Cryo-EM structure of a methanogen nitrogenase-PII protein supercomplex.

Nitrogenases are metalloenzymes that catalyse the reduction of atmospheric dinitrogen to ammonia, sustaining the global nitrogen cycle1,2. Although bacterial nitrogenase has been extensively characterized, the architecture and regulation of archaeal nitrogenases have remained unknown despite longstanding evidence of nitrogen fixation in methanogens. Here we report a cryo-electron microscopy structure of a native nitrogenase-PII protein supercomplex from Methanosarcina acetivorans. The structure reveals an assembly of three NifDK heterotetramers bridged by six NifI1,2 heterotrimeric PII complex

The Ultrastructure of Chloroplasts in Mineral-deficient Maize Leaves.

The ultrastructure of mesophyll chloroplasts in full-nutrient and mineral-deficient maize (Zea mays) leaves was examined by electron microscopy after glutaraldehyde-osmium tetroxide fixation. Nitrogen, calcium, magnesium, phosphorus, potassium, and sulfur deficiencies were induced by growing the plants in nutrient culture. Distinctive chloroplast types were observed with each deficiency. Chloroplasts from nitrogen-deficient plants were reduced in size and had prominent osmiophilic globules and large grana stacks. Magnesium deficiency was characterized by the accumulation of osmiophilic globule

Nitrogen-rich biochar electrodes from urban green waste for microbial CO2 electroreduction in bioelectrochemical systems.

Bioelectrochemical systems (BES), including microbial electrosynthesis (MES), represent a sustainable route for carbon recycling through CO2 electroreduction driven by electroactive microorganisms. However, their performance is often limited by sluggish cathodic reactions and the high cost of efficient electrodes. Nitrogen-rich biochar obtained from biomass provides a low-cost, conductive, and porous matrix with abundant active sites, making it suitable for enhancing electron transfer and catalytic activity. In this study, two biomass precursors with distinct nitrogen contents-hazelnut shells

Light-driven dinitrogen reduction catalyzed by a CdS:nitrogenase MoFe protein biohybrid.

The splitting of dinitrogen (N2) and reduction to ammonia (NH3) is a kinetically complex and energetically challenging multistep reaction. In the Haber-Bosch process, N2 reduction is accomplished at high temperature and pressure, whereas N2 fixation by the enzyme nitrogenase occurs under ambient conditions using chemical energy from adenosine 5'-triphosphate (ATP) hydrolysis. We show that cadmium sulfide (CdS) nanocrystals can be used to photosensitize the nitrogenase molybdenum-iron (MoFe) protein, where light harvesting replaces ATP hydrolysis to drive the enzymatic reduction of N2 into NH3

Segmental spinal instrumentation. A study of the mechanical properties of materials used for sublaminar fixation.

Segmental spinal instrumentation with sublaminar fixation is widely used but a variety of materials and techniques are being employed for both scoliosis and other spinal surgery. This study addresses the question as to which is best. Both the materials and the methods of securing them were studied. The materials tested were stainless steel 316L, Ortron 90 high nitrogen steel, titanium alloys, nylon straps, and Mersilene tapes. Doubled cold-worked 00.84-mm diameter stainless steel formed in a knot with a twist proved to be the strongest and most secure means of fixation with a yield value of 1,

Effects and mechanisms of exogenous acyl-homoserine lactones on nitrogen removal from aquaculture wastewater.

To improve microbial nitrogen removal efficiency in aquaculture wastewater, we added different carbon-chain-length acyl-homoserine lactones (AHLs) to continuous-flow reactors, including N-butyryl-L-homoserine lactone (C4-HSL), N-hexanoyl-L-homoserine lactone (C6-HSL), N-octanoyl-L-homoserine lactone (C8-HSL), N-dodecanoyl-L-homoserine lactone (C12-HSL). We investigated their effects on nitrogen removal performance, sludge characteristics, and microbial community functions, and further analyzed the underlying mechanisms with metagenomics. The results showed that, compared with the nitrate conce

Microbial drivers of biogeochemical cycles in deep sediments of the Kathiawar Peninsula Gulfs of India.

