Reduced blood flow and impaired neurovascular coupling are recognized features of glaucoma, the leading cause of irreversible blindness worldwide, but the mechanisms underlying these defects are unknown. Retinal pericytes regulate microcirculatory blood flow and coordinate neurovascular coupling through interpericyte tunneling nanotubes (IP-TNTs). Using two-photon microscope live imaging of the mouse retina, we found reduced capillary diameter and impaired blood flow at pericyte locations in eyes with high intraocular pressure, the most important risk factor to develop glaucoma. We show that I…

Current research in predicting traumatic brain injury risk focuses on the relationship between head impacts and the likelihood of white matter damage, often overlooking the role of neurovascular coupling in the injury response. To fill this gap, we combined biomechanical and neurodynamic principles to simulate alterations to large-scale resting-state brain activity following head impacts. We simulated cortical neural activity with a network of Kuramoto phase oscillators, using structural connectivity to define coupling and a vascular-informed local resource term to capture neurovascular coupli…

The development of ischemic stroke involves swiftly changing and spatially varied states, including oxidative burst, acidosis, hypoxia, endothelial dysfunction, blood-brain barrier disruption, neuroinflammation, and later tissue remodeling. This temporal structure is poorly matched by conventional single-dose neuroprotectants. Nanozyme-hydrogel systems that respond to stimuli provide a promising approach by integrating catalytic redox control with localized retention, lesion-adaptive mechanics, and release triggered by specific cues. This Mini Review critically examines the design logic of suc

Functional near-infrared spectroscopy (fNIRS) is currently the only non-invasive method allowing for continuous long-term assessment of cere … s were seen to induce widespread HbO increases and HbR decreases. These results suggest that normal neurovascular coupling is partially retained with the hemodynamic response to the detected EEG patterns in these patients. However, the shape and distribution of the response were highly variable. This work highlighted the feasibility of conducting long-term cEEG-fNIRS to …

BACKGROUND: Natalizumab inhibits adherence of leukocytes to the cerebral endothelium. Since leukocytes play a role in regulating vascular tone, natalizumab may also affect cerebral vasoregulation. The aim of this observational study was to investigate whether neurovascular coupling and cerebral autoregulation are altered following routine clinical infusion of natalizumab in patients with relapsing-remitting multiple sclerosis.
METHODS: In 18 patients receiving regular infusion of 300 mg natalizumab, neurovascular coupling to visual stimulation and dynamic cerebral autoregulation (phase and ga…

Neurovascular coupling (NVC) reflects the coordination between brain activity and cerebral blood flow, while glymphatic function indicates the capacity to clear metabolic waste from the brain. However, differences in these two factors between patients on peritoneal dialysis (PD) and hemodialysis (HD), as well as their interrelationship, remain unclear. Functional magnetic resonance imaging, three-dimensional pseudo-continuous arterial spin labeling, and diffusion tensor imaging were prospectively performed in 56 patients on PD, 54 patients on HD, and 52 healthy controls (HC). The study calcula…

INTRODUCTION: Meige syndrome (MS) is an adult-onset segmental dystonia disease, mainly manifested as blepharospasm and involuntary movement caused by dystonic dysfunction of the oromandibular muscles. The changes of brain activity, perfusion and neurovascular coupling in patients with Meige syndrome are hitherto unknown.
METHODS: Twenty-five MS patients and thirty age- and sex-matched healthy controls (HC) were prospectively recruited in this study. All the participants underwent resting-state arterial spin labeling and blood oxygen level-dependent examinations on a 3.0 T MR scanner. The meas…

Type 1 diabetes mellitus (T1DM) can result in complications such as retinopathy, nephropathy, and peripheral neuropathy, which can lead to brain dysfunction. In this study, we investigated the effects of T1DM on cerebral neurovascular function in mice. Streptozotocin (STZ) is known to induce T1DM in animals; thus, we used an STZ-induced diabetes model to evaluate the effects of hyperglycemia on brain morphology and neurovascular tissue. Neurovascular coupling is the connection between neuronal activity and cerebral blood flow that maintains brain function. The ECoG-LSCI technique combines elec…

