Beyond total release: Fragment-resolved metrics for ultrasound microbubble-mediated drug delivery.

Microbubble-assisted ultrasound has emerged as a pivotal technology for breaching biological barriers and achieving localized drug delivery. However, current evaluative strategies are largely confined to total release efficiency as a single metric, overlooking the physicochemical heterogeneity of products generated by microbubble cavitation. This oversimplification contributes to a translational gap in which high in vitro release does not reliably predict in vivo therapeutic outcomes. To address this limitation, this Review introduces the fragment species spectrum as a conceptual and practical

Microbubble-Templated Immunoactive Metal-Phenolic Capsules for Drug Delivery and Enhanced Cancer Immunotherapy.

Advanced drug delivery systems integrating immunomodulatory functions can improve cancer therapy. Herein, we develop a novel metal-phenolic capsule for synergistic drug delivery and immunomodulation. Specifically, a stable metal-phenolic network (MPN), formed by Fe (III), tannic acid, and catechol-modified hyaluronic acid, was assembled onto doxorubicin (DOX)-loaded mannose-glycated bovine serum albumin microbubbles to construct microcapsules with both drug loading and immunomodulatory functions simultaneously. The capsules release DOX in a pH-responsive manner and induce reactive oxygen speci…

Nanosize Polymeric Foams and Microparticles Prepared In Situ From Janus-Type Microbubble Constitutions.

In this work, we generate Janus-type microbubbles that have distinct morphological patterns by utilizing microfluidics technology. A V-Junction Microfluidic (VJM) device was designed through a Solidworks program with three inlets and one microfluidic outlet attached at 30° to produce a closed system. 3% wt. PLA and 3% wt. PEG were chosen owing to the distinct physicochemical features of these polymers. Two hundred and seventy-five microliters/min PLA and 180 μL/min PEG were fed into the system with a nitrogen gas pressure of 12 kPa, where five different solvents produced Janus-type microbubble…

A Case of Pulmonary Arteriovenous Shunt Diagnosed by Microbubble Test via a Swan-Ganz Catheter.

When encountering severe hypoxemia that does not respond to oxygen supplementation, it is essential to consider underlying right-to-left shunting. Among various diagnostic approaches, the microbubble test via transthoracic echocardiography (TTE) is a simple, noninvasive method for detecting pulmonary arteriovenous shunts, particularly in hepatopulmonary syndrome (HPS). Although microbubbles are usually administered peripherally, using a Swan-Ganz (SG) catheter to inject microbubbles directly into the pulmonary artery may provide even more definitive diagnostic information. We report a case of …

Ultrasound-targeted microbubble destruction mediates PDE5i/NO integration for cavernosum remodeling and penile rehabilitation.

Erectile dysfunction (ED) caused by cavernous nerve injury (CNI) is refractory to heal mainly ascribed to the adverse remodeling of the penis induced by ineffectual microvascular perfusion, fibrosis, and neurotrophins scarcity in cavernosum. Phosphodiesterase type V inhibitors (PDE5i) have been regarded as an alternative candidate drug for avoiding penile neuropathy. However, the therapeutic efficacy is severely limited due to poor accumulation under systemic medication and endogenous nitric oxide (NO) deficiency in cavernosum. Herein, an innovative liposomal microbubble (MB) loaded with both

MR-guided ultrasound-stimulated microbubble therapy enhances radiation-induced tumor response.

High intensity focused ultrasound (HIFU) systems have been approved for therapeutic ultrasound delivery to cause tissue ablation or induced hyperthermia. Microbubble agents have also been used in combination with sonication exposures. These require temperature feedback and monitoring to prevent unstable cavitation and prevent excess tissue heating. Previous work has utilized lower power and pressure to oscillate microbubbles and transfer energy to endothelial cells in the absence of thermally induced damage that can radiosensitize tumors. This work investigated whether reduced acoustic power a…

An ultrasound responsive microbubble-liposome conjugate for targeted irinotecan-oxaliplatin treatment of pancreatic cancer.

