Glial cells in schizophrenia: a unified hypothesis.

The cellular neurobiology of schizophrenia remains poorly understood. We discuss neuroimaging studies, pathological findings, and experimental work supporting the idea that glial cells might contribute to the development of schizophrenia. Experimental studies suggest that abnormalities in the differentiation competence of glial progenitor cells lead to failure in the morphological and functional maturation of oligodendrocytes and astrocytes. We propose that immune activation of microglial cells during development, superimposed upon genetic risk factors, could contribute to defective differenti

Subserosal localization of myenteric ganglia in normal human appendix: immunostaining with neuronal and glial markers.

OBJECTIVES: Visualization of unexpected distribution of myenteric ganglia in normal human appendiceal wall by immunofluorescence. BACKGROUND: The myenteric plexus is located between the longitudinal and circular muscle layers of the GIT. However, recently the irregular distribution of myenteric ganglia was revealed in human appendix. METHODS: The cryosections prepared from normal human appendices were examined by immunofluorescence methods using antibodies to neurofilaments (NF) and glial fibrillary acidic protein (GFAP). RESULTS: Indirect immunofluorescence revealed the positive staining o

Glial choristoma of the tongue. Clinicopathological analysis of a case and pathogenetic insights.

The aim of this paper is to review the use of nasal stimulants in interpersonal communication. The literature survey was performed from PubMed, ProQuest Central database of Anadolu University, and Google Scholar. Throughout history, various materials have been used as nasal stimulants to communicate and socialize intensively. Snuff is tobacco inhaled into the nostrils. It goes through the nasal mucosa into the blood and affects the neural system. Maras Powder is also a smokeless tobacco type common in the southeast of Turkey, especially in Gaziantep and Maras, which gave its name to the produc

The role of Müller cells in fibrocontractive retinal disorders.

Despite advances in surgical management of fibrocontractive retinal disorders, proliferative vitreoretinopathy (PVR) and proliferative diabetic retinopathy (PDR) remain major causes of blindness and there is still considerable uncertainty about the origins and roles of the cell types involved. Muller cells and cells identified as retinal glia are consistently identified in epiretinal tissues from both types of disorders. However, their abundance relative to total cell populations is generally low, leaving their role in these disorders uncertain. Studies of Müller cell biology using tissue cult

Patterns of astroglial hypertrophy and neuronal degeneration in the hippocampus of ages, memory-deficient rats.

The brains of aged and young Fischer rats were examined using a modified version of Cajal's gold chloride stain for astrocytes. A sizable loss of pyramidal cells and remarkably hypertrophic astrocytes were found in the hippocampus of the aged animals, while the astrocytic changes were not seen in other forebrain regions. Four major characteristics of the pattern of hippocampal astrogliosis were noted: (1) the astrocytes were often found to be grouped in clusters; (2) grouped astrocytes were commonly seen to have similarly oriented processes; (3) small blood vessels were more often stained in r

The Immuno-Glial Connectome in Alzheimer's Disease: Integrating Central and Peripheral Inflammatory Networks.

Alzheimer’s disease (AD) is increasingly recognized as a disorder of dysregulated neuroimmune connectivity rather than isolated proteinopathy. The immuno-glial connectome, the dynamic interplay between microglia, astrocytes, and peripheral immune systems, constitutes a central driver of disease initiation and progression. Emerging single-cell and spatial transcriptomic studies reveal heterogeneous glial subpopulations with context-dependent transcriptional programs governed by TREM2–APOE, NF-κB, JAK/STAT, and NLRP3 inflammasome signaling. These networks converge to sustain chronic inflammation

Targeting glial-orchestrated neuroinflammation in migraine pathophysiology.

BACKGROUND: Migraine is a chronic neurological disorder characterized by severe headache, nausea, and sensitivity to light and sound, affecting approximately 1 billion people globally. Despite advances in understanding migraine pathophysiology, particularly with the emergence of CGRP-targeted therapies, the mechanisms underlying neuroinflammation and glial contributions remain poorly understood. Current treatments are effective for a subset of patients, yet they don’t tackle the fundamental neurogenic and neuroinflammatory processes that fuel chronic migraine, especially the pathophysiological

Immune Activation and Glial Dysfunction in Spinocerebellar Ataxias: From Cerebellar Landscape to Disease-Driven Mechanisms and Immunomodulation.

Spinocerebellar ataxias (SCAs) comprise a clinically and genetically heterogeneous group of autosomal dominant neurodegenerative disorders. Despite the recognized role of specialized cerebellar glia in cerebellar development and dysfunction, immune activation and non-immune glial responses remain understudied in SCAs. This narrative review compiles evidence from cellular, animal, and human models on the cerebellar immune landscape and the specific pathways that drive homeostatic failure and neuroinflammatory cascades across SCA subtypes. Microgliosis emerges consistently-and often early- as a

Ferroptosis in Alzheimer's Disease: The Regulatory Role of Glial Cells.

