An ultrastructural study of cytodifferentiation in the developing human root-tip.

Transmission electron microscopy showed that in growing human teeth, the root sheath consisted of inner and outer epithelial cells. The inner epithelial cells formed a basal lamina associated filamentous layer which increased in density in coronal direction. Extensions of developing odontoblasts were in contact with the basal lamina. Direct contact with the epithelial plasmalemma was not observed. The odontoblasts obtained their fully-developed cylindrical appearance after making contact with the basal lamina of the inner epithelial cells. But, they possessed already abundant RER before these …

Ultrastructural study of fossil human tooth tissue.

The preservation of fossil human soft tooth tissue from extinct populations which inhabited the northeast of Argentina (fourteenth and fifteenth centuries) is described. Studies were performed using both Scanning Electron Microscopy (SEM) and Transmission Electron Microscopy (TEM). The preservation of the surface structure in Tomes fibrils and in odontoblastic processes was determined by SEM; whereas by TEM we could observe the dentin ducts occupied by acellular material, being the inner structure of the odontoblastic processes poorly preserved. It is suggested that such preservation is due to

Expression of fibroblast growth factor receptor1, -2c, and -3c transcripts in mouse molars after tooth eruption.

A previous study suggested that fibroblast growth factor (FGF) signaling plays an important role in dentin formation during tooth developmen … experimental period. By in situ hybridization, Dspp, Fgfr1, and Fgfr3c transcripts were detected in odontoblasts at ages of 2 and 4 weeks. In addition, Dspp and Fgfr1 transcripts were detected in odontoblasts facing reactionary dentin at 8 weeks old. These results suggest that FGF-FGFR signaling might be involved in the regulation of odontoblasts even after to …

Somatosensory Functions of Melastatin Transient-Receptor Potential Channels in the Teeth: Molecular Basis for Thermal Dentine Hypersensitivity.

Dental pain due to dentine hypersensitivity or pulpitis is characterized by short or lasting episodes of pain triggered by normally innocuous stimuli originating from exposed dentine. Both represent the most frequent pain of the orofacial region. Transient receptor potential (TRP) superfamily of ion channels participates in the detection of different modalities of sensibility in the mammalian sensory teeth system, i.e., trigeminal neurons and odontoblasts. In particular, some members of the melastatin family (TRPM) serve as molecular thermal sensors, and temperature is one of the most potent s…

RANK, RANKL and OPG expressions in a permanent molar with a replacement resorption.

The aim of this study was to explore the expression of RANK, RANKL and OPG during ankylosis. Structural details and immunohistochemical investigations of the expression of RANK, RANKL and OPG in an extracted secondary retained permanent molar of a 12-year-old girl are reported. Woven and lamellar bones were observed in the thickness of the remodeled dental wall and a tertiary dentin was noticed around the pulp cavity. The resorbing multinucleated cells expressed TRAP and RANK but few of them also expressed RANKL. Both odontoblasts and osteoblasts expressed TRAP and RANK, but the expression of …

Pattern-recognition receptors in pulp defense.

Initial sensing of infection is mediated by germline-encoded pattern-recognition receptors (PRRs), the activation of which leads to the expression of inflammatory mediators responsible for the elimination of pathogens and infected cells. PRRs act as immune sensors that provide immediate cell responses to pathogen invasion or tissue injury. Here, we review the expression of PRRs in human dental pulp cells, namely, receptors from the Toll-like (TLR) and Nod-like NLR families, by which cells recognize bacteria. Particular attention is given to odontoblasts, which are the first cells encountered b…

Immunoreactivity for mGluR5 and TRPV1 was also observed in odontoblasts. These results support the possibility that the nerve fibers in the dental pulp mediate inflammatory pain and thermal hyperalgesia through coactivation of mGluR5 and TRPV1 and also suggest a possible role for odontoblasts in the transduction of noci …

The involvement of genes related to bile secretion pathway in rat tooth germ development.