BACKGROUND: Fabry disease (FD) causes cerebrovascular disease (CVD) even if asymptomatic, and this is why it is important to identify non-invasive methods to monitor the disease. We evaluated the usefulness of the cerebral autoregulation, vasoreactivity, and neurovascular coupling assessed by transcranial Doppler (TCD) in FD.
METHODS: Ten adult patients with classic phenotype FD, without clinical expression of CVD, and ten healthy controls, were included. We monitored cerebral blood flow velocity with TCD in the middle and posterior cerebral arteries, blood pressure, heart rate, and non-invas…

BACKGROUND: Clinically, cardiac arrest, cardiopulmonary resuscitation, and ischemic stroke, as major contributors to cerebral ischemia/reperfusion injury (CIRI), leads to cognitive decline, brain dysfunction, and neurological disability. Increasing evidence has highlighted the significant role of neurovascular unit (NVU) damage in the development of CIRI, leading to disruptions in the blood-brain barrier (BBB), alterations in cerebral blood flow regulation, and enhanced neuroinflammation. Over time, the concept of NVU has gained prominence, with research emphasizing the intricate interactions

Amyotrophic lateral sclerosis (ALS) is a fatal neurodegenerative disorder, characterized by the degeneration of motor neurons, resulting in progressive and eventually complete loss of motor function. The phenotypical heterogeneity of ALS poses a challenge to early diagnosis and treatment, with currently available therapeutics only capable of relieving symptoms and slowing progression. While ALS is classified as a motor neuron disease (MND), research has identified significant impairment to the blood-brain barrier (BBB) and neurovascular unit (NVU) of ALS patients. The BBB along with supporting

Simultaneously monitoring cortex-wide microvascular dynamics, blood oxygenation metabolism, and neuronal activities with high spatiotemporal resolution is essential for understanding neurovascular coupling (NVC). However, it remains a challenge for existing neuroimaging tools. Here, we report a linear transducer array-based hybrid microscope (LiTA-HM) that integrates photoacoustic microscopy and confocal fluorescence microscopy to achieve simultaneous neurovascular imaging with a field of view (FOV) of 6 millimeters by 5 millimeters, a spatial resolution of ~6 micrometers, and a temporal resol…

The cerebral microcirculation undergoes dynamic changes in parallel with the development of neurons, glia, and their energy metabolism throughout gestation and postnatally. Cerebral blood flow (CBF), oxygen consumption, and glucose consumption are as low as 20% of adult levels in humans born prematurely but eventually exceed adult levels at ages 3 to 11 years, which coincide with the period of continued brain growth, synapse formation, synapse pruning, and myelination. Neurovascular coupling to sensory activation is present but attenuated at birth. By 2 postnatal months, the increase in CBF of…

Astrocytes bridge neuronal and cerebrovascular systems, regulating neurotransmitter dynamics and modulating neurovascular coupling (NVC). Cocaine disrupts neuronal, astrocytic, and cerebrovascular system function, which in the prefrontal cortex (PFC), could interfere with executive function. Though astrocytes are known to influence neurotoxic and addictive effects of cocaine through their regulations of local neurotransmitter environments, their role in cocaine-induced vasoconstriction and dysregulated NVC remains unclear. To investigate astrocytic regulation of vascular response to cocaine, w…

The high metabolic demand of retinal neurons requires a precisely regulated vascular system that can deliver rapid changes in blood flow in response to neural need. In the retina, this is achieved via the action of a coordinated group of cells that form the neurovascular unit. While cells such as pericytes, Müller cells, and astrocytes have long been linked to neurovascular coupling, more recently the resident microglial population have also been implicated. In the healthy retina, microglia make extensive contact with blood vessels, as well as neuronal synapses, and are important in vascular p…

This review paper explores the critical role of vascular ion channels in the regulation of cerebral artery function and examines the impact … ivity. Here, we briefly review the role of vascular ion channels in cerebral blood flow control and neurovascular coupling. We then examine the vascular defects in AD, the current understanding of how AD pathology affects vascular ion channel function, and how these changes may lead to compromised cerebral blood flow and neurodegenerative processes. Finally, we provide …