Survival rates in pancreatic cancer have remained largely unchanged over the past four decades with less than 5% of patients surviving five years following initial diagnosis. FOLFIRINOX chemotherapy, a combination of folinic acid, 5-fluoruracil, irinotecan and oxaliplatin, has shown the greatest survival benefit for patients with advanced disease but is only indicated for those with good physical performance status due to its extreme off-target toxicity. Ultrasound targeted microbubble destruction (UTMD) has emerged as an effective strategy for the targeted delivery of drug payloads to solid t

Testing the 'microbubble effect' using the Cavitron technique to measure xylem water extraction curves.

Plant resistance to xylem cavitation is a major drought adaptation trait and is essential to characterizing vulnerability to climate change. … rifuge-based vulnerability curves: the microbubble effect. The microbubble effect hypothesizes that microbubbles introduced to open vessels, either through sample flushing or injection of solution, travel by buoyancy or mass flow towards the axis of rotation where they artefactually nucleate cavitation. To test the microbubble effect, we constructed vulnerabil …

Enhanced homing of mesenchymal stem cells to the ischemic myocardium by ultrasound-targeted microbubble destruction.

In recent years, ultrasound-targeted microbubble destruction (UTMD) has been utilised for the targeted delivery of stem cells. We tested the effects of the myocardial micro-environment changes induced by UTMD on promoting the homing of mesenchymal stem cells (MSCs) to the ischemic myocardium. Dogs were randomly divided into two groups and treated with or without UTMD after the establishment of myocardial infarction models. 4,6-diamino-2-phenyl indole (DAPI) labelled MSCs were transplanted via coronary injections 2 weeks after myocardial infarction in both groups. The results from real-time PCR

Assessing vesicoureteral reflux in live inbred mice via ultrasound with a microbubble contrast agent.

Vesicoureteral reflux (VUR) is a common pediatric anomaly linked to renal scarring and hypertension. Although there are many mouse VUR models, cystograms have previously only been performed in euthanized animals, thus preventing serial assessments for VUR in the same animal and not delineating "live" physiology. Our purpose was to develop a live murine cystogram assay that could be used serially to track reflux. We injected microbubbles via transurethral catheters into bladders of C57BL6/J and C3H/HeJ inbred mouse strains that are known to have low and high VUR rates, respectively. We performe…

From microbubbles to macro-control: Linking ultrasound-induced microarchitecture to precise drug release kinetics from acoustically responsive scaffolds.

Spatiotemporally controlled drug delivery systems are pivotal for advancing personalized medicine, yet programming precise release kinetics within implantable materials remains a significant challenge. Acoustically responsive scaffolds (ARSs), which utilize ultrasound to trigger payload release from phase-change emulsions embedded within a hydrogel matrix, offer a promising solution. However, the complex interplay of three ultrasound-based effects within an ARS - drug release kinetics, bubble morphology, and matrix mechanics - remains poorly understood. Here, we utilized a 24 full factorial de

S. aureus biofilm disruption using ultrasound and microbubbles: Influence of radiation force, bubble dynamics and biofilm growth conditions.

Staphylococcus aureus is a human pathogen and a major cause of bloodstream infections, which can readily form biofilms on implanted medical devices. Here, we utilise a combination of lipid-shelled microbubbles (MBs) and ultrasound (US) to physically disperse the biofilm from the growth surface. The effects of two peak negative pressures (PNPs) and the direction of the acoustic radiation force (ARF) were evaluated. At 1.1 MHz, a clinically relevant frequency, and low PNP of 360 kPa, no significant biofilm dispersal occurred regardless of ultrasound (US) orientation. In contrast, at a high PNP o…

Cationic microbubbles loading shFOXO4/SDF1 rejuvenate the aged heart and alleviate myocardial ischemia-reperfusion injury in the elderly.

Aging is a significant risk factor for cardiovascular diseases, with ischemic heart disease (IHD) being the leading cause of cardiovascular-related mortality. Inhibition of FOXO4, which selectively eliminates senescent cells, offers protective effects on the aging myocardium. However, the removal of senescent cells may lead to a reduction in tissue cell density, thereby exacerbating tissue space formation and perivascular fibrosis. Therefore, selectively eliminating senescent cells in the aging heart, while simultaneously replenishing therapeutic bone marrow-derived mesenchymal stem cells (BMS