Deep marine sediments are rich in microbial diversity, which holds metabolic repertoire to modulate biogeochemical cycles on a global scale. We undertook the environmental microbiome inhabiting the Gulf of Kathiawar Peninsula as a model system to understand the potential involvement of the deep marine sediment microbial community and as a cohort in the carbon, nitrogen, and sulfur biogeochemical cycles. These gulfs are characterized by dynamic tidal variations, diverse sediment textures, and nutrient-rich waters, driven by coastal processes and the interaction between natural coastal dynamics

Salinity-induced variations in bacterial composition and co-occurrence patterns within Salicornia-based constructed wetlands in mariculture.

Constructed wetlands use vegetation and microorganisms to remove contaminants like nitrogen and phosphorus from water. For mariculture, the impact of salinity on the efficiency of wastewater treatment of wetlands is unneglectable. However, little is known about their impact on the microbiome in constructed wetlands. Here, we set four salinity levels (15, 22, 29, and 36) in Salicornia constructed wetlands, and the experiment was conducted for a period of 72 days. The 15 group exhibited the highest removal rates of nitrogen compounds and phosphate, compared to the other salinity groups, the nosZ

The improvement of spinach growth by nano-anatase TiO2 treatment is related to nitrogen photoreduction.

The improvement of spinach growth is proved to relate to N2 fixation by nano-anatase TiO2 in this study. The results show that all spinach leaves kept green by nano-anatase TiO2 treatment and all old leaves of control turned yellow white under culture with N-deficient solution. And the fresh weight, dry weight, and contents of total nitrogen, NH4(+), chlorophyll, and protein of spinach by nano-anatase TiO2 treatment presented obvious enhancement compared with control. Whereas the improvements of yield of spinach were not as good as nano-anatase TiO2 treatment under N-deficient condition, confi

Anabaena, a multicellular model for cyanobacterial cell biology and biotechnology.

Heterocyst-forming cyanobacteria (order Nostocales) are a monophyletic group of organisms that grow as filaments usually made of dozens to hundreds of cells (termed vegetative cells) that can perform oxygenic photosynthesis. Under combined nitrogen deficiency, some cells in the filament differentiate into N2-fixing heterocysts, which do not produce oxygen and, indeed, keep a microoxic environment appropriate for the expression and functioning of the N2-fixing enzyme, nitrogenase. Thus, in the diazotrophic filament, oxygenic photosynthesis and N2 fixation are segregated into two different cell

Metagenomic analysis reveals a green sulfur bacterium as a potential coral symbiont.

Coral reefs are ecologically significant habitats. Coral-algal symbiosis confers ecological success on coral reefs and coral-microbial symbiosis is also vital to coral reefs. However, current understanding of coral-microbial symbiosis on a genomic scale is largely unknown. Here we report a potential microbial symbiont in corals revealed by metagenomics-based genomic study. Microbial cells in coral were enriched for metagenomic analysis and a high-quality draft genome of "Candidatus Prosthecochloris korallensis" was recovered by metagenome assembly and genome binning. Phylogenetic analysis show

The electron transfer flavoprotein fixABCX gene products from Azospirillum brasilense show a NifA-dependent promoter regulation.

The complete nucleotide sequence of the A. brasilense fixA, fixB, fixC, and fixX genes is reported here. Sequence similarities between the protein sequences deduced from fixABCX genes and many electron transfer flavoproteins (ETFs) have been noted. Comparison of the amino acid sequences of both subunits of ETF with the A. brasilense fixA and fixB gene products exhibits an identity of 30%. The amino acid sequence of the other two genes, fixC and fixX, revealed similarity with the membrane-bound electron transfer flavoprotein ubiquinone oxidoreductase (ETF-QO). Using site-directed mutagenesis, m

Prokaryotic communities from a lava tube cave in La Palma Island (Spain) are involved in the biogeochemical cycle of major elements.

Lava caves differ from karstic caves in their genesis and mineral composition. Subsurface microbiology of lava tube caves in Canary Islands, a volcanic archipelago in the Atlantic Ocean, is largely unknown. We have focused the investigation in a representative lava tube cave, Fuente de la Canaria Cave, in La Palma Island, Spain, which presents different types of speleothems and colored microbial mats. Four samples collected in this cave were studied using DNA next-generation sequencing and field emission scanning electron microscopy for bacterial identification, functional profiling, and morph

Coming out of the cold: Vanadium is an important player in N-fixation in warm tropical environments.

