Male F344 rats, 9 weeks of age, were given multiple intrarectal administrations of N-methyl-N-nitrosourea (NMU) at 0.5 mg/dose twice a week for a total of 8, 12, or 16 doses. Five days after the final NMU instillation, rats were placed on one of four diets; chow with retinoid vehicle, chow with 654 mg N-ethylretinamide per kg diet, chow with 686 mg N-(2-hydroxethyl)retinamide per kg diet, or 406 mg retinylidene dimedone per kg diet. The influence of dietary protein content and dietary vitamin B12 supplement on the hepatotoxicity and carcinogenicity of aflatoxin in rat liver was studied. In animals fed a low-protein diet, aflatoxin induced extensive toxic and carcinogenic effects. Cirrhosis was significantly prevented to a certain level by vitamin B12 administration, but the incidence of cholangiofibrosis and hyperplastic nodules was unchanged. A DNA region not associated with conventional immunoglobulin gene rearrangement is rearranged in many lymphoid tumors. Experimental demonstration that vessels draining large tumours are impermeable to cellular elements. Thus the pulmonary vessels of animals bearing large, primary (other than gastro-intestinal) cancers can become impermeable to tumour emboli. This imperviousness prevents the establishment of secondaries in the lung and promotes the transpulmonary passage of tumor cells. This phenomenon may account for the development of paradoxical metastases.

We exposed embryos (83 hours postfertilizaton) and fry (3 weeks posthatch) to N-methyl-N'-nitro-N-nitrosoguanidine (MNNG) by immersion in aqueous solutions of 0-10 ppm for 1 hour (embryo) or 0-2 ppm for 24 hours (fry). Zebrafish embryos were microinjected with MNNG at levels of 0 or 96 ng/egg. Diets containing 0-2,000 ppm MNNG were fed to juvenile zebrafish for 3 months beginning at 2 months posthatch.