Tooth formation is accomplished under strict genetic control procedures. Therefore, exploring the gene network system of tooth development h … the postnatal 1 and postnatal 7, SLC2A1, SLC4A4 and ABCC3 were highly expressed in ameloblasts and odontoblasts while ADCY5, ATP1B1 and SLC10A1was expressed moderately only in odontoblasts. This finding illustrated that the bile secretion pathway related genes may participate in the development of tooth germ.

Effects of fibroblast growth factor-2 on the expression and regulation of chemokines in human dental pulp cells.

BACKGROUND: Fibroblast growth factor-2 (FGF-2) participates in both hematopoiesis and osteogenesis; however, the effects of FGF-2 on chemokines during odontoblastic differentiation have not been reported. This study investigated whether human dental pulp cells (HDPCs) treated with FGF-2 could express chemokines during differentiation into odontoblastic cells and sought to identify its underlying mechanism of action. METHODS: To analyze differentiation, we measured alkaline phosphatase (ALP) activity, calcified nodule formation by alizarin red staining, and marker RNA (mRNA) expression by reve

Dentin sialoprotein: biosynthesis and developmental appearance in rat tooth germs in comparison with amelogenins, osteocalcin and collagen type-I.

A non-collagenous protein, extracted from rat incisor dentin, is a dentin sialoprotein (DSP). We examined immunohistochemically the developmental appearance and tissue distribution of DSP in 1 to 3-day-old rat molar and incisor tooth germs. The earliest staining for DSP was observed in newly differentiated odontoblasts. In more advanced stages, immunostaining for DSP gradually increased in pre-dentin, odontoblasts and dentin, and appeared in many cells of the dental papilla. In early stages of development before the breakdown of the dental basement membrane, pre-ameloblasts were also positive …

Notch signalling pathway in tooth development and adult dental cells.

Notch signalling is a highly conserved intercellular signal transfer mechanism that includes canonical and non-canonical pathways. It regulates differentiation and proliferation of stem/progenitor cells by means of para-inducing effects. Expression and activation of Notch signalling factors (receptors and ligands) are critical not only for development of the dental germ but also for regeneration of injured tissue associated with mature teeth. Notch signalling plays key roles in differentiation of odontoblasts and osteoblasts, calcification of tooth hard tissue, formation of cusp patterns and g…

An Innovative "Tooth-On-Chip" Microfluidic Device Emulating the Structure and Physiology of the Dental Pulp Tissue.

The dental pulp is a highly vascularized and innervated connective tissue composed of various cell types, including fibroblasts, odontoblasts, mesenchymal stem cells, neuronal, and endothelial cells. The interplay between these diverse cell populations is pivotal for dental pulp tissue homeostasis and regeneration after carious infections and traumatic tooth lesions. Despite the great clinical need, comprehensive in vitro models that accurately recapitulate the complexity of the dental pulp are still missing, hampering the development of novel, faster, and more effective therapies. In this stu…

Investigating the Effects of Conditioned Media from Stem Cells of Human Exfoliated Deciduous Teeth on Dental Pulp Stem Cells.

Pulp regeneration has recently attracted interest in modern dentistry. However, the success ratio of pulp regeneration is low due to the compromising potential of stem cells, such as their survival, migration, and odontoblastic differentiation. Stem cells from human exfoliated deciduous teeth (SHED) have been considered a promising tool for regenerative therapy due to their ability to secrete multiple factors that are essential for tissue regeneration, which is achieved by minimally invasive procedures with fewer ethical or legal concerns than those of other procedures. The aim of this study i

Hormone-responsive cells derived from human dental papilla: characterization in vitro and in vivo in diffusion chambers.

Cells of the dental papilla are capable of odontoblastic, fibroblastic, and endothelial differentiation and formation of dentin and the dental pulp. In the present study dental papilla cells, obtained from human tooth buds (HDP cells), were cultured in vitro through 3 to 7 passages. After exposure to prostaglandin E2 there was a marked decrease in intracellular cyclic AMP (cAMP) levels as compared to hormone-free controls. Parathyroid hormone and calcitonin had stimulatory effects with 1 and 2 log increases in cAMP, respectively. The HDP cells showed moderate activity of alkaline phosphatase,