Nitrogen (N)-fixation is a crucial source of reactive N in terrestrial environments. The nitrogenase enzyme (Nase), responsible for this conversion, has three isoforms: molybdenum (Mo)-, vanadium (V)-, and iron (Fe)-Nases. The Mo-Nase is found in all N-fixers, but the distribution pattern and drivers of V- and Fe-Nases remain poorly understood. Carbon and micronutrients (Mo and V) might be key contributors to N-fixation by affecting the metabolic demand for N and participating in Nase synthesis, respectively. Here, we investigated the presence of V-Nase genes in a tropical cyanolichen and expl

Cell-specific CO2 fixation rates of two distinct groups of plastidic protists in the Atlantic Ocean remain unchanged after nutrient addition.

To assess the role of open-ocean ecosystems in global CO2 fixation, we investigated how picophytoplankton, which dominate primary production, responded to episodic increases in nutrient availability. Previous experiments have shown nitrogen alone, or in combination with phosphorus or iron, to be the proximate limiting nutrient(s) for total phytoplankton grown over several days. Much less is known about how nutrient upshift affects picophytoplankton CO2 fixation over the duration of the light period. To address this issue, we performed a series of small volume (8-60 ml) - short term (10-11 h) n

Microbial dark matter spearheading the biogeochemical cycle in the Solar Lake of Taba, Egypt.

Microbial dark matter (MDM) represents a vast, uncultured fraction of microbial life with largely unknown ecological roles, particularly in extreme environments. This study investigates MDM in the hypersaline microbial mats of Solar Lake, Taba, Egypt, using shotgun metagenomics (∼70 M reads/sample; triplicate sampling across four sites). A total of 364 metagenome-assembled genomes (MAGs) were recovered, of which 116 (∼30 %) were classified as MDM, comprising 55 % archaeal and 45 % bacterial lineages. Functional annotation revealed that ∼14 % of the MDM MAGs had the genetic potential to fix car

Molecular insights into bacteroid development during Rhizobium-legume symbiosis.

Rhizobial soil bacteria can form a symbiosis with legumes in which the bacteria fix atmospheric nitrogen into ammonia that can be utilized by the host. The plant, in turn, supplies the rhizobia with a carbon source. After infecting the host cell, the bacteria differentiate into a distinct bacteroid form, which is able to fix nitrogen. The bacterial BacA protein is essential for bacteroid differentiation in legumes producing nodule-specific cysteine-rich peptides (NCRs), which induce the terminal differentiation of the bacteria into bacteroids. NCRs are antimicrobial peptides similar to mammali

Modeling and Simulation of a New Proposed System for the Elimination of Nitrogen Contained in Natural Gas.

This work presents the modeling and simulation of a new biotechnological system for nitrogen removal from natural gas, based on immobilized diazotrophic microorganisms operating in a packed-bed bioreactor. The model incorporates a first-order apparent reaction rate and a diffusion-reaction formulation within porous supports, coupled with reactor-scale mass balances and pressure-drop effects. Because system-specific kinetic data for gas-solid diazotrophic systems are not yet available, an apparent first-order rate was adopted as a nominal reference value, and its influence on system behavior wa

Fundamental Role of N-O Bond-Containing Compounds in Prebiotic Synthesis.

The emergence of biomolecules on the primordial Earth represents a pivotal scientific question for understanding the origin of life. Recent studies show that compounds containing nitrogen-oxygen bonds could serve as important feedstocks in prebiotic synthesis and intermediates in primitive metabolic pathways. Simple N-O-compounds have been identified in the interstellar medium and were likely formed from molecular nitrogen in the early Earth's atmosphere in the course of high-energy events. N-O-compounds are reactive species with rich chemistry enabling abiotic nitrogen fixation processes. Mor

Physiological and gene expression responses to nitrogen regimes and temperatures in Mastigocladus sp. strain CHP1, a predominant thermotolerant cyanobacterium of hot springs.

Cyanobacteria are widely distributed primary producers with significant implications for the global biogeochemical cycles of carbon and nitrogen. Diazotrophic cyanobacteria of subsection V (Order Stigonematales) are particularly ubiquitous in photoautotrophic microbial mats of hot springs. The Stigonematal cyanobacterium strain CHP1 isolated from the Porcelana hot spring (Chile) was one of the major contributors of the new nitrogen through nitrogen fixation. Further morphological and genetic characterization verified that the strain CHP1 belongs to Stigonematales, and it formed a separate clad
