory rats. This excessive water-drinking behavior has not received any convincing biological and physiological interpretation for the last 50 years. This review offers such an interpretation and suggests that cholinergically induced drinking response, mostly by carbachol, is associated with activation of the ascending mesolimbic cholinergic system that serves for initiation of emotional aversive arousal of the organism. The ascending cholinergic system originates from the laterodo

Involvement of opioid signaling in food preference and motivation: Studies in laboratory animals.

Motivation is a complex neurobiological process that initiates, directs, and maintains goal-oriented behavior. Although distinct components of motivated behavior are difficult to investigate, appetitive and consummatory phases of motivation are experimentally separable. Different neurotransmitter systems, particularly the mesolimbic dopaminergic system, have been associated with food motivation. Over the last two decades, however, research focusing on the role of opioid signaling has been particularly growing in this area. Opioid receptors seem to be involved, via neuroanatomically distinct me

[Cholinergic modulation on stereotyped behavior and behavioral hypersensitivity (reverse tolerance) in rats].

The failure in the mutual balance between striatal/mesolimbic dopaminergic neurons and the inhibitory system, and a decreased activity of mesocortical dopaminergic neurons may account for biological base of paranoid schizophrenia. We tested this hypothesis by investigating stereotyped behavior and "reverse tolerance" in dopaminergic supersensitivity co-existing with a low-level activity of the inhibitory cholinergic system induced by the chronic administration of methamphetamine (MAP) 4 mg/kg in combination with scopolamine (SCOP) 0.5 mg/kg, in male Wistar rats. Lack of association of a functional catechol-O-methyltransferase gene polymorphism with risk of tobacco smoking: results from a multicenter case-control study.

BACKGROUND: The catechol-O-methyltransferase (COMT) modulates dopaminergic neurotransmission in the prefrontal cortex as well as in the mesolimbic reward system. Since the reward system mediates addictive behavior, the COMT gene is a strong candidate gene regarding the pathophysiology of tobacco dependence and smoking behavior. Because of rather conflicting results in previous studies, the purpose of the present study was to test for association between a functional genetic variant in the COMT gene (single nucleotide polymorphism [SNP] rs4680) and tobacco smoking behavior. METHODS: In a popul