Alzheimer's disease (AD) is a neurodegenerative disease characterized by the formation of amyloid plaques, neurofibrillary tangles and progressive cognitive decline. Amyloid-beta peptide (Aβ) monoclonal antibody therapeutic clinical trials have nearly failed, raising significant concerns about other etiological hypotheses about AD. Recent evidence suggests that AD patients also exhibit persistent neuronal loss and neuronal death accompanied by brain iron deposition or overload-related oxidative stress. Ferroptosis is a type of cell death that depends on iron, unlike autophagy and apoptosis. In

An Ex Vivo Explant Model for Studying Glial Interactions in the Mouse Retina.

The role of glia in glaucoma is an increasingly prominent research topic, but much remains unknown about how populations of these support cells - namely astrocytes and microglia - influence retinal ganglion cell survival. While in vivo and in vitro models provide a degree of insight, both approaches have significant limitations, such as the impact of peripheral immune response in the former and changes to physiological function induced by cell isolation and culture in the case of the latter. To minimize these confounding factors, we have developed an ex vivo retinal explant system in which ast

From glia limitans to glial scars: in vitro co-culture studies of the astrocyte and meningeal interaction.

The brain-meninges interface, comprising of astrocytes and meningeal cells seperated by a shared basement membrane, plays critical roles in the central nervous system. Recent work has shown the importance of signalling between the brain and the meninges in neurodevelopment, health, disease, and in stem cell migration. Despite this recent research, the brain-meninges interface is significantly understudied. This systematic review evaluates 27 studies which examine astrocyte-meningeal cell co-culture models. The papers were identified from a search of PubMed, Scopus, and Web of Science and scree

Glial phagocytosis for synapse and toxic proteins in neurodegenerative diseases.

Glia, as resident immune and supportive cells of the central nervous system, play a critical role in maintaining brain homeostasis. One of their key homeostatic functions is phagocytic capacity in pruning synapses and removing cellular debris/protein aggregates, a process vital for synaptic plasticity and brain maintenance. However, these phagocytic functions are often dysregulated with aging and in neurodegenerative diseases (NDs), such as Alzheimer's disease, Parkinson's disease, Huntington's disease, amyotrophic lateral sclerosis, and frontotemporal dementia. This review aims to examine the

Addressing the Effect of Exercise on Glial Cells: Focus on Ependymal Cells.

A growing body of research highlights the positive impact of regular physical activity on improving physical and mental health. On the other hand, physical inactivity is one of the leading risk factors for noncommunicable diseases and death worldwide. Exercise profoundly impacts various body districts, including the central nervous system. Here, overwhelming evidence exists that physical exercise affects neurons and glial cells, by promoting their interaction. Physical exercise directly acts on ependymal cells by promoting their proliferation and activation, maintaing brain homeostasis in heal

Mechanisms of axonal support by oligodendrocyte-derived extracellular vesicles.

Extracellular vesicles (EVs) have recently emerged as versatile elements of cell communication in the nervous system, mediating tissue homeostasis. EVs influence the physiology of their target cells via horizontal transfer of molecular cargo between cells and by triggering signalling pathways. In this Review, we discuss recent work revealing that EVs mediate interactions between oligodendrocytes and neurons, which are relevant for maintaining the structural integrity of axons. In response to neuronal activity, myelinating oligodendrocytes release EVs, which are internalized by neurons and prov

New Insights in the Complexity and Functionality of the Neurovascular Unit.

The neurovascular unit (NVU) encompasses all brain cells and underlines that neurons, glia and brain vasculature are in intimate physical and functional association. Brain function is dependent on blood flow and local increases in blood flow in response to neural activity - functional hyperaemia takes place at the NVU. Although this is a vital function of the NVU, many studies have demonstrated that the NVU also performs other tasks. Blood vessels in the brain, which are composed of multiple cell types, are essential for correct brain development. They constitute the niche for brain stem cells

NG2-glia crosstalk with microglia in health and disease.

Neurodegenerative diseases are increasingly becoming a global problem. However, the pathological mechanisms underlying neurodegenerative diseases are not fully understood. NG2-glia abnormalities and microglia activation are involved in the development and/or progression of neurodegenerative disorders, such as multiple sclerosis, Alzheimer's disease, Parkinson's disease, and cerebrovascular diseases. In this review, we summarize the present understanding of the interaction between NG2-glia and microglia in physiological and pathological states and discuss unsolved questions concerning their fat
